Developing a novel quantitative parameter for characterizing spatial distribution of fish following exposure to chemicals and wastewater:behavioral Gini coefficient

While the spatial distribution pattern of fish is increasingly used for toxicological test o chemicals or wastewater,no ideal parameter is available for quantitative assessment of spa tial distribution,especially uneven distribution with multiple ***,to develop a quantitative assessment parameter for spatial distribution,the zebrafish were exposed to ethanol,pentylenetetrazole(PTZ),paraquat dichloride(paraquat)and wastewater,followed by a behavioral test in a narrow *** data was acquired and analyzed by id Tracker and *** comparing the effects of all treatments on behavior parameters we confirmed that the spatial distribution was more easily altered rather than general loco motor parameters,e.g.0.7-70 mg/L PTZ and 5-20 mg/L paraquat being effective for altering spatial distribution but having little effects on general locomotor *** on the heatmap,i.e.,the cumulative proportion of grids and that of frequency in grids,we calcu lated the behavioral Gini coefficient(G_(b))for quantitative assessment of fish spatial distri *** Gini coefficient ranged from zero to 1,with larger values meaning poorer even ness of spatial *** note,G_(b)showed smaller coefficient of variations(CV)with3%-19%between replicate tanks in all treatments than the highest frequency(4%-79%),dis playing well ***,G_(b)addressed the challenge of the complicated heatmap with multiple ***,the behavioral Gini coefficient we established is an idea parameter to quantitatively assess spatial distribution of fish shoal,which is expected to be applied in toxicity testing for chemicals and wastewater and automatic quality monitoring for surface water and aquaculture water.

A modified light-dark box test for the common marmoset

The common marmoset （Callithrix jacchus） has attracted extensive attention for use as a non-human primate model in biomedical research, especially in the study of neuropsychiatric disorders. However, behavioral test methods are still limited in the field of marmoset research. The light-dark box is widely used for the evaluation of anxiety in rodents, but little is known about light-dark preference in marmosets. Here, we modified the light-dark test to study this behavior. The modified apparatus consisted of three compartments： one transparent open area and two closed opaque compartments. The closed compartments could be dark or light. We found that both adult and young marmosets liked to explore the open area, but the young animals showed more interest than adults. Furthermore, when one of the closed compartments was light and the other dark, the adult marmosets showed a preference for the dark compartment, but the young animals had no preference. These results suggest that the exploratory behavior and the light-dark preference in marmosets are age-dependent. Our study provides a new method to study exploration, anxiety, and fear in marmosets.

Biochemical and behavioral characterization of the double transgenic mouse model (APPswe/PS1dE9) of Alzheimer’s disease

Objective The double transgenic mouse model （APPswe/PSldE9） of Alzheimer＇s disease （AD） has been widely used in experimental studies. β-Amyloid （Aβ） peptide is excessively produced in AD mouse brain, which affects synaptic function and the development of central nervous system. However, little has been reported on characterization of this model. The present study aimed to characterize this mouse AD model and its wild-type counterparts by biochemical and functional approaches. Methods Blood samples were collected from the transgenic and the wild-type mice, and radial arm water maze behavioral test was conducted at the ages of 6 and 12 months. The mice were sacrificed at 12-month age. One hemisphere of the brain was frozen-sectioned for immunohistochemistry and the other hemisphere was dissected into 7 regions. The levels ofAβ1-40, Aβ1-42 and 8-hydroxydeoxyguanosine （8-OHdG） in blood or/and brain samples were analyzed by ELISA. Secretase activities in brain regions were analyzed by in vitro assays. Results The pre-mature death rate of transgenic mice was approximately 35% before 6-month age, and high levels of Aβ1-40 and Aβ1-42 were detected in these dead mice brains with a ratio of 1 ： 1 0. The level of blood-borne Aβ at 6-month age was similar with that at 12-month age. Besides, Aβ1-40 level in the blood was significantly higher than Aβ1-42 level at the ages of 6 and 12 months （ratio 2.37：1）. In contrast, the level of Aβ1-42 in the brain （160.6 ng/mg protein） was higher than that of Aβ1-40 （74 ng/mg protein）（ratio 2.17：1）. In addition, the levels of Aβ1-40 and Aβ1-42 varied markedly among different brain regions. Aβ1-42 level was significantly higher than Aβ1-40 level in cerebellum, frontal and posterior cortex, and hippocampus. Objective The common marmoset（Callithrix jacchus） has attracted much attention for application as a powerful non-human primate model in recent neuroscience research. In the present study, we conducted a direct comparison of the cognitive function performance between the young and aged common marmosets to explore the relationship between the age and cognitive ability of common marmoset. Methods Ten aged and four young marmosets were trained to take a two-alternative forced-choice cognitive function task by using the modified apparatus in their home cages. After food training and nondelay stages, the delay intervals between presentations of the comparison stimuli were fixed at 0 and 3 s in the initial phases in a session. The task content within a set was gradually increased after the marmosets achieved the pre-established criterion levels of each task. Results The results demonstrated that the young marmosets performed better and learned more quickly in the cognitive tasks, compared to the aged marmosets. Hence, these results reflect that the behavior in marmosets is likely age-dependent. Conclusion The current study provides novel insights of cognitive function performance in marmosets.

Objective Autism spectrum disorder（ASD） is a highly heterogeneous neurodevelopmental disorder characterized by impaired social communication, higher anxiety and cognitive deficits. Emerging studies increasingly suggest that immune dysfunction is a viable risk factor contributing to the neurodevelopmental deficits observed in ASD. Molecule hydrogen has anti-oxidative and anti-inflammatory activities and neuroprotective effects. However, the potential role of molecular hydrogen in autistic behaviors are still elusive. The present study aims to investigate the effects of molecule hydrogen on autistic behaviors in the mice. Methods Mice were free access with hydrogen-rich water（HRW） before and/after the valproic acid（VPA） treatment during the pregnant and postnatal stages. Open filed test（OFT）, novelty suppressed feeding（NSF）, social interaction test（SI） and contextual fear conditioning（CFC） were used to assess the behavioral deficits in VPA-exposed mice offspring with or without HRW treatment. Results The results showed that VPA-exposed mice offspring exhibited significant autistic behaviors. HRW treatment could significantly rescue the behavioral deficits, mainly including the decreased time in central zone in the OFT, the increased latency time to feeding in the NSF test, the decreased social index in the SI test and impairment of memory in the CFC in VPA-exposed developmental mice offspring. These results warrant further exploration for developing molecular hydrogen as a novel strategy to prevent the occurrence of autism and related disorders.
