Relationship of serum GDF11 levels with bone mineral density and bone turnover markers in postmenopausal Chinese women

Growth differentiation factor 11 （GDF11） is an important circulating factor that regulates aging. However, the role of GDF11 in bone metabolism remains unclear. The present study was undertaken to investigate the relationship between serum GDF11 level, bone mass, and bone turnover markers in postmenopausal Chinese women. Serum GDF11 level, bone turnover biochemical markers, and bone mineral density （bmd） were determined in 169 postmenopausal Chinese women （47-78 years old）. GDF11 serum levels increased with aging. There were negative correlations between GDF11 and bmd at the various skeletal sites. After adjusting for age and body mass index （BMI）, the correlations remained statistically significant. In the multiple linear stepwise regression analysis, age or years since menopause, BMI, GDF11, and estradiol were independent predictors of bmd. A significant negative correlation between GDF11 and bone alkaline phosphatase （BAP） was identified and remained significant after adjusting for age and BMI. No significant correlation was noted between cross-linked N-telopeptides of type I collagen （NTX） and GDF11. In conclusion, GDF11 is an independent negative predictor of bmd and correlates with a biomarker of bone formation, BAP, in postmenopausal Chinese women. GDF11 potentially exerts a negative effect on bone mass by regulating bone formation.

A genome-wide association analysis implicates SOX6 as a candidate gene for wrist bone mass

Osteoporosis is a highly heritable common bone disease leading to fractures that severely impair the life quality of *** fractures caused by osteoporosis are largely due to the scarcity of wrist bone *** we report the results of a genome-wide association study (GWAS) of wrist bone mineral density (bmd).We examined ～500000 SNP markers in 1000 unrelated homogeneous Caucasian subjects and found a novel allelic association with wrist bmd at rs11023787 in the SOX6 (SRY (sex determining region Y)-box 6) gene (P=9.00×10-5).Subjects carrying the C allele of rs11023787 in SOX6 had significantly higher mean wrist bmd values than those with the T allele (0.485:0.462 g cm-2 for C allele vs.T allele carriers).For validation,we performed SOX6 association for bmd in an independent Chinese sample and found that SNP rs11023787 was significantly associated with wrist bmd in the Chinese sample (P=6.41×10-3).Meta-analyses of the GWAS scan and the replication studies yielded P-values of 5.20×10-6 for *** of this study,together with the functional relevance of SOX6 in cartilage formation,support the SOX6 gene as an important gene for bmd variation.

New simulation model for bone formation markers in osteoporosis patients treated with once-weekly teriparatide

Daily 20-mg and once-weekly 56.5-mg teriparatide（parathyroid hormone 1–34） treatment regimens increase bone mineral density（bmd） and prevent fractures, but changes in bone turnover markers differ between the two regimens. The aim of the present study was to explain changes in bone turnover markers using once-weekly teriparatide with a simulation model. Temporary increases in bone formation markers and subsequent decreases were observed during once-weekly teriparatide treatment for 72 weeks. These observations support the hypothesis that repeated weekly teriparatide administration stimulates bone remodeling, replacing old bone with new bone and leading to a reduction in the active remodeling surface. A simulation model was developed based on the iterative remodeling cycle that occurs on residual old bone. An increase in bone formation and a subsequent decrease were observed in the preliminary simulation. For each fitted time point, the predicted value was compared to the absolute values of the bone formation and resorption markers and lumbar bmd. The simulation model strongly matched actual changes in bone turnover markers and bmd. This simulation model indicates increased bone formation marker levels in the early stage and a subsequent decrease. It is therefore concluded that remodeling-based bone formation persisted during the entire treatment period with once-weekly teriparatide.

The objective of this study was to characterize sympathetic nervous system （SNS） activity in girls with adolescent idiopathic scoliosis （AIS） and investigate the correlation of such activity with body weight and bone mass. Fifty-eight girls with AIS and 21 healthy controls were recruited. Abnormal growth pattern （lower weight, lower body mass index （BMI）, and longer arm span） and significantly increased adrenaline （AD） level, norepinephrine （NE） level, and low-frequency （LF） were found in girls with AIS. Circulating AD level, NE level, and LF power were negatively correlated with body weight and BMI but significantly correlated with bone quality.

老年髋部骨折患者骨密度阈值的初步探讨

作者收集３９例６０岁以上髋部骨折病人作为病例组，其中男１７例，女２２例。另选８０例６０岁以上正常老年人作为对照组。其中男女各４０例。采用双能量骨密度仪分别测定其脊柱及髋部各部位的骨密度值。测量结果表明无论是脊柱还是髋部，骨折组骨密度均显著低于对照组（Ｐ＜０．０１）。其中髋部Ｗａｒｄ’ｓ三角区骨密度下降最为明显。骨折组Ｗａｒｄ’ｓ三角区ＢＭＤ值平均为０．４８８８±０．１１０８。作者提出对高危人群进行普查，如发现髋部Ｗａｒｄ’ｓ三角区ＢＭＤ值低于０．５ｇ／ｃｍ２时，对其采取治疗及保护措施是十分必要的。

Osteoporosis,which is characterized by bone loss,increases the risk of fractures and s and seriously affected the amputee’s *** mass is an important factor affecting bone mineral density(bmd).Muscular atrophy and decreased muscle mass can cause bmd to *** bone quantity and quality are used to assess by Microcomputer tomography(micro-CT)with *** investigate the performance of micro-CT in evaluating amputation-induced osteopenia in rats,the micro-CT was used to evaluate the change of the microstructure in trabecular bone and cortical bone of *** morphology was analyzed by micro-CT and transmission electron *** Sprague–Dawley rats(8 week old)were divided into two groups:control(CON)groups and left hind limb amputation(LHLA).At 35 days,microstructure and volumetric bone mineral density(vbmd)of the tibia and femur were detected through *** results showed:(1)the trabecular and cortical vbmd of the proximal tibia and distal femur in the LHLA group were significantly lower than those in the CON group.(2)Compared with the CON group,the trabecular number,bone volume fraction and trabecular thickness of proximal tibia and distal femur in LHLA group decreased significantly,whereas the trabecular separation,structure model index and bone surface/bone volume increased significantly in LHLA group.(3)The whole cross-sectional area(CSA)of whole calf muscles of left limb in LHLA group decreased significantly compared with CON *** general,hind limb amputation could cause bone loss,changes in the microstructure of trabecular bone and muscle ***-CT is sensitive in evaluating bone microstructure destruction caused by amputation,and can be used for early diagnosis or monitor the development of bone loss in amputation in vivo.

目的了解不同的年龄绝经后妇女不同体重质量指数(BMI)与不同部位骨密度(bmd)的关系.方法来自门诊健康体检的557名绝经后妇女(肝肾等疾病除外),年龄范围50～78岁.用双能X线吸收法(DEXA)测定腰椎(L2-4)、股骨颈、Ward's三角及Troch等部位的bmd,同时计算BMI.根据BMI将研究对象分为3组:低体重组(BMI≤20 kg/m2)、正常体重组(25≥BMI＞20kg/m2)及超重组(BMI＞25 kg/m2).根据年龄又分为3组(50～,60～,70～78岁),用方差分析的方法进行各组间均数的统计学分析.结果不同BMI组总体不同部位的bmd方差分析比较有极显著意义P＜0.01,高BMI组的bmd均值明显增高;进一步对同一年龄组不同BMI组的不同部位的bmd均值比较,各组之间均有显著性差异(P＜0.01),特别是低体重组的骨密度明显低于其他两组BMI组的bmd.结论BMI与绝经后妇女的骨密度有显著相关,但低体重的绝经后妇女作为骨折的危险人群应受到更多的关注:对于超重的绝经后妇女,通过增加体重的方式增加bmd是危险的,因为超重与高血压、冠心病、糖尿病、心肌梗死及中风有一定的关系;通过其他途径:如年轻时加强运动、多饮牛奶、不盲目减肥等,提高妇女峰值骨含量是非常重要的.

目的　本文旨在探明绝经后妇女脊椎压缩性骨折的高发部位、患病率随年龄变化的规律以及ＢＭＤ、ＢＭＣ等指标的变化规律。方法　以居住在成都地区具有正式户口年龄在５０～８９岁的健康人群为调查对象，进行随机抽样。对每名受试者进行胸腰椎侧位摄片，若楔形指数（椎体前缘／后缘）≤０．８即诊断为压缩性骨折，同时采用ＤＸＡ对全部受试者腰椎正位及髋部的ＢＭＤ进行测定。计算不同部位及不同年龄段的脊椎压缩性骨折的患病情况。配对后对骨折组和对照组的ＢＭＤ及其他各项指标进行ｔ检验。结果　胸椎压缩性骨折患病率为８．４９％，占脊椎压缩性骨折的７６．１％，其中Ｔ６、Ｔ７、Ｔ１１、Ｔ１２的患病率高，共占胸椎压缩性骨折的６９．１４％；腰椎压缩性骨折的患病率相对较低，占脊椎压缩性骨折的２３．９％，主要集中于Ｌ１、Ｌ２，占腰椎压缩性骨折的９３．５５％。脊椎压缩性骨折的患病率随年龄的增加而逐渐上升。骨折组腰椎ＢＭＤ、ＢＭＣ明显低于对照组（Ｐ＜０．０１），但二者的面积无显著性差异（Ｐ＞０．０５）。结论　绝经后妇女随着年龄的增加脊椎压缩性骨折的患病率逐渐升高。胸椎压缩性骨折的患病率高于腰椎，主要发生于Ｔ６、Ｔ７、Ｔ１１、Ｔ１２。

目的了解老年腰椎及髋部骨密度(bmd)变化,并探讨不同部们测量点结果对骨质疏松症诊断的影响。方法用法国产Lcxxos型双能x线骨密度测量仪(DEXA)对辽宁地区45-89岁老人腰椎正位及髋部骨密度测定,按年龄分组进行统计分析。结果腰椎正位及髋部本组两性问bmd值差异有显著性(Pbmd值保持稳定,并稍有升高趋势。各组腰椎bmd值明显高于髋部bmd值,差异有显著性(Pbmd值诊断敏感性明显低于髋部bmd值。结论在诊断骨质疏松症中髋部bmd值较腰椎正位bmd值更精确,且腰椎bmd值受影响因素较多,腰椎骨质增生、腰椎骨折畸形等。轻者应对异常高密区加以删除后分析,重者应避免应用压缩骨折明显或侧弯,前后凸明显,较重的腰椎增生性骨关节的腰椎前后位bmd值进行临床分析。这样才能保证诊断更准确。
