cardiovascular disease is the number one cause of death in ***,cardiotoxicity is one of the most important adverse effects assessed by arrhythmia recognition in drug ***,cell-based techniques developed for arhythmia recognition primarily employ linear methods such as time-domain analysis that detect and compare individual waveforms and thus fall short in some applications that require automated and efficient arrhythmia recognition from large *** carried out the frst report to develop a biosensing system that integrated impedance measurement and multiparameter nonlinear dynamic algorithm(MNDA)analysis for druginduced arrhythmia recognition and *** biosensing system cultured cardiomyocytes as physiologically relevant models,used interdigitated electrodes to detect the mechanical beating of the cardiomyocytes,and employed MNDA analysis to recognize drug-induced arrhythmia from the cardiomyocyte beating *** best performing MNDA parameter,approximate entropy,enabled the system to recognize the appearance of sertindoleand norepinephrine-induced arrhythmia in the *** MNDA reconstruction in phase space enabled the system to classify the different arrhythmias and quantify the severity of *** new biosensing system utilizing MNDA provides a promising and alternative method for drug-induced arrhythmia recognition and classification in cardiological and pharmaceutical applications.

Non-small cell lung cancer(NSCLC)is a leading cause of cancer mortality *** epidermal growth factor receptor tyrosine kinase inhibitors(EGFR-TKIs)have dramatically improved the life expectancy of patients with NSCLC,concerns about TKI-induced cardiotoxicities have ***0010,a novel third-generation TKI,was developed to overcome drug resistance induced by EGFR-T790M ***,the cardiotoxicity of AC0010 remains *** evaluate the efficacy and cardiotoxicity of AC0010,we designed a novel multifunctional biosensor by integrating microelectrodes(MEs)and interdigital electrodes(IDEs)to comprehensively evaluate cell viability,electrophysiological activity,and morphological changes(beating of cardiomyocytes).The multifunctional biosensor can monitor AC0010-induced NSCLC inhibition and cardiotoxicity in a quantitative,label-free,noninvasive,and real-time ***0010 was found to significantly inhibit NCI-H1975(EGFR-L858R/T790M mutation),while weak inhibition was found for A549(wild-type EGFR).Negligible inhibition was found in the viabilities of HFF-1(normal fibroblasts)and *** the multifunctional biosensor,we found that 10μM AC0010 significantly affected the extracellular field potential(EFP)and mechanical beating of *** amplitude of EFP continuously decreased after AC0010 treatment,while the interval decreased first and then *** analyzed the change in the systole time(ST)and diastole time(DT)within a beating interval and found that the DT and DT/beating interval rate decreased within 1 h after AC0010 *** result probably indicated that the relaxation of cardiomyocytes was insufficient,which may further aggravate the ***,we found that AC0010 significantly inhibited EGFR-mutant NSCLC cells and impaired cardiomyocyte function at low concentrations(10μM).This is the first study in which the risk of AC0010-induced cardiotoxicity was *** addition,novel multifunctional bi

目的以乌头碱为工具药,建立基于实时细胞分析(real-time cell analysis,RTCA cardio)系统的体外心律失常检测技术,为抗心律失常药物的研发提供可靠方法。方法利用八导生理记录仪从整体动物水平检测乌头碱对实验大鼠心律的影响;体外培养心肌细胞,首先利用RTCA方法考察心肌细胞的接种密度,利用RTCA cardio系统监测乌头碱对心肌细胞搏动的影响,分析细胞的CI值、细胞搏动频率、振幅、不规则性节律等。结果八导生理记录仪检测50 mg·g^-1乌头碱对整体动物的影响结果发现乌头碱可诱发室速、室颤、RR间期缩短以及心率增加等心律失常的表现;RTCA cardio系统显示2~8μmol·L^-1乌头碱可呈剂量依赖性的导致心肌细胞搏动频率增加、搏动幅度降低、搏动状态异常等心律失常现象。结论RTCA cardio系统可以快速、敏感、准确检测乌头碱诱导的心肌细胞心律失常现象,为抗心律失常药物的研发提供了方法学参考。

Raised levels of the cardiac biomarker, Troponin I, are frequently encountered in hemodialysis patients and appear to be prognostic indicators for cardiovascular risk. Though evidence suggests that control of secondary hyperparathyroidism may reduce cardiac endpoints, the effect of the calcimimetic agent, cinacalcet, remains controversial. This retrospective study aimed at evaluating troponin levels in hemodialysis patients with severe secondary hyper parathyroidism (SHPT) who are on cinacalcet vs controls on conventional treatment. In addition, clinical outcomes including all-cause, cardiovascular morbidity and mortality were compared among both groups. A decline in Troponin I levels was observed in the cinacalcet group, this however was not translated clinically into improved survival. In fact, all-cause and cardiac mortality was similar in the two groups. Conversely, comparison of the incidence of cardiovascular events revealed lower rates in the cinacalcet group including cardiac, cerebral and peripheral vascular complications. Given some of our study limitations, further long-term, placebo-controlled trials are necessary to definitively establish the effect of cinacalet on cardiac biomarkers and ultimately its impact on clinical outcomes.

The effects of fresh zucchini on nutritional status, and biological indicators for the prevention of cardiovascular disease in rats fed high-fat diets investigated in this study. Thirty Sprague Dawley rats were randomly divided into two main groups the first, negative control group Co (-) (n = 6), fed basal diet, The second group (n = 24) fed high-fat diet (containing basal diet + 5% tallow + 1% cholesterol + 0.02% bile salt). This group was divided into four subgroups each group 6 rats: group positive control co (+) fed high-fat diet only, group 1 (G (1)) fed high-fat diet plus 10% zucchini, group 2 (G (2) fed high-fat diet plus 15% zucchini and group 3 (G (3)) fed high-fat diet plus 20% zucchini. The levels of serum total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C) were measured after eight wk. of experimental treatment. The pathologic changes of the heart, spleen and kidney were evaluated. SPSS, one way ANOVA was used to analyze the results. The results indicated that the mean values of body weight gain (BWG) and feed efficiency ratio (FIR) in G (2) and G (3) showed significant decrease compared to co (-), co (+) and G 1. The results showed that in groups (G1, G2 and G3) the relative weight of heart had significant increase when compared with control negative group. But liver and spleen weight had no significant difference when compared with control negative and positive group, the levels of TC, TG and LDL-C were significantly increased in co (+) (P < 0.05) Compared to co (-), all groups fed on high fat diet containing different levels of zucchini (10%, 15% and 20%) had significant decrease in TC and LDL-C compared with co (+), also G2 and G3 had improve significant in HDL-C when compared with co.
