Somatic mutations in FAT cadherin family members constitute an underrecognized subtype of colorectal adenocarcinoma with unique clinicopathologic features

BACKGROUND The FAT cadherin family members(FAT1,FAT2,FAT3 and FAT4)are conserved tumor suppressors that are recurrently mutated in several types of human cancers,including colorectal carcinoma(CRC).AIM To characterize the clinicopathologic features of CRC patients with somatic mutations in FAT cadherin family *** We analyzed 526 CRC cases from The Cancer Genome Atlas PanCancer Atlas *** samples were subclassified into 2 groups based on the presence or absence of somatic mutations in FAT1,FAT2,FAT3 and *** clinicopathological data were collected after digital slide *** analysis was performed using t tests and chi-square *** This CRC study cohort had frequent mutations in the FAT1(10.5%),FAT2(11.2%),FAT3(15.4%)and FAT4(23.4%)*** hundred CRC patients(38.0%)harbored somatic mutations in one or more of the FAT family genes and were grouped into the FAT mutated CRC *** FAT-mutated CRC subtype was more commonly located on the right side of the colon(51.0%)than in the rest of the cohort(30.1%,Pclinicopathologic features,including a lower rate of positive lymph nodes(pN1-2:33.5%vs 46.4%,P=0.005),a lower rate of metastasis to another site or organ(pM1:7.5%vs 16.3%,P=0.006),and a trend toward an early tumor stage(pT1-2:25.0%vs 18.7%,P=0.093).FAT somatic mutations were significantly enriched in microsatellite instability CRC(28.0%vs 2.1%,Pclinicopathologic behaviors,as well as a trend of a better diseasefree survival rate(hazard ratio=0.539;95%confidence interval:0.301-0.967;log-rank P=0.073).CONCLUSION FAT cadherin family genes are frequently mutated in CRC,and their mutation profile defines a subtype of CRC with favorable clinicopathologic characteristics.

Ultrasonographic and clinicopathologic features of benign Brenner tumors of the ovary

Objective The aim of this study was to summarize and analyze the ultrasonographic and clinicopathologic features of benign brenner tumors of the *** Forty-six patients with brenner tumors of the ovary were included,and the imaging and pathologic features of the tumors were *** Thirty-eight cases were unilateral,while eight cases were *** tumors were located only in the left ovary in 32 patients and in the right ovary in six *** median diameter of the tumors was 62 mm,and the diameter ranged from 15 to 270 *** tumors were not *** of the tumors(22/46)showed micro-perfusion,while 16 tumors showed no blood flow *** signal shadows after the cysts were observed in 26 tumors,accounting for 57%of all brenner *** patients showed unilocular or multilocular tumors with no distinct characteristics on *** masses showed a multilocular solid structure,accompanied by calcification and a post-mass shadow;the solid structure showed mild-to-moderate vascularization on Doppler *** masses showed a pure solid structure,accompanied by calcification,resulting in an acoustic shadow behind the tumor;the solid structure showed mild-to-moderate vascularization on Doppler *** Some ultrasonographic features,including calcification with shadow,poor blood circulation of solid components,and mass localization,are helpful in the diagnosis of benign brenner tumors.

Etiological and clinicopathologic characteristics of intrahepatic cholangiocarcinoma in young patients

AIM:To investigate the prevalence,risk factors,and clinicopathologic characteristics of intrahepatic cholangiocarcinoma(ICC)in young ***:A retrospective analysis was performed in ICC patients referred to the Eastern Hepatobiliary Surgery Hospital in Shanghai,*** 317 consecutively enrolled patients,40 patients were aged ≤40 years(12.61%).We compared the risk factors and clinicopathologic characteristics of these patients(groupⅠ:n=40)with those aged>40 years(group Ⅱ:n=277).RESULTS:Group I had distinct features compared with groupⅡ,including a low frequency of hepatolithiasis(P=0.000);a high positive rate of serum hepatitis B surface antigen(P=0.000)and hepatitis B virus(HBV)associated cirrhosis(P=0.038);a high frequency ofα-fetoprotein(>400μg/L)(P=0.011);a low frequency of carbohydrate antigen 19-9(>37 U/mL)(P=0.017);and a high frequency of liver histological inflammation(P=0.002).Although there was no significant difference between the two groups in regards to hepatic schistosomiasis,alcohol-associated cirrhosis and cirrhosis due to other causes(P>0.05),they only occurred in the elderly ***:The risk factors are significantly different between young and elderly ICC *** and HBV-associated cirrhosis are the most important risk factors for young ICC patients.

Mucinous cystic neoplasms （MCNs） of the pancreas are a distinct entity, account for 1% of pancreatic exocrine tumors. MCNs can be classified histologically as adenomas, borderline tumors, or carcinomas. Because several evidences showing that mucinous cystadenomas are potentially malignant and may transform into cystadenocarcinomas, particularly if treated by drainage, these tumors should be identified accurately.

Prostate cancer:a presentation of climcopathologic prognosticators among Filipino and American men at radical prostatectomy

Lower incidence and mortality rates from prostate cancer(PCa)have been shown in Asian men in general compared to *** is the first study detailing the clinicopathologic features of resected prostate cancer in Filipino men living in the Philippines(PH)_This study investigated the supposed“lower risk”Filipino and“higher risk”American PCa patients from the PH and the United States of America(USA),*** examined 348(176 from PH,172 from USA)radical prostatectomy *** clinicopathologic features of both groups(age at time of diagnosis,preoperative prostate-specific antigen[pre-op PSA]level,Gleason score[GS],Grade groups[GG],margin involvement,extra prostatic extension[EPE],seminal vesicle invasion[SVI],and regional lymph node[RLN]metastasis)were *** of seven prognosticators examined were more strongly associated with Filipinos than with *** were older at diagnosis(PH:64.32±6.56 years vs USA:58.98±8.08 years)and had higher pre-op PSA levels(PH:21.39±46.40 ng ml^(-1) vs USA:7.63±9.19 ng ml^(-1)).Filipino men had more advanced grade,GG 2 with minor pattern 5(PH:6.2%vs USA:2.9%)and GG 5(PH:14.8%vs USA:3.5%).Likewise,other adverse pathological features in margin positivity(PH:52.3%vs USA:23.8%),focal EPE(PH:14.2% vs USA:2.3%),and SVI(PH:17.1%vs USA:5.8%)were more commonly observed in *** study reveals the prognostic disadvantage of Filipinos versus Americans and highlights an important difference of Filipinos from other studied Asian ethnicities that have repeatedly been shown to have lower-risk *** study,the first on Filipino PCa patients with RP,suggests the need to modify Western-based risk stratification when employed in other countries like the PH.
