Recently, immunoglobulins （Igs） were unexpectedly found to be expressed in epithelial cancers. Immunoglobulin class switching or class switch recombination （CSR） is a natural biological process that alters a B cell＇s production of antibodies （immunoglobulins） from one class to another. However, the mechanism of CSR of Iggenes in cancer is still unknown. Here, we confirmed by detecting the hallmark of CSR that the Iga gene in cancer underwent CSR. Then we focused on activation-induced cytidine deaminase （aid）, a crucial factor for initiating CSR. Further studies using tumor necrosis factor （TNF）-α stimulation and specific inhibitor of NF-KB revealed that TNF-α could increase aid expression through NF-κB signaling. Finally, we demonstrated that aid could co-localize with protein kinase A and bind to the switching （Sα） region of the Igα gene. Overexpression of aid obviously enhanced Igα heavy chain expression and its binding ability to the Sa region. These findings indicated that TNF-α-induced aid expression is involved with CSR in cancer.

China’s Foreign aid,Recipient Countries’Economic Growth,and the Mediating Effect of aid in Infrastructure

Based on the panel data of China’s foreign aid in 2003-2014 from aidData,this paper empirically investigates the relationship between China’s foreign aid and economic growth in recipient *** the principal component analysis(PCA),the authors propose a variable,*** infrastructure index,to measure the effects of aid on the economic growth of recipient *** study shows that aid from China has significantly improved the level of economic growth in recipient countries,which provides a new evidence of China’s aid *** analysis of the ways by which aid in infrastructure induces economic growth,the authors conclude that the improvement of local infrastructure accounts for 55.30%of the aggregate economic growth effect of China’s aid to recipient countries.

Iranian teacher’s knowledge about first aid in the school environment

Purpose: School students are at risk of unintentional injuries. Teachers are the main caregivers and the first protectors for school students and must be able to deliver first aid. This study aimed to assess the school t*eachers knowledge about first aid in Kashan, Iran. Methods: A cross-sectional study was undertaken on a sample of 200 teachers working in public and private primary, sec on dary and high schools in Kashan city, Iran. Data collection was achieved via a questionnaire consisting of items on teachers' characteristics and 20 questions on basic first-aid practices. One point was given for each correct answe匚 The lowest and highest possible scores were 0 and 20. Then, the sum scores 0-9, 10—16, and 17—20 were ranked as weak, moderate, and good first aid knowledge, respectively. Statistical analyses were performed using descriptive statistics. Chi-square and Fisher's exact tests, and regression analysis. Results: The mean age and working experienee of teachers were respectively (41.37 ± 6.35) years and (20.07 ± 7.29) years. Among all teachers 51.0% were males and 66.3% were working in public schools. Totally, 40.3% of the teachers had passed courses on first aid and 48.5% had confronted w让h situations needing first-aid;59.7% evaluated their own knowledge of first aid as moderate. The mean score of firstaid knowledge was 10.00 ± 2.58. Teachers who had received first aid training and those with previous experiences of situations requiring first-aid possessed higher scores (p aid in a child with a fracture (93.4%) or struck by electricity (85.2%). Conclusion: Most of the teachers had insufficient knowledge on first aid. They mostly did not pass any training on first aid. Urgent action seems necessary for training teachers on first aid through in-service training programs.

目的:本课题组前期利用基因芯片技术对DLBCL石蜡标本及作为对照的反应性增生淋巴结石蜡标本进行miRNA表达谱分析,旨在寻找差异表达的miRNA。本研究第一部分将筛选出在DLBCL中差异表达明显的miRNAs,然后进行扩大样本再验证。筛选发现miR-106a-5p在DLBCL石蜡标本中呈现一致性的高表达,后进一步探讨miR-106a-5p在DLBCL中的表达与患者临床预后指标的相关性及其预后价值。另外利用细胞转染技术在细胞水平探讨miR-106a-5p在弥漫大B细胞瘤的发生发展过程中的生物学功能。本研究第二部利用GEPIA网站预测aid在DLBCL中的表达水平,并应用组织芯片技术和免疫组化染色验证其在DLBCL组织中的蛋白表达水平,结合临床信息,进一步探讨aid表达的临床意义及预后价值。方法:一、基于基因芯片筛选的miR-106a-5p在DLBCL中的表达及其临床意义收集2016年到2019年期间105例弥漫性大B细胞淋巴瘤患者的石蜡样本,另外选择10例反应性增生淋巴结石蜡标本作为对照组,提取标本中的总RNA。通过基因芯片火山图(差异倍数变化Flod change上调大于等于1.5或下调小于0.5且P值aid并验证其在DLBCL中的表达及临床意义应用GEPIA网站观测aid在DLBCL和正常对照中的表达,对石蜡组织芯片进行免疫组化染色从蛋白水平分析aid在DLBCL中的表达,以组织化学评分(H-score)对aid进行半定量分析。定期随访病人,Kaplan-Meier法分析DLBCL中aid的表达对患者总生存期的影响。利用生物信息学网站Star Base共表达分析观测两者表达的相关性,并在组织中进行验证分析,以期为后续具体的分子机制研究提供依据。结果:1.共筛选出13个miRNAs进行样本扩大再验证,其中6个miRNAs显著上调,与基因芯片结果一致,另外7个miRNAs在芯片结果中显示下调,却在样本扩大验证中呈现上调。以扩大验证结果与基因芯片差异表达谱结果呈现一致性上调的miR-106a-5p作为研究指标进一步探讨。研究显示患者miR-106a-5p的高表达与临床病理指标均无相关性,但Kaplan-Meier生存分析显示,低表达miR-106a-5p的患者的总生存期优于高表达miR-106a-5p患者,且差异具有统计学意义(P=0.0038)。***-106a-5p在DLBCL细胞如SU-DHL-4,SU-DHL-8,OCI-LY3,OCI-LY19,WSU-NHL,RC中均为表达上调。瞬时转染miR-106a-5p inhibitor使SU-DHL-4,OCI-LY3细胞内miR-106a-5p水平有效降低。转染后发现DLBCL细胞增殖活性降低,凋亡增加,细胞周期阻滞于G/G期。***网站显示,相较于正常对照组,aid在DLBCL中呈现显著高表达。基于组织芯片进行的免疫组化染色结果显示DLBCL患者肿瘤组织中aid表达具有明显的异质性,non-GCB亚型患者的H-score分数显著高于GCB亚型的患者,且差异具有统计学意义(P=0.046)。以H-score为10作为界值,发现H-score≥10分的患者生存显著差于H-scoreaid在DLBCL中的表达呈现正相关,但组织验证中两者表达并无显著相关性。结论:基因芯片差异表达谱与q PCR验证均显示DLBCL存在异常表达miRNAs。miR-106a-5p在DLBCL中表达水平上调,且高表达miR-106a-5p患者显示更差的预后,可用来预测患者生存。此外,DLBCL细胞中干扰miR-106a-5p水平可以明显抑制细胞的增殖并

近年来,全球范围内甲状腺乳头状癌(papillary thyroid carcinoma, PTC)的发病率递增,已成为头颈外科最常见的恶性肿瘤之一。国内外学者对PTC的机制研究成为临床和基础研究的热点,其发生、发展过程涉及多种遗传和表观遗传学改变,其中B型丝氨酸/苏氨酸蛋白激酶(B typeserine/threonine protein kinases, BRAF)密码子600处谷氨酸通常替换缬氨酸(BRAFV600E)在PTC中发生突变频率颇高;活性诱导性胞嘧啶脱氨基酶(activation-induced cytidine deaminase, aid)的累积异常表达可引起癌基因或抑制基因突变,aid高表达可能与BRAFV600E突变型PTC具有交互作用,要通过aid基因水平改变来影响细胞的分化程度、调控肿瘤的生长和转移概率,还需要进一步的研究。本文拟针对PTC与BRAFV600E、ADI之间的关联性作一综述。

First aid Knowledge and Attitude of Secondary School Students in Saudi Arabia

Background: Increasing the number of qualified people, including school students who can play a significant role in first-aid (FA) and basic life support (BLS) arena is a public health demand. Aim: To assess the awareness of Saudi secondary school students regarding FA and BLS. Methodology: 360 male secondary school children in Taif, Saudi Arabia were surveyed. Results: The mean students’ age was 17.4 ± 1.21 years. The mean participants’ FA knowledge score was 64.8% ± 11%. Generally, trained students reported both better FA knowledge and skills than untrained counter-parts;for 79.6% trained-compared to 53.7% untrained-students recorded such score > 70% [χ2 (df 1) = 11.60, p 70%) [62.3% 2(df 1) = 5.90, p = 0.02] was. Trained students better deal with critical cases, bleeding and bodily injury compared to untrained peers [89.8% vs. 55.9%, χ2 (df 1) = 20.3, p 2 (df 1) = 11.62, p = 0.04;81.6% vs. 67.2%, χ2 (df 1) = 4.13, p = 0.04, respectively]. Only 37.2% of schools had FA incorporated in the education curriculum. Eventually, FA training and the presence of FA group were significant predictors for improved FA knowledge among students [odds ratio (OR) 3.35, 95% CI 1.60 - 7.06;OR 2.28, 95% CI 1.34 - 3.95, respectively). Conclusions: First-aid training is crucial to enhance school children’s FA skills and to thrust their contribution to health emergency control efforts. Incorporating FA training within the educational curriculum of Saudi schools is a step forward to disseminate FA message in the community.

目的探讨幽门螺杆菌(Hp)侵袭内化对细胞aid表达的影响及相关机制。方法选择不同侵袭力的Hp,分别感染AGS细胞,以抗β1整合素抗体抑制侵袭为对照组,经庆大霉素保护性侵袭实验,用real-time RT-PCR和细胞免疫荧光分别检测aid mRNA和蛋白表达水平。用SN-50抑制AGS细胞NF-κB活性,同法检测Hp对aid表达的影响。结果高侵袭力的Hp诱导的aid蛋白和mRNA的表达量均高于低侵袭力的Hp,Paid蛋白表达均减少(Paid蛋白和mRNA表达水平均降低,Paid的异常表达,这种诱导涉及NF-κB信号通路,细菌的诱导能力与内化细菌的数量呈正相关。Hp内化诱导宿主细胞异常表达aid可能是导致Hp相关肿瘤发生的机制之一。
