Objective The present study eval.ated the antinociceptive activity of Cal.ndul. officinal.s l.(Ca)cream on infl.mmatory *** Creams with different Ca concentrations were tested for their abil.ty to amel.orate pain-rel.ted behavior and edema in rats using formal.n test,carrageenan(Cg)-induced acute infl.mmation model.bradykinin(BK)-induced acute infl.mmation model.and compl.te Freund’s adjuvant(CFA)-induced chronic infl.mmation *** the formal.n test,we al.o examined the impl.cation of peripheral.opioid receptors in the antinociceptive mechanisms of Ca cream,by means of Q-nal.xone,a peripheral.y acting nonsel.ctive opioid ***,the effects of Ca cream compared with dicl.fenac on BK-induced edema were examined when the kininase II in tissue was preserved or inhibited by *** l.cal.production of redox biomarkers in formal.n model.tumor necrosis factor-α(TNF-α)in Cg model.and histopathol.gical.changes in CFA model.were al.o *** A singl. appl.cation of Ca cream at a dose of 10%or 30%(w/w)decreased the formal.n-induced l.cking/biting behavior during both phases of this test in a Q-nal.xone-sensitive *** effect was associated with the reduction of oxidative stress in the injured paw and the edema associated with the l.ter phase of formal.n-induced pain.A singl. appl.cation of Ca cream(10%or 30%)reduced paw edema and thermal.hypernociception in Cg-induced acute infl.mmation,corresponding with a l.cal.decrease in TNF-α.Ca cream al.o inhibited BK-induced spontaneous nociceptive behavior and paw infl.mmation in both paradigms *** appl.cations of Ca cream al.o decreased CFA-induced chronic infl.mmation,mechanical.hypernociception and histopathol.gical.changes in the *** These resul.s reveal.the topical.antinociceptive and antiedematogenic effects of Ca cream.A modul.tory action on peripheral.opioid receptors associated with its antioxidant mechanism may be invol.ed,at l.ast in par
