Comparison of Ventricular Electrophysiological Effects of Amiodarone in Canine Models With Congestive Heart Failure and Normal Dogs

Objectives This study compared the effects of amiodarone on ventricular electrophysiological properties in normal dogs and CHF dogs. Methods Dogs （n=44） were randomized into four groups： Group 1 （n=10） was the control. Group 2 （n=10） was given amiodarone orally 300 mg·d^-1 for 4 to 5 weeks. Group 3 （n=12） was the congestive heart failure （CHF） models induced by right ventricular rapid pacing （240 pulses·min^-1 for 4 to 5 weeks）. Group 4 （n=12） was the CHF models given amiodarone orally 300 mg·d^-1 for 4 to 5 weeks. The ventricular electrophysiological variables were evaluated by standard electric stimulation and monophasic action potential （MAP） recording. Results Amiodarone prolonged sinus cycle length （SCL）, intra-ventricular conduction time （IVCT）, MAP duration （MAPD90）, ventricular effective period （VERP）, ventricular activation time （VAT） and ventricular recovery time （VRT） without significant effects on the ratio of VERP to MAPD90 （VERP/MAPD90）, ventricular fibrillation threshold （VFT）, the dispersion of VRT （VRT-D）, and ventricular late repolarization duration （VLRD） in normal dogs. However, amiodarone did not further prolong the prolonged SCL, MAPD90, VERP, VAT and VRT, but further prolonged IVCT in CHF dogs. Amiodarone normalized the abnormal ventricular electrophysiological properties in CHF dogs as manifested by increasing the decreased VERP/MAPD90 and VFT, shortening the prolonged VLRD, and decreasing the increased VRT-D. Amiodarone did not worsen the hemodynamic parameters in normal and CHF dogs. Conclusions Amiodarone had different effects on ventricular electrophysiological properties in normal and CHF dogs. The favorable effects of amiodarone in normalizing some abnormal cardiac electrophysiological properties in CHF models may have potential value on the prevention and treatment of ventricular arrhythmias and sudden cardiac death in CHF.

The use of immunohistochemical techniques has enhanced the diagnostic intensities in pathology. The sensitivity and specificity of any immunohistochemical protocol mainly depends on the ability to unmask antigen from formalin fixed tissue. Various combinations of antigen retrieval protocols have been used to unmask antigen efficiently but none has been established as a flawless one. These protocols either have one or the other lacunae, lacking repeatability under same conditions. In our study, we tried to develop a standardized immunohistochemical protocol for the identification of blood and lymphatic vessels in tissue sections of Canine mammary tumour (CMT) using recently identified markers. The combined effects of antigen retrieval (AR) methods including pH shock by Heat-induced epitope retrieval (HIER) and Protease-induced epitope retrieval (PIER) were found highly effective in retrieval of tedious antigens of lymphangiogenesis and angiogenesis. The use of combined antigen retrieval technique for the unmasking of antigens from over fixed, old formalin fixed paraffin embedded (FFPE) Canine mammary tumour sections resulted in efficient unmasking of epitopes when compared with the individual antigen retrieval methods.

Oligonucleotide of cFIX eDNA (Canine FIX, cFIX) was used to transcript mRNA of dog liver cell to cDNA by RT-PCR, and further construct it on the plasmid vector pGEM-T. The correct sequence of cFIX eDNA was obtained which covered the entire cFIX coding region. Furthermore, GlNaCcIX (driven by hCMV promoter) and GlNaMBcIX (driven by MCK enhancer and β-aetin promoter) were constructed using the retroviral vector backbone of GlNa. Canine skin fibroblast (CSF) was used as target cell, transduced with the above constructors respectively. The results showed that these modified CSF cells could express cFIX and that the expression levels were 173 ng/10~6 cell/24 h (GlNaCcIX) and 211 ng/10~6 cell/24 h (GlNaMBcIX) respectively. Those data offered a promising result for further animal study.

Neutrophils are the most important circulating phagocytes. Circulating mono-cytes and precursors of tissue macrophages also have the ability to phagocytize. Pidotimod (ADIMODTM) exerts immunostimulatory and immunoregulatory effects through the stimulation and regulation of cellular immune responses by lymphocytes Canine herpesvirus (CHV) mainly affect puppies between the first and second weeks of age, causing high morbidity in the litter. To date, there is only one commercial vaccine in Europe to prevent disease. In this work, inactivated CHV cultures were inoculated in rabbits, adsorbed and not adsorbed to chitosan nanoparticles. Phagocytosis in the presence or absence of specific antibodies was measured. Response of virus neutralizing antibodies was also evaluated. AdimodTM enhanced the nonspecific and specific phagocytotic response. The association of the virus to the nanoparticles increased the phagocytic ability of blood cells;however, AdimodTM alone had a greater effect on phagocytic activity and generated a stronger immune response that corresponded to the increased phagocytosis (p TM was used.

Objective: The aim of study was to evaluate the nature of mesiodistal angulation of the posterior teeth in patients with maxillary high Canine, and to identify possible associations of crowding with mesial axial angulation of the maxillary posterior teeth. Methods: The present study included seven Japanese male and 23 Japanese female patients. Study models were analyzed to evaluate the effectiveness of orthodontic treatment. A protractor was used to measure crown angulations, which were measured with reference to the posterior angle between the functional occlusal plane (FOP) and the long axis of the clinical crown of the lateral teeth and first molars. A sliding digital caliper was used to measure dental arch widths. Changes in the rotation of the maxillary molars were analyzed using “occlu-sograms”, which were printed from photographs of the occlusal aspect of the study model. Results: The maxillary teeth interacted with one another in the alveolar bone, resulting in progressive mesial tipping of the posterior teeth before orthodontic treatment. After orthodontic treatment, the angle relative to the FOP exhibited a significant increase, indicating that the maxillary teeth underwent distally directed uprighting (P P Conclusions: Based on the results, both uprighting of mesially tipped lateral teeth and expansion of narrow dental arches could prove to be keys to success in space regaining or correction of high Canines and mild crowding.

Aurora kinases play an important role in the cell cycle. These enzymes help establish mitotic spindles by directing centrosome duplication and separation and by regulating the spindle assembly checkpoint thereby helping control cytokinesis. An over-expression of aurora kinases has been reported in a variety of human tumors. In this study, we identified the expression of aurora-A and aurora-B kinases in Canine malignant lymphoid cells. We also evaluated the effects of the aurora kinase inhibitor (ZM447439), and found that this inhibitor decreases cell viability, increases DNA content change, and leads to apoptosis in Canine B- and T-cell lymphoid cell lines. The lymphotoxicity induced by ZM447439 in these Canine lymphoid cell lines suggests that further in vivo evaluation of aurora kinase inhibitors as a potential treatment for Canine malignant lymphoid tumors is warranted.

Potent Anticancer Effect of PET-Fraction (PET-F) in Comparison with Other Commercial Products on Canine Cancer Cells

A variety of nutritional supplements are commercially available for the potential therapeutic purpose for dogs and other domestic animals with cancers. However, the efficacy of these supplements on those cancer-bearing animals has not been verified and remains uncertain, due to the lack of the sufficient scientific studies/data. To address this important issue, we examined actual anticancer effects of five selected commercial products on the two types of Canine cancer models in vitro. Among them, PET-Fraction (PET-F) obtained from maitake mushroom has been shown to have potent anticancer effect on Canine cancer cells. In comparison with this PET-F, possible anticancer effects of the rest of four products were examined. Two of four products were found to have the significant anticancer effects on both cancer cells, while other two products had little effects. Nevertheless, PET-F exhibited the best anticancer effect. Cell cycle analysis indicated that the PET- F-induced growth reduction was attributed to a G1 cell cycle arrest, and Western blot analysis further revealed that such a growth reduction was coupled with apoptosis. Moreover, anticancer activity of PET-F was remarkably enhanced with vitamin C, but no such synergistic effect was seen with other four products. Therefore, although two other products besides PET-F yet have anticancer activities, PET-F appears to be the most potent and promising commercial product that can be useful in effectively and safely treating Canine cancers.

Objective: To collect high quality, representative tissue material from tumors and manage its distribution to different laboratories. Design: Prospective controlled study. Animals: Thirty-six dogs with mast cell tumors. Procedures: The samples were submitted for the following analyses: stereology;histopathology;cell culture;breakdown for cytogenetic analysis of chromosomes (based on the Boxer breed published genome);Cell lysis for Real Time PCR and quantification of gene expression of CX 43, 32 and E-cadherin in Canine mast cells. Results: Cytogenetic chromosome analysis, 90.9% of the samples were considered to be of good quality. For gene expression quantification of CX 43, 32 and E-cadherin in Canine mast cell tumors (MCT), 95.5% of samples were considered to be of good quality. Conclusions and Clinical Relevance: We seek to assess the importance of surgical collection and post-surgical tissue preparation on laboratory testing by collecting surgical material appropriately to allow accurate diagnosis and reliable clinical prognosis and minimize errors caused by inadequate preparation of samples.

Evaluation of Real-Time PCR and Culturing for the Detection of Leptospires in Canine Samples

Validated real-time PCRs detecting leptospires for veterinary purposes are not readily available. This paper describes the prospective evaluation of a SYBR Green-based real-time PCR on serum samples collected from experimentally infected dogs. Compared to culturing, the assay had a diagnostic sensitivity and specificity of 91.7% and 90.0%, respectively. Culturing for part is complementary to PCR and preferably both should be applied for diagnosis and vaccine challenge experiments. In a subsequent prospective study on the dynamics of experimental infections with serovars Canicola and Copenhageni and serovar complex Bananal-Liangguang in young and adult dogs, the PCR was applied on serum samples in conjunction with culturing on blood, urine and kidney samples with the following results: 1) Leptospires persisted longer in the blood of young dogs than of adult ones;2) Numbers of viable leptospires in the blood are rapidly reduced but DNA remains occasionally detectable up to 7 days post infection;3) Appearance of viable leptospires in the urine follows a biphasic dynamics;4) EDTA hampers effective culturing from blood samples;5) Serovar Canicola persists longer in the blood of dogs than Copenhageni and Bananal-Liangguang. Together with a markedly higher recovery rate from kidney samples (71% compared to respectively 33% and 0% in young dogs), this probably reflects the adaptation power of Canicola to its Canine maintenance host. Appearance of leptospires in the urine samples indicates that experimental infections have been successful. PCR presents a valuable adjunct to the diagnosis of veterinary leptospirosis and the follow-up in vaccine protection experiments.
