Thrombolytic activity of cheonggukjang kinase in recovery from brain damage in a rat cerebral embolic stroke model

Cheonggukjang is a soybean paste made by fermenting whole cooked soybeans with Bacillus subtilis. Cheonggukjang contains a fibrinolytic enzyme that could provide clinical applications for removing blood clots. In the present study, the term ＂cheonggukjang kinase＂（CGK） was used to refer to this fibrinolytic enzyme. The thrombolytic effects of CGK were analyzed in a rat model of cerebral embolic stroke produced by middle cerebral artery occlusion （MCAO）. Results from fibrin and platelet-rich clot lysis assays demonstrated that thrombolytic activity was greatest in CGKs, which were cultured for 40 hours. In addition, T50, the time needed to decompose 50% of the clot, did not change with plasminogen treatment, indicating that CGK was not a plasminogen activator, but was rather presumed to act as a plasmin-like protein. An intravenous infusion of CGK （1 U plasmin-like activity/100 μg CGK/kg） at 1 hour after MCAO resulted in removal of clots in a rat model of cerebral embolic stroke. CGK-treated groups exhibited a significant dose-dependent reduction in infarct volume. CGK treatment also improved functional recovery, as assessed by neurological deficit scores. Decreased infarct volume and improved functional recovery following CGK treatment was greater compared with recombinant tissue plasminogen activator （10 mg/kg）. These results suggested that CGK effectively reduced infarct volume and improved functional recovery following ischemic brain injury. CGK exhibits a number of potential clinical applications ir the treatment of cerebral embolic stroke.

Negative regulation of miRNA-9 on oligodendrocyte lineage gene 1 during hypoxic-ischemic brain damage

Oligodendrocyte lineage gene 1 plays a key role in hypoxic-ischemic brain damage and myelin repair, miRNA-9 is involved in the occurrence of many related neurological disorders. Bioin- formatics analysis demonstrated that miRNA-9 complementarily, but incompletely, bound oligodendrocyte lineage gene 1, but whether miRNA-9 regulates oligodendrocyte lineage gene 1 remains poorly understood. Whole brain slices of 3-day-old Sprague-Dawley rats were cultured and divided into four groups： control group; oxygen-glucose deprivation group （treatment with 8% O2 ＋ 92% N2 and sugar-free medium for 60 minutes）; transfection control group （after oxygen and glucose deprivation for 60 minutes, transfected with control plasmid） and miRNA-9 transfection group （after oxygen and glucose deprivation for 60 minutes, transfected with miRNA-9 plasmid）. From the third day of transfection, and with increasing culture days, oligodendrocyte lineage gene 1 expression increased in each group, peaked at 14 days, and then decreased at 21 days. Real-time quantitative PCR results, however, demonstrated that oligoden- drocyte lineage gene 1 expression was lower in the miRNA-9 transfection group than that in the transfection control group at 1, 3, 7, 14, 21 and 28 days after transfection. Results suggested that miRNA-9 possibly negatively regulated oligodendrocyte lineage gene 1 in brain tissues during hypoxic-ischemic brain damage.

Role of Toll-like receptor 4 and Janus kinase and signal transducer and activator of transcription signal transduction pathway in sepsis-induced brain damage

The Janus kinase and signal transducer and activator of transcription （JAK/STAT） signal transduction pathway is involved in sepsis-induced functional damage to the heart, liver, kidney, and other organs. However, the cellular and molecular mechanisms underlying sepsis-induced brain damage remain elusive. In the present study, we found severe loss of neurons in the hippocampal CA1 region in rats with sepsis-induced brain damage following intraperitoneal injection of endotoxin, The expression of toll-like receptor 4, tumor necrosis factor a, and interleukin-6 was significantly increased in brain tissues following lipopolysaccharide exposure. AG490 （JAK2 antagonist） and rapamycin （STAT3 antagonist） significantly reduced neuronal loss and suppressed the increased expression of toll-like receptor 4, tumor necrosis factor a, and interleukin-6 in the hippocampal CA1 region in sepsis-induced brain damaged rats. Overall, these data suggest that blockade of the JAK/STAT signal transduction pathway is neuroprotective in sepsis-induced brain damage via the inhibition of toll-like receptor 4, tumor necrosis factor a, and interleukin-6 exoression.

Role of gap junction and connexin-43 in hypoxic-ischemic brain damage

OBJEETIVE ： Gap junction （GJ） is the structural basis tor direct intercellular communication OT nerve CellS. Connexin （Cx） is the protein subunit for constructing GJ channel. Among them, Cx43 is closely related with nervous system. Both Cx43 and nervous system play an important role in the pathophysiological development of hypoxic-ischemic injury. We are in attempt to investigate G J, Cx43 and their correlations with hypoxic-ischemic brain damage by research. DATA SOURCES ： Using the terms ＂ brain gap junction＂ in English and ＂gap junction＂ in Chinese, we searched the Medline database and Chinese BioMedical Literature Database as well as China Hospital Knowledge Database to identify the articles published from 1996 to 2006 about GJ and brain hypoxic-ischemic injury. STUDY SELECTION ： The articles were selected firstly and abstracts of 250 articles were read through. Articles in which the experimental design met randomized controlled principle were included, and study articles and case reports with repetitive contents were excluded. DATA EXTRACTION ： Among 53 included correlative articles, 23 were excluded for repetitive contents and the other 30 were analyzed. DATA SYNTHESIS： GJ, widely existing in nervous system, plays a key role in maintaining normal differentiation and development as well as physiological function of brain tissue. GJ channel is a hydrophilic, Iow-selectivity and lowohmic channel, which can provide direct channel for intercellular substance transmission and information communication. It plays an important role in the differentiation and development of nerve cells and regulation of physiological function. The functions of GJ channel are regulated by many factors, which involved intracellular pH value, Ca^2＋ concentration, ATP concentration, phosphorylation of Cx, transchannel pressure, some neurohormonal factors, regulatory factors of protein and so on. Level of serum neuron-specific enolase and brain damage in children with febrile seizures

BACKGROUND ： Febrile seizure （FS） has good prognosis in the majority of cases. But there is an ongoing debate on the relationship between complicated febrile seizure （CFS） and later development of temporal lobe epilepsy （TLE） due to hippocampal sclerosis.OBJECTIVE： To evaluate the level of serum neuron-specific enolase （S-NSE） of children with simple febrile seizure （SFS） and complicated febrile seizure and compare with children with non-FS respiratory tract infection. DESIGN： Contrast observation.SETTING ： Department of Pediatrics and Department of Laboratory, Fujian Provincial Hospita. PARTICIPANTS： Forty-nine patients who were admitted to Department of Pediatrics of Fujian Provincial Hospital from June 2002 to September 2003 with FS were enrolled in this study. There were 28 boys and 21 girls aged from 5 to 72 months. All children were divided into 2 groups based on frequency and duration. Thirty-two children whose FS occurred within 24 hours （lasting shorter than single and twice durations and also shorter than 10 minutes） were regarded as SFS group; meanwhile, 17 children whose FS occurred within 24 hours （lasting longer than single and twice durations and also longer than 10 minutes） were regarded as CFS group. Another 23 patients who were admitted to our hospital with respiratory tract infection in the same period, without the history and positive symptoms/features of neurological dysfunction, were enrolled as control group. There were 13 boys and 10 girls aged from 5 months to 86 months. All parents were told the facts. METHODS： 2 mL venous blood was collected from FS children within 24 hours after the seizures. Meanwhile, 2 mL venous blood was also collected from children in the control group. Level of S-NSE was measured with enzyme immunoassay procedure, and differences among groups were compared with ttest. MAIN OUTCOME MEASURES： Level of S-NSE in each group RESULTS： A total of 49 children in FS group and 23 ones in control group were involved

Changes of neuronal calcium channel following brain damage induced by injection of pertussis bacilli in rats

Objective: To explore changes of neuronal calcium channel following brain damage induced by injection of pertussis bacilli in rats, and to investigate the relationship between cytosolic free calcium concentration ([Ca 2+ ] i) in the synaptosome and Ca 2+ ATPase activities of mitochondria. Methods: The level of [Ca 2+ ] i in the synaptosome and Ca 2+ ATPase activities of mitochondria in the acute brain damage induced by injection of pertussis bacilli (PB) in rat was determined and nimodipine was administrated to show its effects on [Ca 2+ ] i in the synaptosome and on alteration of Ca 2+ ATPase activity in the mitochondria. Seventy three rats were randomly divided into four groups, ie, normal control group (Group A), sham operation control group (Group B), PB group (Group C) and nimodipine treatment group (Group D). Results: The level of [Ca 2+ ] i was significantly increased in the PB injected cerebral hemisphere in the Group C as compared with that in the Group A and the Group B at 30 minutes after injection of PB. The level of [Ca 2+ ] i was kept higher in the 4 hours and 24 hours subgroups after the injection in the Group C (Pbrain damage through stimulating the activities of Ca 2+ ATPase in the mitochondria, and decrease the level of [Ca 2+ ] i in the synaptosome. Treatment with nimodipine dramatically reduces the effects of brain damage induced by injection of PB.

Repair of abnormal perfusion foci in idiopathic epilepsy patients under long-term antiepileptic treatment

Epileptic seizure control and the disappearance of epileptJform discharge are not indicative of the absence of abnormal perfusion foci. Perfusion abnormalities are a major cause of epileptic discharge, and the existence of abnormal perfusion loci implies possible relapse. Very little is known about perfusion abnormality repair in epilepsy. The present study selected 43 cases of idiopathic epilepsy under antiepileptic drug control for an average of 24 months. Comparisons between interictal single-photon emission CT （SPECT） images and long-term electroencephalogram （EEG） pre- and post-treatment showed that cases of normal SPECT increased by 48% （12/25） following treatment, with a total number of 15 reduced loci （,36%, 15/41 ）. Perfusion foci, Le., region of interest, were altered following treatment. These changes included： normal to abnormal in 3 cases （7%, 3/43; 2 hyperperfusion and 1 hypoperfusion）; abnormal to normal in 14 cases （32%, 14/43; 10 pre-treatment hypopeffusion and 4 hyperperfusion）; abnormal to abnormal in 7 cases （16%, 7/43; hyperperfusion to hypoperfusion in 5 cases, hypoperfusion to hyperpeffusion in 2 cases）. Long-term EEG revealed in an increase in the number of normal cases by 20 （40%, 20/39）, and there were 25 fewer cases with epileptiform discharges （66%, 25/38）. These findings demonstrate that long-term control of anti-epileptic drugs partially repaired cerebral perfusion abnormalities and reduced epileptiform discharges in idiopathic epilepsy.

Teriflunomide provides protective properties after oxygen-glucose-deprivation in hippocampal and cerebellar slice cultures

One of the major challenges in emergency medicine is out-of-hospital cardiac arrest(OHCA).Every year,about 53–62/100000 people worldwide suffer an out-of-hospital cardiac arrest with serious consequences,whereas persistent brain injury is a major cause of morbidity and mortality of those surviving a cardiac ***,only few and insufficient strategies are known to limit neurological damage of ischemia and reperfusion *** aim of the present study was to investigate whether teriflunomide,an approved drug for treatment of relapsing-remitting-multiple-sclerosis,exerts a protective effect on brain cells in an in vitro model of ***,organotypic slice cultures from rat hippocampus and cerebellum were exposed to oxygen-glucose-deprivation and subsequently treated with *** administration of teriflunomide in the reperfusion time on both hippocampal and cerebellar slice cultures significantly decreased the amount of detectable propidium iodide signal compared with an untreated culture,indicating that more cells survive after ***,hippocampal slice cultures showed a higher vulnerability to ischemic conditions and a more sensitive response to teriflunomide compared with cerebellar slice *** study suggests that teriflunomide,applied as a post-treatment after an oxygenglucose-deprivation,has a protective effect on hippocampal and cerebellar cells in organotypic slice cultures of *** procedures were conducted under established standards of the German federal state of North Rhine Westphalia,in accordance with the European Communities Council Directive 2010/63/EU on the protection of animals used for scientific purposes.

Assessment tools for differential diagnosis of neglect: Focusing on egocentric neglect and allocentric neglect

BACKGROUND There are very few studies on the differential diagnosis between egocentric neglect(EN)and allocentric neglect(AN).AIM To investigate the overall trend of the previously developed assessment tools by conducting a descriptive review of the studies on assessment tools that can perform a differential diagnosis of EN and *** The data were collected by using databases such as Google Scholar,PubMed,and *** most commonly used search terms were“neglect”,“stroke”,“egocentric neglect”,and“allocentric neglect”.RESULTS A total of seven studies that met the inclusion criteria were selected and *** were able to confirm the research process,test method,and differential diagnosis criteria of the seven presented assessment tools from four studies on paper-based tests and three studies on computerized *** majority of the tests were carried out via the cancellation method using stimuli such as everyday objects or *** distinguished the left from right based on the test paper,while AN distinguished the left from right based on *** order to perform differential diagnosis,the difference in the number of left and right responses or non-responses was used based on the EN and AN *** It was confirmed that all the seven assessment tools can effectively perform differential diagnosis of EN and *** study may provide important data that can be used in clinical practice for differential diagnosis and future intervention planning for neglect patients.
