An array of natural products bearing chiral peroxide motifs display antitumor,anticancer,and antiparasitic activities.However,there are very few catalytic asymmetric syntheses of chiral peroxides.Moreover,effective catalysts for asymmetric catalytic peroxidations are limited to chiral amines.To further expand synthetic access to chiral peroxides,the development of new catalysts realizing catalytic asymmetric peroxidation is highly desirable yet challenging.We report here a catalytic asymmetric peroxidation of anα,β-unsaturated triflone-kinetic resolution cascade reaction,which furnishes chiral peroxides in greater than 99.9%ee.The realization of this cascade reaction resulted from the development of two betaines as novel catalysts for chiral peroxide synthesis;one betaine promoted enantioselective peroxidation ofα,β-unsaturated triflones via conjugate addition affording peroxides in 80-92%ee,while the other betaine catalyzed kinetic resolution of the newly generated chiral peroxides to further increase the ee to greater than 99.9%.

Background:Nucleic acid amplification enhancers suitable for use in a recombinase-aided amplification(RAA)assay were studied for the first time,and amplification of a long-fragment(509 bp)was initially explored.Methods:Using recombinant plasmids and clinical samples,RAA fluorescence and basic methods were used to evaluate the efficacy.The fluorescence method was evaluated by threshold time and fluorescence value,and the basic method was characterized by 2%agarose gel electrophoresis.Results:Taking a previously established RAA assay for HPV18 as an example,we demonstrated that the addition of 0.2 M,0.4 M,and 0.6 M betaine and 10%pullulan could enhance the RAA.The new RAA assays with betaine and pullulan were named B-RAA and P-RAA,respectively.Using the B-RAA and P-RAA fluorescence methods,the threshold time values could be shortened by 1.72-2.32 minutes and 2.60 minutes,respectively,and the fluorescence values could be enhanced by 8847.25-9094.37 mv and 5250 mv,respectively.Using the basic method,the sensitivity could be increased 10-fold.We successfully amplified a long-fragment of 509 bp using a P-RAA assay with a sensitivity of 102 copies/μL(compared with 103 copies/μL in the RAA assay).Conclusions:Thus,we concluded that betaine and pullulan are effective additives to enhance the sensitivity of RAA assays.

betaine has been demonstrated to improve growth performance and antioxidant status of animals under various stress conditions.However,there is no literature on the effects of betaine in animals exposed to mycotoxins,which are among the most prevalent contaminants in feed.Therefore,this study was conducted to evaluate the influence of dietary betaine on broilers fed a diet based on mold-contaminated corn(MCC).A total of 192 Ross 308 male broiler chicks at 1 d of age were randomly divided into 4 groups with 6 replicates and fed an MCC-based diet supplemented with 0,250,500,and 1,000 mg/kg betaine,respectively.betaine increased average daily gain(linear,P=0.030)and decreased feed conversion ratio(linear,P=0.027)of broilers during d 1-21,and decreased feed conversion ratio during d 22-42(linear,P=0.012;quadratic,Pbetaine.betaine linearly increased breast muscle yield(P=0.003)and pH24 h(P=0.008),and decreased drip loss(P=0.022).betaine increased(linear,P=0.025;quadratic,P=0.016)total superoxide dismutase activity in breast muscle and reduced malondialdehyde content in serum(linear,P=0.006),liver(quadratic,P=0.006)and breast muscle(linear,P=0.003).Moreover,the zearalenone concentrations in breast muscle were linearly decreased by betaine(P=0.006).It was concluded that betaine could improve growth performance,liver health,antioxidant status,and breast meat yield and quality,and reduce zearalenone residue in broilers fed the MCC-based diet,especially at 500 or 1,000 mg/kg.

Background： betaine affects fat metabolism in animals, but the specific mechanism is still not clear. The purpose of this study was to investigate possible mechanisms of betaine in altering lipid metabolism in muscle tissue in finishing pigs.Methods： A total of 120 crossbred gilts（Landrace × Yorkshire × Duroc） with an average initial body weight of 70.1 kg were randomly allotted to three dietary treatments. The treatments included a corn–soybean meal basal diet supplemented with 0, 1250 or 2500 mg/kg betaine. The feeding experiment lasted 42 d.Results： betaine addition to the diet significantly increased the concentration of free fatty acids（FFA） in muscle（P 〈 0.05）. Furthermore, the levels of serum cholesterol and high-density lipoprotein cholesterol were decreased（P 〈 0.05） and total cholesterol content was increased in muscle（P 〈 0.05） of betaine fed pigs. Experiments on genes involved in fatty acid transport showed that betaine increased expression of lipoprotein lipase（LPL）, fatty acid translocase/cluster of differentiation（FAT/CD36）, fatty acid binding protein（FABP3） and fatty acid transport protein（FATP1）（P 〈 0.05）. The abundance of fatty acid transport protein and fatty acid binding protein were also increased by betaine（P 〈 0.05）. As for the key factors involved in fatty acid oxidation, although betaine supplementation didn＇t affect the level of carnitine and malonyl-CoA, betaine increased mR NA and protein abundance of carnitine palmitransferase-1（CPT1）and phosphorylated-AMPK（P 〈 0.05）.Conclusions： The results suggested that betaine may promoted muscle fatty acid uptake via up-regulating the genes related to fatty acid transporter including FAT/CD36, FATP1 and FABP3. On the other hand, betaine activated AMPK and up-regulated genes related to fatty acid oxidation including PPARα and CPT1. The underlying mechanism regulating fatty acid metabolism in pigs supplemented with betaine is associated with the up-regulation of genes involved in fatty acid transport and fatty acid oxidation.

AIM:To test whether ethanol feeding could induce Toll-like receptor 4(TLR4)responses,assess the hepatoprotective effect of betaine and its inhibitive effect on TLR4 in animal models of alcoholic liver injury.METHODS:Forty-eight female Sprague-Dawley rats were randomly divided into four groups as control,model,low and high dose betaine groups.Except control group,all rats were fed with high fat-containing diet plus ethanol and fish oil gavages for 8 wk.betaine was administered intragastrically after exposure of ethanol for 4 wk.The changes of liver histology were examined.The expression of TLR4 mRNA and protein was detected by RT-PCR and Western blotting,respectively.The serum aminotransferase activity alanine transarninase(ALT),aspartate aminotransferase(AST),serum endotoxin,and liver inflammatory factors tumor necrosis factor-α(TNF-α),interferon-γ(IFN-γ),interleukin-18(IL-18)were also assayed.RESULTS:Compared with control group,rats of model group developed marked liver injury,accompanied by an increase of ALT(159.41±7.74 U/L vs 59.47± 2.34 U/L,Pbetaine feeding resulted in significant decreases of ALT(64.93 ±6.06 U/L vs 159.41±7.74 U/L,Pbetaine also improved liver steatosis.The expression levels of TLR4 mRNA or protein in liver tissues were significantly

The objective of the study is to evaluate and compare the effects of betaine or glycine on carcass trait,meat quality and lipid metabolism of finishing Huan Jiang mini-pigs.betaine called trimethylglycine is a methyl derivative of glycine,but few researches were conducted to compare the impact of dietary betaine and glycine on pigs.One hundred and forty-four Huan Jiang mini-pigs(body weight=10.55±0.15 kg;70 d)were randomly divided to 3 treatment groups(basal diet,glycine or betaine).Results indicated that dietary betaine increased the average daily gain(ADG)and final weight(Pbetaine markedly reduced average backfat thickness(Pbetaine significantly improved the redness(a*)and tenderness(shear force)of the longissimus dorsi(LD)muscle(Pbetaine and equimolar amounts of glycine could regulate cascass trait and meat quality of finishing Huan Jiang mini-pigs,and the effect of betaine was superior to that of glycine.

A potential adsorbent based on betaine-modified magnetic iron oxide nanoparticles(BMNPs) was successfully synthesized by facile method, characterized and applied for methyl blue(MB) removal from aqueous solution. The characterization results of FTIR, transmission electron microscopy(TEM), X-ray diffraction(XRD) and vibrating sample magnetometer(VSM) showed that the prepared nanoparticles could be well dispersed in water and exhibited excellent superparamagnetism. These properties imply the potential to recycle BMNPs from wastewater through magnetic field. In the adsorption process, the effects of main experimental parameters such as p H of MB solution, initial concentration of MB, contact time, and adsorption capacity for MB were studied and optimized. These results demonstrated that large amounts of quaternary ammonium groups existing on the surface of BMNPs could promote absorption of MB via electrostatic forces. Additionally, the adsorption kinetics of MB was found to follow a pseudosecond-order kinetic model and the adsorption equilibrium data fitted very closely to the Langmuir adsorption isotherm model. The maximum adsorption capacity for MB was calculated to be 136 mg g 1at room temperature. Moreover, the BMNPs showed good reusability with 73.33% MB adsorption in the5 th cycle.

Sow performance in response to natural betaine fed during lactation and post-weaning during summer and non-summer months

Background: Two studies were conducted to evaluate the effects of dietary natural betaine on sow reproductive performance during summer(Exp. 1) and non-summer months(Exp. 2). Treatments were designed as a 2 × 2 factorial arrangement with factors including dietary betaine(0 or 0.2%) and period of supplementation(lactation or post-weaning until 35 days post-insemination). In Exp. 1, 322 and 327 sows and in Exp. 2, 300 and 327 sows representing young(parity 1 and 2) and mature(parity 3 to 6) sows, respectively, were used.Results: In Exp. 1, supplementation of betaine during lactation increased sow body weight losses(-11.95 vs.-14.63 kg;P = 0.024), reduced feed intake(4.12 vs. 4.28 kg/d;P = 0.052), and tended to reduce percentage of no-value pigs(P = 0.071). betaine fed post-weaning reduced weaning-to-estrus interval(5.75 vs. 6.68 days;P = 0.054) and farrowing rate(86.74% vs. 91.36%;P = 0.060), regardless of parity group. Post-hoc analysis with sows clustered into 3 parity groups(1, 2 and 3, and 4+) indicated that betaine fed in lactation to parity 4+ sows(P = 0.026) and betaine fed post-weaning to parity 1 sows increased the number of pigs born in the subsequent cycle(P ≤ 0.05). In Exp. 2, betaine fed during lactation tended to reduce the weaning-to-estrus interval(6.64 vs. 7.50 days;P = 0.077) and farrowing rate(88.23% vs.83.54%;P = 0.089), regardless of parity group. Feeding betaine post-weaning reduced number of pigs born(13.00 vs.13.64;P = 0.04) and pigs born alive(12.30 vs. 12.82;P = 0.075), regardless of parity group.Conclusions: Using 0.2% betaine during the non-summer months did not benefit sow performance. During the summer, betaine supplementation in lactation increased subsequent litter size in parity 4+ sows. betaine fed during the post-weaning period reduced the wean-to-estrus interval and farrowing rate, increased total number of pigs born for parity 1 sows and reduced total number of pigs born to parity 4+ sows. Background:Laying hens supplemented with betaine demonstrate activated adrenal steroidogenesis and deposit higher corticosterone(CORT)in the egg yolk.Here we further investigate the effect of maternal betaine on the plasma CORT concentration and adrenal expression of steroidogenic genes in offspring pullets.Results:Maternal betaine significantly reduced(Pbetaine-supplemented hens.Furthermore,expression of transcription factors,such as steroidogenic factor-1,sterol regulatory element-binding protein 1 and c AMP response element-binding protein,was significantly(Pbetaine-supplemented offspring pullets is linked to suppressed cholesterol trafficking into the mitochondria,despite the activation of cholesterol and corticosteroid synthetic genes associated with promoter hypomethylation.
