Oils with different degree of saturation:effects on ileal digestibility of fat and corresponding additivity and bacterial community in growing pigs

Background Oils are important sources of energy in pig *** combination of oils with different degree of saturation contributes to improve the utilization efficiency of the mixed oils and may reduce the cost of oil *** experiment was conducted to evaluate the effects of oils with different degree of saturation on the fat digestibility and corresponding additivity and bacterial community in growing *** Eighteen crossbred(Duroc×Landrace×Yorkshire)barrows(initial body weight:29.3±2.8 kg)were surgically fitted with a T-cannula in the distal *** experimental diets included a fat-free basal diet and 5 oil-added *** 5 oil-added diets were formulated by adding 6%oil with different ratio of unsaturated to saturated fatty acids(U:S)to the basal *** 5 oils were palm oil(U:S=1.2),canola oil(U:S=12.0),and palm oil and canola oil were mixed in different proportions to prepare a combination of U:S of 2.5,3.5 and 4.5,*** The apparent and standardized ileal digestibility(AID and SID)of fat and fatty acids increased linearly(P<0.05)as the U:S of dietary oils increased except for SID of fat and C18:*** AID and SID of fat and fatty acids differed among the dietary treatments(P<0.05)except for SID of unsaturated fatty acids(UFA)and C18:*** one-slope broken-line analyses for the SID of fat,saturated fatty acids(SFA)and UFA indicated that the breakpoint for U:S of oil was 4.14(R^(2)=0.89,P<0.01),2.91(R^(2)=0.98,P<0.01)and 3.84(R^(2)=0.85,P<0.01),*** determined SID of fat,C18:1,C18:2 and UFA in the mixtures was not different from the calculated SID of fat,C18:1,C18:2 and ***,the determined SID of C16:0,C18:0 and SFA in the mixtures were greater than the calculated SID values(P<0.05).The abundance of Romboutsia and Turicibacter in pigs fed diet containing palm oil was greater than that in rapeseed oil treatment group,and the two bacteria were negatively correlated with SID of C16:0,C18:0 and SFA(P<0.05).

Response of bacterial community and Enzyme Activity of Greenhouse Tomato under Different Irrigation Systems

bacterial community response to petroleum contamination in brackish tidal marsh sediments in the Yangtze River Estuary, China

The brackish tidal marsh in the Baimaosha area of the Yangtze River Estuary was severely contaminated by 400 tons of heavy crude petroleum from a tanker that sank in December *** spill accident led to severe environmental damage owing to its high toxicity,persistence and wide *** communities play vital roles in petroleum degradation in marsh ***,taxonomic analysis,high-throughput sequencing and 16 S rRNA functional prediction were used to analyze the structure and function of microbial communities among uncontaminated(CK),lightly polluted(LP),heavily polluted(HP),and treated(TD)*** bacterial communities responded with increased richness and decreased diversity when exposed to petroleum *** dominant class changed from Deltaproteobacteria to Gammaproteobacteria after petroleum *** phylum Firmicutes increased dramatically in oil-enriched sediment by 75.78%,346.19%and 267.26%in LP,HP and TD,*** of the suspected oil-degrading genera,Dechloromonas,increased the most in oil-contaminated sediment,by 540.54%,711.27%and 656.78%in LP,HP and TD,*** protease,quinate dehydrogenase(quinone)and glutathione-independent formaldehyde dehydrogenase,three types of identified enzymes,increased enormously with the increasing petroleum *** conclusion,petroleum contamination altered the community composition and microorganism structure,and promoted some bacteria to produce the corresponding degrading ***,the suspected petroleum-degrading genera should be considered when restoring oil-contaminated sediment.

bacterial community succession during the enrichment of chemolithoautotrophic arsenite oxidizing bacteria at high arsenic concentrations

To generate cost-effective technologies for the removal of arsenic from water, we developed an enrichment culture of chemolithoau- totrophic arsenite oxidizing bacteria （CAOs） that could effectively oxidize widely ranging concentrations of As（III） to As（V）. In addition, we attempted to elucidate the enrichment process and characterize the microbial composition of the enrichment culture. A CAOs enrichment culture capable of stably oxidizing As（III） to As（V） was successfully constructed through repeated batch cultivation for more than 700 days, during which time the initial As（iiI） concentrations were increased in a stepwise manner from 1 to 10-12 mmol/L. As（III） oxidation activity of the enrichment culture gradually improved, and 10-12 mmol/L As（III） was almost completely oxidized within four days. Terminal restriction fragment length polymorphism analysis showed that the dominant bacteria in the enrichment culture varied drastically during the enrichment process depending on the As（III） concentration. Isolation and characterization of bacteria in the enrichment culture revealed that the presence of multiple CAOs with various As（Ⅲ） oxidation abilities enabled the culture to adapt to a wide range of As（Ⅲ） concentrations. The CAOs enrichment culture constructed here may be useful for pretreatment of water from which arsenic is being removed.

Soil microbial biomass and community structures are commonly used as indicators for soil quality and fertility. A investigation was performed to study the effects of long-term natural restoration, cropping, and bare fallow managements on the soil microbial biomass and bacterial community structures in depths of 0-10, 20 30, and 40-50 cm in a black soil （Mollisol）. Microbial biomass was estimated from chloroform fumigation-extraction, and bacterial community structures were determined by analysis of 16S rDNA using polymerase chain reaction-denaturing gradient gel electrophoresis （PCR- DGGE）. Experimental results showed that microbial biomass significantly declined with soil depth in the managements of restoration and cropping, but not in the bare fallow. DGGE profiles indicated that the band number in top 0-10 cm soils was less than that in depth of 20-30 or 40-50 cm. These suggested that the microbial population was high but the bacterial community structure was simple in the topsoil. Cluster and principle component analysis based on DGGE banding patterns showed that the bacterial community structure was affected by soil depth more primarily than by managements, and the succession of bacterial community as increase of soil depth has a similar tendency in the three managements. Fourteen predominating DGGE bands were excised and sequenced, in which 6 bands were identified as the taxa of Verrueomicrobia, 2 bands as Actinobacteria, 2 bands as α-Proteobacteria, and the other 4 bands as 8-Proteobacteria, Aeidobacteria, Nitrospira, and unclassified bacteria. In addition, the sequences of 11 DGGE bands were closely related to uncultured bacteria. Thus, the bacterial community structure in black soil was stable, and the predominating bacterial groups were uncultured.

Objective It aims to investigate the changes in composition and structure of bacterial communities de-veloping on biological activated carbon （BAC） particles, and the bacterial functions. Method A pilot plant had been in service for 180 days, aiming to develop bacterial communities on acti-vated carbon naturally. After 180 days of operation, the bacterial communities were determined by dena-turing gradient gel electrophoresis （DGGE） analyses of PCR-amplified 16S rRNA genes. The study on community composition and the phylogenetic relationships of the organisms was complemented by a se-quence analysis of cloned PCR products from 16S rRNA genes. Gas chromatorgaphy-mass （GC-MS） mea-surement was used to determine organic chemical composition of inflow and outflow water on the 300th day. TOC and NH 4 ＋ -N were also tested in this experiment. Results It showed that the stable bacterial structure did not develop on BAC particles until the 9th month during running time of the BAC filter. The communities were finally dominated by Pseudomonas sp., Ba-cillus sp., Nitrospira sp., and an uncultured bacterium. Stable bacterial communities played an important role in removal of NH 4 ＋ -N and total organic carbon （TOC）. Results from gas chromatorgaphy-mass （GC-MS） showed that 36 kinds of chemicals in feed water were eliminated, and concentrations of 5 kinds of chemicals decreased. These chemicals served as nutrients for the dominant bacteria. Conclusion The findings from the study suggested that the stability of microbial structure was beneficial for improving NH 4 ＋ -N and TOC removal efficiencies. The dominant bacteria had the advantage of biode-grading a wide range of organic chemicals and NH 4 ＋ -N.

The structures of bacterial communities in the brown planthopper （BPH） Nilaparvata lugens （St^l） from different geographic and resistant virulent populations were analyzed by using denatured gradient gel electrophoresis （DGGE）. Results showed that the bacterial communities in BPH nymph from the first to the fifth instars varied with nymphal growth and development. The bacterial communities in the first-instar BPH nymph were similar to those in adults. Nine geographic BPH populations were divided into three groups based on the cluster analysis of DGGE fingerprint. The first group was from the Philippines; the second group was from Thailand and Hainan, Yunnan and Zhejiang provinces of China; and the third group was from Vietnam and Guangxi, Hunan and Jiangxi provinces of China. BPH populations adapted to different resistant rice varieties. The BPH populations from Mudgo （with resistant gene Bphl） and ASD7 （with resistant gene bph2） differed with those of the susceptible rice variety TNI.

Microbial enzymes are crucial for material biotransformation during the composting *** this study,we investigated the effects of adding bamboo charcoal(BC)(i.e.,at 5%,10%,and 20%corresponding to BC5,BC10,and BC20,respectively)on the enzyme activity levels during chicken manure *** results showed that BC10 could increase the cellulose and urease activities by 56%and 96%,*** bacterial community structure in BC10 differed from those in the other treatments,and Luteivirga,Lactobacillus,Paenalcaligenes,Ulvibacter,Bacillus,Facklamia,Pelagibacterium,Sporosarcina,Cellvibrio,and Corynebacterium had the most important roles in *** with other treatments,BC10 significantly enhanced the average rates of degradation of carbohydrates(Dxylose(40%)andα-D-lactose(44%))and amino acids(L-arginine(16%),L-asparagine(14%),and L-threonine(52%)).We also explored the associations among the bacterial community and their metabolic functions with the changes in the activities of *** analysis demonstrated that BC10 altered the co-occurrence patterns of the bacterial communities,where Ulvibacter and class Bacilli were the keystone bacterial taxa with high capacities for degrading carbon source,and they were related to increases in the activities of cellulase and urease,*** results obtained in this study may help to further enhance the efficiency of composting.

Investigating the bacterial community and amoebae population in rural domestic wastewater reclamation for irrigation

Reclamation of domestic wastewater for agricultural irrigation is viewed as a sustainable option to create an alternative water source and address water scarcity. Free-living amoebae（FLA）, which are amphizoic protozoa, are widely distributed in various environmental sources. The FLA could cause considerable environmental and health risks. However, little information is available on the risk of these protozoa. In this study, we evaluated the feasibility using rural domestic wastewater for agricultural irrigation, and analyzed dynamic changes of the microbial community structure and FLA populations in raw and treated wastewater, as well as the phyllosphere and rhizosphere of lettuce production sites that were irrigated with different water sources. The bacterial community dynamics were analyzed by terminal restriction fragment length polymorphism（T-RFLP）. The bacterial community structures in the influent were similar to that in the effluent, while in some cases relative abundances varied significantly. The populations of Acanthamoeba spp. and Hartmannella vermiformis in the anaerobically treated wastewater were significantly higher than in the raw wastewater. The vegetables could harbor diverse amoebae, and the abundances of Acanthamoeba spp. and H. vermiformis in the rhizosphere were significantly higher than in the phyllosphere. Accordingly, our studies show insight into the distribution and dissemination of amoebae in wastewater treatment and irrigation practices.
