Osteoblasts are essential in the maintenance of human bone homeostasis.1 The abnormal formation and impaired differentiation ability of osteoblasts are pivotal factors leading to bone-related diseases,suggesting that an indepth study of osteoblasts can provide novel treatment strategies for these diseases.2 Human primary osteoblasts are precious experimental models in bone-related research.3 However,the isolation and culture of human primary osteoblasts still have some difficulties and limitations.Here,we refined a method to isolate and culture human primary osteoblasts by taking cancellous bone through a bone tissue sampler and pre-digesting it with pancreatin,which makes the process easier,cheaper,and more efficient.Furthermore,we compared the effects of the subtle changes in isolation and culture methods on the phenotype of human primary osteoblasts and examined the differences in morphology,proliferation,and differentiation between micro-explants cultured osteoblasts(MECOBs)and explants cultured osteoblasts(EC-OBs).

Tennessee Williams is the most famous Southern playwright in the history of 20th century American literature,and southern identity is a thread that runs throughout Williams’work.The Glass Menagerie is one of his highly representative dramatic works.The play tells the story of how an ordinary family in deep identity crisis escapes the harsh reality and cultural trauma during the Great Depression in the United States.Using trauma theory as a tool,this paper explores the complex relationship between the deep-seated causes of the tragedy of the two female characters in the play and the historical context through the traumatic events experienced by the characters in the play and the different attitudes they show in the face of trauma.Through this,the vulnerability and identity crisis of individuals during the period of social change and cultural breaks are analyzed.The purpose of the paper is to arouse human beings’thoughts on how to cope with cultural integration and foreign cultural invasion,and how to obtain salvation when they are amid an identity crisis.

Gene-editing technologies have ushered in a significant advancement in plant genetics research and molecular breeding.However,a critical challenge hindering the widespread adoption of these technologies is the efficient delivery of gene-editing tools.The predominant methods for introducing these tools into plants typically involve Agrobacterium tumefaciens-mediated transformation or particle bombardment(Mao et al.,2019).Unfortunately,these traditional gene delivery methods require delicate and timeconsuming tissue culture procedures and show limited success,especially in medicinal and other less studied plants.A recent breakthrough is the development of the Cut-Dip-Budding(CDB)gene delivery system.The CDB system is highly effective for plants with root-suckering capabilities.It allows the delivery of transgenes and gene-editing tools into plants through hairy root induction followed by shoot regeneration from transformed hairy roots,bypassing the need for tissue culture processes(Cao et al.,2023).

In this review of ten years of social innovation research(2012e2022),we define and explore three paradigms in the field:instrumentalist,strong,and democratic.We investigate how language usage and geography play a central role in identifying which paradigms recently published scholarship falls into.While we do not insist that sharp divisions exist between each paradigm,we do find that on the“instrumentalist”side,language tends to abstract or neutralize power relations.Further,these perspectives tend to derive from Western or Eurocentric orientations or biases.The“strong”paradigm accepts the necessity of institutional and stakeholder engagement and seeks to engage socially excluded populations.In contrast,geographical diversity,attendance to historicized and systemic inequalities,and elevation of the most marginalized communities are more likely to be centered in the“democratic”paradigm.We apply this discussion to recent research in arts-related social innovation and the related field of social entrepreneurship.

BACKGROUND Timing of passaging,passage number,passaging approaches and methods for cell identification are critical factors influencing the quality of neural stem cells(NSCs)culture.How to effectively culture and identify NSCs is a continuous interest in NSCs study while these factors are comprehensively considered.AIM To establish a simplified and efficient method for culture and identification of neonatal rat brain-derived NSCs.METHODS First,curved tip operating scissors were used to dissect brain tissues from new born rats(2 to 3 d)and the brain tissues were cut into approximately 1 mm^(3)sections.Filter the single cell suspension through a nylon mesh(200-mesh)and culture the sections in suspensions.Passaging was conducted with TrypLTM Express combined with mechanical tapping and pipetting techniques.Second,identify the 5th generation of passaged NSCs as well as the revived NSCs from cryopreservation.BrdU incorporation method was used to detect self-renew and proliferation capabilities of cells.Different NSCs specific antibodies(anti-nestin,NF200,NSE and GFAP antibodies)were used to identify NSCs specific surface markers and muti-differentiation capabilities by immunofluorescence staining.RESULTS Brain derived cells from newborn rats(2 to 3 d)proliferate and aggregate into spherical-shaped clusters with sustained continuous and stable passaging.When BrdU was incorporated into the 5th generation of passaged cells,positive BrdU cells and nestin cells were observed by immunofluorescence staining.After induction of dissociation using 5%fetal bovine serum,positive NF200,NSE and GFAP cells were observed by immunofluorescence staining.CONCLUSION This is a simplified and efficient method for neonatal rat brain-derived neural stem cell culture and identification.
