Dear Editor,We read with great interest the recent article by Tapper et al.[1].The authors aimed to determine the clinical predictors of advanced histology in the referral *** multivariate logistic regression analyses,they have demonstrated that body mass index(BMI)30 kg/m2,female gender,and aspartate aminotransferase(AST)>40 IU/L were associated with a non-alcoholic fatty liver disease(NAFLD)activity score>*** have also shown that ASTto-platelet ratio index(APRI)>1 was the most significant predictor of advanced ***,they have suggested that patients with suspected NAFLD should be routinely evaluated for advanced liver disease,including non-invasive indices of fibrosis such as APRI,and serious consideration given to liver *** the value of non-invasive indices of fibrosis such as APRI in patients with NAFLD,we consider this large,retrospective study to be very important;however,we have some questions that should be mentioned as contributory factors.

AIM:To determine the upper cut-off values of serumalanine aminotransferase(ALT)and aspartate aminotransferase(AST)in a Northern Chinese ***:A total of 3769 subjects in Jilin Province Northeast China were stratified to determine the potential factors affecting serum ALT and AST *** upper cut-off values of serum ALT and AST in these subjects were determined using receiver operating characteristic analysis and their sensitivity and specificity were ***:Stratification analysis revealed that serum ALT and AST levels were associated with gender,alcohol consumption,serum cholesterol and triglyceride levels,and body mass *** upper cut-off values of serum ALT and AST were 22.15 U/L and 25.35 U/L for healthy men and 22.40 U/L and 24.25 U/L for healthy women,*** new cut-off values had a higher sensitivity,but a slightly lower specificity than the current ***:Our results indicate that the new upper cut-off values of serum ALT and AST are markedly lower than current standards and may be valuable for the evaluation of liver function.

Validation of aspartate aminotransferase to platelet ratiofor diagnosis of liver fibrosis and prediction of postoperativeprognosis in infants with biliary atresia pathological Metavir fibrosis score of the liver wedgespecimens of 91 BA infants. The prognostic value ofpreoperative APRI for jaundice persistence, liver injury,and occurrence of cholangitis within 6 mo after KP wasstudied based on the follow-up data of 48 BA ***： APRI was significantly correlated withMetavir scores （rs = 0.433; P 〈 0.05）. The mean APRIvalue was 0.76 in no/mild fibrosis group （Metavir scoreF0-F1）, 1.29 in significant fibrosis group （F2-F3）, and2.51 in cirrhosis group （F4）（P 〈 0.001）. The areaunder the ROC curve （AUC） of APRI for diagnosingsignificant fibrosis and cirrhosis was 0.75 （P 〈 0.001）and 0.81 （P = 0.001）, respectively. The APRI cut-offof 0.95 was 60.6% sensitive and 76.0% specific forsignificant fibrosis diagnosis, and a threshold of 1.66was 70.6% sensitive and 82.7% specific for *** preoperative APRI in infants who maintainedjaundice around 6 mo after KP was higher than thatin those who did not （1.86 ± 2.13 vs 0.87 ± 0.48, P 〈0.05）. The AUC of APRI for prediction of postoperativejaundice occurrence was 0.67. A cut-off value of0.60 showed a sensitivity of 66.7% and a specificityof 83.3% for the prediction of jaundice *** APRI had no significant association withlater liver injury or occurrence of ***： Our study demonstrated that APRIcould diagnose significant liver fibrosis, especiallycirrhosis in BA infants, and the elevated preoperativeAPRI predicts jaundice persistence after KP.

Excitatory amino acid transporters(EAATs) are responsible for excitatory amino acid transportation and are associated with auto-immune diseases in the central nervous system and peripheral ***, the subcellular location and function of EAAT2 in macrophages are still obscure. In this study,we demonstrated that LPS stimulation increases expression of EAAT2(coded by Slc1a2) via NF-κB signaling. EAAT2 is necessary for inflammatory macrophage polarization through sustaining mTORC1 activation. Mechanistically, lysosomal EAAT2 mediates lysosomal glutamate and aspartate efflux to maintain V-ATPase activation, which sustains macropinocytosis and mTORC1. We also found that mice with myeloid depletion of Slc1a2 show alleviated inflammatory responses in LPS-induced systemic inflammation and high-fat diet induced obesity. Notably, patients with type Ⅱ diabetes(T2D) have a higher level of expression of lysosomal EAAT2 and activation of mTORC1 in blood macrophages. Taken together, our study links the subcellular location of amino acid transporters with the fate decision of immune cells,which provides potential therapeutic targets for the treatment of inflammatory diseases.

The title complex Ni(C4H5NO4)(C7H6N2)31.75(H2O) (C25H26.5N7NiO5.75, Mr = 575.74) has been prepared and its crystal structure was determined by X-ray diffraction method. The crystal of this chiral complex belongs to monoclinic, space group P21 with a = 13.208(1), b = 10.741(1), c = 19.398(1) ? b = 104.39(1)o, V = 2665.8(4) 3, Z = 4, Dc = 1.435 g/cm3, m(MoKa) = 0.780 mm-1 and F(000) = 1198. The final R = 0.0504 and wR = 0.1267 for 9159 observed reflections (I > 2s(I)), and R = 0.065 and wR = 0.1299 for 11006 independent reflections. The complex assumes a distorted octa- hedral coordinate geometry formed by one aspartate dianion and three benzimidazole ligands. The aspartate chelates to the Ni atom as a tridentate ligand. The lattice water molecules are hydrogen- bonded to the Ni complex molecule.

Glutamic acid(Glu)and aspartic acid(Asp)are acidic amino acids with regulatory roles in nutrition,energy metabolism,and oxidative *** study aimed to evaluate the effects of low-protein diets supplemented with Glu and Asp on the intestinal barrier function and energy metabolism in weaned piglets challenged with hydrogen peroxide(H2O2).Forty piglets were randomly divided into 5 groups:NC,PC,PGA,PG,and PA(n=8 for each group).Pigs in the NC and PC groups were fed a low-protein diet,while pigs in the PGA,PG,or PA groups were fed the low-protein diet supplemented with 2.0%Glu+1.0%Asp,2.0%Glu,or 1.0%Asp,*** day 8 and 11,pigs in the NC group were intraperitoneally injected with saline(1 mL/kg BW),while pigs in the other groups were intraperitoneally administered 10%H2O2(1 mL/kg BW).On day 14,all pigs were sacrificed to collect jejunum and ileum following the blood sample collection in the ***,low-protein diets supplemented with Glu or Asp ameliorated the intestinal oxidative stress response in H2O2-challenged piglets by decreasing intestinal expression of genes(P<0.05)(e.g.,manganese superoxide dismutase[MnSOD],glutathione peroxidase[Gpx]-1,and Gpx-4)encoding oxidative stress-associated proteins,reducing the serum concentration of diamine oxidase(P<0.05),and inhibiting apoptosis of the intestinal *** and Asp supple-mentation attenuated the upregulated expression of energy metabolism-associated genes(such as hexokinase and carnitine palmitoyltransferase-1)and the H2O2-induced activation of acetyl-coenzyme A carboxylase(ACC)in the jejunum and adenosine monophosphate-activated protein kinase-acetyl-ACC signaling in the *** Glu and Asp also ameliorated intestinal barrier damage as indicated by restored intestinal histology and *** conclusion,low-protein diets supplemented with Glu and Asp protected against oxidative stress-induced intestinal dysfunction in piglets,suggesting that this approach could be used as a nut

Abnormal postoperative neurobehavioral performance(APNP)is a common phenomenon in the early postoperative *** disturbed homeostatic status of metabolites in the brain after anesthesia and surgery might make a significant contribution to *** dynamic changes of metabolites in different brain regions after anesthesia and surgery,as well as their potential association with APNP are still not well ***,we used a battery of behavioral tests to assess the effects of laparotomy under isoflurane anesthesia in aged mice,and investigated the metabolites in 12 different sub-regions of the brain at different time points using proton nuclear magnetic resonance('H-NMR)*** abnormal neurobehavioral performance occurred at 6 h and/or 9 h,and recovered at 24 h after anesthesia/*** with the control group,the altered metabolite of the model group at 6 h was aspartate(Asp),and the difference was mainly displayed in the cortex;while significant changes at 9 h occurred predominantly in the cortex and hippocampus,and the corresponding metabolites were Asp and glutamate(Glu).All changes returned to baseline at 24 *** altered metabolic changes could have occurred as a result of the acute APNP,and the metabolites Asp and Glu in the cortex and hippocampus could provide preliminary evidence for understanding the APNP process.
