Antioxidant capacity and genoprotective effect of ethanol fruit extract from Detarium microcarpum Guill. and Perr.(Caesalpiniaceae)

Objective: To evidence the ability of ethanol fruit extract from Detarium microcarpum(D. microcarpum) to preserve DNA integrity against oxidative genomic ***: Ethanol extract from D. microcarpum fruit pulp was analyzed for its antioxidant capacity using ferric reducing antioxidant power, 2,2-diphenyl-1-picrylhydrazyl,2,20-azinobis-3-ethyl-ethylbenzothiazoline-6-sulphonate, superoxide anion, deoxyribose degradation and lipid peroxidation models. The genoprotective activity was assessed ex vivo by comet assay, on liver cells of NMRI female mice using cyclophosphamide(CP) as genotoxic ***: Ethanol extract from D. microcarpum fruit pulp exhibited interesting antioxidant activity in 2,2-diphenyl-1-picrylhydrazyl, deoxyribose degradation and lipid peroxidation assays. The extract did not present any genotoxic effect but protected DNA against CP-induced damages with a dose-dependent manner. The genoprotective effect observed was related to the antioxidant molecules of the fruit that scavenged the hydroxyl radical(generated by the metabolism of CP) as well as the peroxyl and alkoxyl radicals issued from lipid peroxidation. Other mechanisms such as inactivation of CP metabolism to genotoxic end products, induction of the expression of antioxidant and DNA repair enzymes have been ***: Our results suggest that the wild edible fruit from D. microcarpum could be beneficial on consumer's health by its antioxidant and genoprotective effects, particularly during chemotherapies exhibiting genotoxic effects like CP in cancer treatment.

A potential strategy for patients with estrogen receptor(ER)-positive breast cancer is necessary to replace neoadjuvant chemotherapy which has limited *** endocrine therapy(NAE)has been indicated to be a favorable alternate approach to downstage large or locally advanced breast cancer in ER-positive,human epidermal growth factor receptor 2(HER2)-negative(ER+/HER2-)patients,especially postmenopausal *** studies have demonstrated the efficacy of various endocrine agents in *** inhibitors(AIs)have proven superiority over tamoxifen as a suitable choice to optimize treatment *** was recently reported as an effective agent,similar to ***,the addition of targeted agents exerts synergistic antiproliferative effects with endocrine agents and rapidly improves response rates in both endocrine sensitive and resistant *** neoadjuvant platform provides a unique opportunity to define the appropriate strategy and address the mechanisms of endocrine *** addition,the predictive value of biomarkers and genomic assays in NAE is under investigation to evaluate individual effects and validate biomarker-based *** this review,we discuss the most relevant evidence on the potential of NAE for ER+breast *** current understanding also offers new insights into the identification of the optimal settings and valuable predictive tools of NAE to guide clinical treatment decisions and achieve beneficial therapeutic effects.

4-(2-QUINOLYLAZO)-1, 3-DIHYDROXY-NAPHTHALENE——A NEW SENSITIVE ANALYTICAL REAGENT FOR TRACE COPPER DETERMINATION

The azo compound 4-(2-quinolylazo)-1,3-dihydroxynaphthalene (2-QADNm) has been synthesized and used as a new sensitive spectrophotometric and electroanalytical reagent for determination of trace amounts of copper. In dilute alkaline solutions, 2-QADNm shows a single-sweep polarographic wave at -0.95 V (vs. SCE). In the presence of copper(Ⅱ) two new waves appear at -0.52 and -0.79 V. Using the wave at-0.79 V, a trace amount of copper down to 10^(-8) mol/L can be determined. The electroanalytical characteristics of 2-QADNm and the electrochemical reaction mechanism of the copper(Ⅱ)-2-QADNm complex have been studied in detail by the use of various electrochemical techniques. In alkaline solutions, 2-QADNm forms red complex with copper(Ⅱ) which has an absorption peak at 526 nm, the molar absorptivity being 6.4×10~4 1·mol^(-1)·cm^(-1). In the presence of a non-ionic surfactant the molar absorptivity of this complex is 1.6×10~5 1·mol^(-1)·cm^(-1).

Trichothecene mycotoxin T-2 is a secondary metabolite of several Fusarium species, and is known as a natural contaminant of stored grain and other agricultural products. This compound is a potent inhibitor of protein synthesis and has been implicated as the causative agent of several mycotoxicoses. There is also a close relationship between T-2 toxin and esophageal carcinoma, Keshan, Kaschin-Beck diseases in China.

Ameliorative effect of grapefruit juice on amiodarone-induced cytogenetic and testicular damage in albino rats

Objective:To evaluate the ameliorative role of grapefruit juice on the cytogenetic and testicular damage induced by the antiarrythmic drug amiodarone in albino ***:Animals were divided into four *** I was considered as *** II was given grapefruit juice at a dose level of 27 mL/kg body *** III was orally administered amiodarone(18 mg/kg body weight)daily for 5 *** were sacrificed after 5 weeks of *** marrow was collected from the femurs for analysis of chromosomal aberrations and mitotic *** were removed and stained with H&E for histological *** were collected from epidedymis for detection of sperm head *** assay was used to detect DNA ***:Amiodarone treatment caused a significant increase in the percentage of chromosomal aberrations,decreased the mitotic index and increased DNA *** testis showed many histopathological alterations,inhibition of spermatogenesis and morphometric *** number of sperm head abnormalities was *** animals with amiodarone and grapefruit juice caused a reduction in chromosomal aberrations,mitotic index,DNA damage and testicular alterations caused by ***:The results of this study indicated that grapefruit juice ameliorates the cytotoxicty and testicular alterations induced by amiodarone in albino rats and this is may be due to the potent antioxidant effects of its components.

Pedalium murex Linn(Pedaliaceae) fruits:a comparative antioxidant activity of its different fractions

Objective:To examine the antioxidant activity and total phenolic content of different solvent fractions of Pedulium murex(***)Linn fruits(Family:Pedaliaceae)as well as the correlation between the total antioxidant capacity and total phenolic ***:In the present study,the antioxidant activities of *** were evaluated using six in-vitro assays,namely total antioxidant assay,DPPH assay,reducing power,nitric oxide scavenging,hydrogen peroxide scavenging and deoxyribose scavenging assays,and total phenol contents were also ***:The ethyl acetate(EA)fraction was found to have high levels of phenolic content(298.72±2.09 mg GAE/g).The EA fraction exhibit higher total antioxidant capacity,higher percentage of DPPH radical scavenging activity(135.11±2.95μg/mL),nitric oxide(200.57±4.5lμg/mL),hydrogen peroxide(2I7.91±6.12μg/mL),deoxyribose(250.01±4.68μg/mL)and higher reducing *** coefficient(r^2=0.914)was found to be significant between total phenolic content and total antioxidant ***:In general,the results indicate that the EA fractions are rich in phenolic antioxidants with potent free radical scavenging activity implying their importance to human health.

Isolation of a small molecule with anti-MRSA activity from a mangrove symbiont Streptomyces ***-1 and its biomedical studies in Zebrafish embryos

Objective:The aim of the present study was to isolate the anti-MRSA(Methicillin Resistant Staphylococcus aureus)molecule from the Mangrove symbiont Streptomyces and its biomedical studies in Zebrafish ***:MRSA was isolated from the pus samples of Colachal hospitals and confirmed by amplification of mecA ***-MRSA molecule producing strain was identified by!6s rRNA gene ***-MRSA compound production was optimized by Solid State Fermentation(SSF)and the purification of the active molecule was carried out by TLC and *** inhibitory concentration and LC_(50)were calculated using Statistical software *** Biomedical studies including the cardiac assay and organ toxicity assessment were carried out in ***:The bioactive anti-MRSA small molecule A,was purified by TLC with Rf value of 0.37 with 1.389 retention time at *** Inhibitory Concentration of the purified molecule A_2 was 30μg/mL but,the inhibitory concentration of the MRSA in the infected embryo was 32-34μg/mL for TLC purified molecule A,with LC_(50)mean value was61.504μg/*** toxicity was assessed in 48-60μg/mL by observing the physiological deformities and the heart beat rates(HBR)of embryos for anti MRSA molecule showed the mean of 41.33-41.67 HBR/15 seconds for 40μg/mL and control was 42.33-42.67 for 15 seconds which significantly showed that the anti-MRSA molecule A_2 did not affected the ***:Anti-MRSA molecule from Streptomyces sp PVRK-I was isolated and biomedical studies in Zebrafish model assessed that the molecule was non toxic at the minimal inhibitory concentration of MRSA.
