Objective adipose-derived stem cells(ADSCs) are suggested to possess a highly plastic ability to differentiate into several specific cell types in addition to adipocyte lineages, including germ layer tissue-specific cell lineages such as chondrocyte, myocyte, neuronal, and osteoblast lineages. The aim of this study is to establish an in vitro culture technique for ADSCs in an adult guinea pig model that facilitate their differentiation into hair cell-like cells. Materials and Methods cells from inguinal fat pads in adult guinea pigs were cultured with β-mercaptoethanol, RA, Forskorin, Heregulin, bFGF, BDNF and EGF. cellular differentiation was examined using immunocytochemistry techniques. Results The ADSCs demonstrated hair cell immunophenotypes with expression of epitopes of the hair cell marker protein myosin Ⅶa. Conclusion ADSCs from adult guinea pig adipose tissue can differentiate into hair cell-like cells when cultured in vitro. ADSCs may serve as seed cells for tissue engineering.

The slow regenerating rate and misdirected axonal growth are primary concerns that disturb the curative outcome of peripheral nerve repair.Biophysical intervention through nerve scaffolds can provide efficient,tunable and sustainable guidance for nerve regrowth.Herein,we fabricate the reduced graphene oxide(rGO)/polycaprolactone(PCL)scaffold characterized with anisotropic microfibers and oriented nanogrooves by electrospinning technique.adipose-derived stem cells(ADSCs)are seeded on the scaffolds in vitro and the viability,neural differentiation efficiency and neurotrophic potential are investigated.RGO/PCL conduits reprogram the phenotype of seeded cells and efficiently repair 15 mm sciatic nerve defect in rats.In summary,biophysical cues on nerve scaffolds are key determinants to stem cell phenotype,and ADSC-seeded rGO/PCL oriented scaffolds are promising,controllable and sustainable approaches to enable peripheral nerve regeneration.

The high incidence and disability rates of stroke pose a heavy burden on society.Inflammation is a significant pathological reaction that occurs after an ischemic stroke.Currently,therapeutic methods,except for intravenous thrombolysis and vascular thrombectomy,have limited time windows.Mesenchymal stem cells(MSCs)can migrate,differentiate,and inhibit inflammatory immune responses.Exosomes(Exos),which are secretory vesicles,have the characteristics of the cells from which they are derived,making them attractive targets for research in recent years.MSC-derived exosomes can attenuate the inflammatory response caused by cerebral stroke by modulating damage-associated molecular patterns.In this review,research on the inflammatory response mechanisms associated with Exos therapy after an ischemic injury is discussed to provide a new approach to clinical treatment.

Osteoporosis(OP)is a noncommunicable bone disease caused by a shift in the balance between os-teoblasts and osteoclasts,and can severely affect the health of elderly persons.Autologous stem-cell transplantation can improve reduced bone density and weakened fracture healing abilities in patients with OP.However,OP can adversely affect the osteogenesis and proliferation abilities of autologous adipose-derived stem cells(ASCs).Therefore,an effective drug is required to facilitate autologous ASCs to recover their osteogenic and proliferative potential.Tetrahedral framework nucleic acid(tFNA)is a new type of nanomaterial that has ability to regulate the biological behavior of cells effectively and en-hance the bioactivity of stem cells.In this study,we examine the effects of tFNAs on the osteogenic differentiation and proliferation abilities of ASCs in rats with OP.The results indicate that the 250 nmol/L tFNAs can considerably increase the expression of osteogenesis-related markers,effectively promote the proliferation and osteogenic differentiation of osteoporotic ASCs(OP-ASCs),and help them to regain their osteogenic and proliferative potential.In short,tFNAs can enable OP-ACSs to recover their osteogenic po-tential and promote their proliferation and,therefore,can play a key regulatory role in autologous ASC transplantation.

Adult mesenchymal stem cells, specifically adipose-derived stem cells have self-renewal and multiple differentiation potentials and have shown to be the ideal candidate for therapeutic applications in regenerative medicine, particularly in peripheral nerve regeneration. adipose-de- rived stem cells are easily harvested, although they may show the effects of aging, hence their potential in nerve repair may be limited by cellular senescence or donor age. cellular senescence is a complex process whereby stem cells grow old as consequence of intrinsic events （e.g., DNA damage） or environmental cues （e.g., stressful stimuli or diseases）, which determine a permanent growth arrest. Several mechanisms are implicated in stem cell senescence, although no one is exclusive of the others. In this review we report some of the most important factors modulating the senescence process, which can influence adipose-derived stem cell morphology and function, and compromise their clinical application for peripheral nerve regenerative cell therapy.

To investigate the effects of allogeneically transplanted, adipose-derived stem cells in aging rats, in the present study, we established a rat model of subacute aging using continuous subcutaneous injections of D-galactose. Two weeks after the adipose-derived stem cells transplantations, serum superoxide dismutase activity was significantly increased, malondialdehyde content was significantly reduced, hJppocampal neuronal degeneration was ameliorated, the apoptotic index of hippocampal neurons was decreased, and leaming and memory function was significantly improved in the aging rats. These results indicate that allogeneic transplantation of adipose-derived stem cells may effectively delay D-galactose-induced aging.

Objective:This study aimed to investigate the differentiation of human adipose-derived stem cells(hASCs)into endometrial epithelial cells(EECs)under certain induction conditions and to a further step provide a promising approach for ASCs in clinical practice to the treatment of severe intrauterine adhesion.Methods:Four groups of hASCs were separately cultured as follows:in Group 1,hASCs were cultured in a control medium(5%fetal bovine serum[FBS]+α-minimum Eagle’s medium[α-MEM]);in Group 2,hASCs were cultured in an induction medium(5%FBS+α-MEM+[1×10-7 mol/L 17β-estradiol]+10 ng/mL transforming growth factorβ1[TGF-β1]+10 ng/mL epidermal growth factor[EGF]+10 ng/mL platelet-derived growth factor BB[PDGF-BB]);in Group 3,hASCs and human endometrium cells(hEMCs)were cocultured in the control medium;and in Group 4,hASCs and hEMCs were cocultured in the induction medium.Results:When cocultured with hEMCs,the morphology of hASCs became similar with EECs,and the addition of factors such as EGF,TGFβ,PDGF-BB,and 17β-estradiol promoted differentiation.This study,for the first time,demonstrated estrogen receptor(ER)αand ERβexpression in hASCs and preliminarily explored changes in ERα,ERβ,β-catenin,and H19 mRNA expression during hASC differentiation.Furthermore,we concluded that H19 mRNA expression was negatively correlated with differentiation,which is seemingly related to the estrogen signaling pathway.Conclusions:hASCs revealed the potential for differentiating to EECs when cocultured with hEMCs.

Fibrosis is a condition in which connective tissue replaces normal parenchymal tissue,resulting in significant tissue remodeling.Fibrosis can affect several organs and pose a serious threat to human health and life.adiposederived stem cells(ASCs)have been suggested as promising candidates for antifibrotic therapies.Paracrine secretion is one of the key processes in stem cell therapy due to its critical function in cellular communication.ASC-derived exosomes(ASC-exos)are used as tools for restoring and regenerating damaged tissue,and they are now thought to orchestrate antifibrosis-related events.In this review,we summarize the recent findings and present an extensive view of the therapeutic applications of ASC-exos in fibrotic diseases.

Objective To investigate the effects of combination of PRP,PRF and adipose-derived stem cells(ADSC)for wound healing of chronic skin ulcer,and provide evidences for clinical application.Methods Twenty-four SD rats aged 6-8 weeks were used in this study.5 grams of groin adipose tissue were derived from each rat.After collagenase digestion,mesenchyma stem cells derived from adipose tissue(ADSCs)were determined by immunocytochemical method and flow cytometry.After continuous culturing,the 3th passage of ADSCs in good condition was prepared for transplantation.Ten milliliters of whole blood were extracted from one rat for making PRP and PRF backup.Full thickness skin defect model with six wounds with an area of 1.5cm*1.5cm on each side of the spine on the back was established in rats after anesthesia.Then the rats were randomly divided into six groups.Skin wounds were injected with PRF combining ADSC in group A,PRF in group B,ADSC in group C,ADSC combining PRP in group D,PRP in group E,and same amount of normal saline(NS)in group F.Histological analysis was performed to evaluate the velocity and the quality of the wound healing 7 days,14 days,21days and 28 days after treatment,and comparing the regeneration of epidermis,corium,fibroblast,blood vessel and skin appendages respectively.Results The woulds in each group healed well,without symptoms of infection.Skin wounds were almost healed in 21 days,and no significant difference in healing velocity was found among these groups.There were no apparent inflammatory cells in 7 days.The wounds were filled with granulation formed by proliferation of connective tissue with visible new blood capillary and repair cells like fibroblast.Epidermis grew out and became thick,and gradually grew to the wounds,with active proliferation of re-epithelization.Re-epithelization was completed in 14 days,with granulation tissue covered by epidermis and fibrosis showing a significant increase in collagenous fiber,and visible skin appendages like new hair follicle,glandula sebacea,sweat gland
