AIM To observe the effect of acupuncture and moxibustion on the expression of IL 1β and IL 6 mRNA in ulcerative colitis rats. METHODS The SD rat ulcerative colitis model was created by immunological method associated with local stimulation. Colonic mucosa was prepared from human fresh surgical colonic specimens, homogenized by adding appropriate amount of normal saline and centrifuged at 3000*!r/*!min . The supernatant was collected for measurement of protein conentration and then mixed with Freund adjuvant. This antigen fluid was first injected into the plantae of the model group rats, and then into their plantae, dorsa, inguina and abdominal cavities (no Freund adjuvant for the last injection) again on the 10th, 17th, 24th and 31st day. When a certain titer of serum anti colonic antibody was reached, 2% formalin and antigen fluid (no Freund adjuvant) were administered separately by enema. The ulcerative colitis rat model was thus set up. The animals were randomly divided into four groups: model control group (MC, n =8), electro acupuncture group (EA, n =8), herbs partition moxibustion group (HPM 8), normal control group (NC, n =8). HPM: Moxa cones made of refined mugwort floss were placed on the medicinal pad (medicinal pad dispensing: Radix Aconiti praeparata, cortex Cinnamomi, etc ) for Qihai (RN 6) and Tianshu (ST 25, bilateral) and ignited. Two moxa cones were used for each acupoint once a day and 14 times in all. EA: Tianshu (bilateral) and Qihai were stimulated by the intermittent pulse with 2Hz frequency, 4mA intensity for 20 minutes once a day and 14 times in all. After treatment, rats of all four groups were killed simultaneously. The spleen was separated and the distal colon was dissected. Total tissue RNA was isolated by the guanidinium thiocyanate phenol chloroform extraction method. RT PCR technique was used to study the expression of IL 1β and IL 6 mRNA. Up-regulation of P2X7 Receptors Contributes to Spinal Microglial Activation and the Development of Pain Induced by BmK-I

Previous work has demonstrated that the sensitization of spinal neurons and microglia is important in the development of pain behaviors induced by BmK I,a Na^+ channel activator and a major peptide component of the venom of the scorpion Buthus martensi Karsch(BmK).We found that the expression of P2X7 receptors(P2X7Rs)was up-regulated in the ipsilateral spinal dorsal horn after BmK I injection in rats.P2X7R was selectively localized in microglia but not astrocytes or neurons.Similarly,interleukin 1β(IL-1β)was selectively up-regulated in microglia in the spinal dorsal horn after BmK I injection.Intrathecal injection of P2X7R antagonists largely reduced BmK I-induced spontaneous and evoked pain behaviors,and the up-regulation of P2X7R and IL-1β in the spinal cord.These data suggested that the up-regulation of P2X7Rs mediates microglial activation in the spinal dorsal horn,and therefore contributes to the development of BmK I-induced pain.

Role of NLRP3 inflammasome in inflammatory bowel diseases

Inflammasomes are multiprotein intracellular complexes which are responsible for the activation of inflammatory responses. Among various subtypes of inflammasomes, NLRP3 has been a subject of intensive investigation. NLRP3 is considered to be a sensor of microbial and other danger signals and plays a crucial role in mucosal immune responses, promoting the maturation of proinflammatory cytokines interleukin 1β(IL-1β) and IL-18. NLRP3 inflammasome has been associated with a variety of inflammatory and autoimmune conditions, including inflammatory bowel diseases(IBD). The role of NLRP3 in IBD is not yet fully elucidated as it seems to demonstrate both pathogenic and protective effects. Studies have shown a relationship between genetic variants and mutations in NLRP3 gene with IBD pathogenesis. A complex interaction between the NLRP3 inflammasome and the mucosal immune response has been reported. Activation of the inflammasome is a key function mediated by the innate immune response and in parallel the signaling through IL-1β and IL-18 is implicated in adaptive immunity. Further research is needed to delineate the precise mechanisms of NLRP3 function in regulating immune responses. Targeting NLRP3 inflammasome and its downstream signaling will provide new insights into the development of future therapeutic strategies.

Metabolic approach for regulation of IL-1β in vivo

IL-1β, the autoantibodies to IL-1β and IL-1β-containing immune complex in pleural effusion and cerebrospinal fluid were detected using anti-recombinant human IL-1β monoclonal antibody. The results indicate that IL-1β is detectable in some cases, and the amounts decrease gradually with the chemotherapy following the autoantibodies and the specific detectable immune complex. These autoantibodies may have a feedback effect, inhibiting the IL-1β activity, and forming the immune complex to speed the clearance of activated 1L-1β.

AIM.:To evaluate the association between the interleukin 1β(IL-1β) polymorphisms and the pancreatic neuroendocrine tumor(pN ET) development.METHODS.:A case-control study was conducted analyzing IL-1β polymorphisms using germline DNA collected in a population-based case-control study of pancreatic cancer(51 pN ET cases,85 pancreatic ductal adenocarcinoma cases,19 intraductal papillary mucinous neoplasm and 98 healthy controls).RESULTS.:The distribution of genotypes for the-511C/T polymorphism in the pN ET patient groups showed significant difference compared to the control group.It is known that the carriers of the IL-1β-511 T allele have increased concentrations of IL-1β.The-511 CT and TT high-expression genotypes were over-represented in pN ET patients.CONCLUSION.:The findings of this study suggested a possible role of IL-1β-511 C/T genotypes in the pathogenesis of pN ETs since the presence of the IL-1β-511 CT and TT genotypes and the T allele was associated with an increased risk of pN ET only.

目的探讨STAT-1对白介素1β(IL-1β)诱导的关节软骨细胞衰老的影响及可能机制。方法软骨细胞同步化后分为:正常对照组(正常培养的软骨细胞);IL-1β组(加入IL-1β,浓度为10 ng/ml);阴性对照组(加入IL-1β和空白质粒);STAT-1转染组(加入IL-1β和siRNA-STAT-1重组质粒)。检测各组细胞的β-半乳糖苷酶染色阳性率;采用MTT法检测4组细胞培养24、48、72 h时的增殖情况;Western blotting法检测4组细胞STAT-1蛋白的表达水平;采用端粒酶检测试剂盒进行端粒酶活性测定。结果细胞培养24 h时,各组细胞的增殖率比较,差异无统计学意义(F=3.037,P=0.087)。细胞培养48 h和72 h时,各组细胞的增殖率比较,差异均有统计学意义(F=3.754,P1,P1β组、阴性对照组细胞增殖率均分别低于正常对照组、STAT-1转染组,差异均有统计学意义(P12.11±1.34)%,IL-1β组为(65.62±2.56)%,阴性对照组为(60.73±3.21)%,STAT-1转染组为(21.32±2.21)%,4组细胞β-半乳糖苷酶染色阳性率比较,差异有统计学意义(F=2.877,P1β组和阴性对照组均高于正常对照组和STAT-1转染组,差异均有统计学意义(P1表达水平比较,差异有统计学意义(F=236.150,P1β组和阴性对照组STAT-1表达水平均高于正常对照组和STAT-1转染组,差异均有统计学意义(P1.65±5.28)%,IL-1β组为(60.32±5.32)%,STAT-1转染组为(76.76±3.45)%。3组端粒酶活性比较,差异有统计学意义(F=26.040,P1β组端粒酶活性低于正常对照组和STAT-1转染组,差异均有统计学意义(P1在IL-1β诱导的衰老关节软骨细胞中表达水平升高,可能通过影响端粒酶活性而参与软骨细胞的衰老。

目的:探讨甲状腺乳头状癌组织中炎性因子的表达及其与甲状腺乳头状癌临床病理特征的关系,为指导临床用药和判断预后奠定实验基础。方法:选取穿刺确诊为甲状腺乳头状癌的74例住院患者作为实验组,另选同期26名体检健康者作为正常对照组。2组受试者均空腹抽取静脉血,实验组患者于术后留取癌组织、癌旁正常组织(距癌组织2cm)及术后7d再次空腹抽取静脉血。采用酶联免疫法检测血清、癌旁正常组织及癌组织中白细胞介素1α(IL-1α)、IL-1β和环氧合酶-2(COX-2)蛋白的表达水平;采用实时荧光定量法和免疫组织化学方法检测甲状腺乳头状癌组织及癌旁正常组织中IL-1α、IL-1β和COX-2的mRNA和蛋白表达水平;分析IL-1α、IL-1β和COX-2表达与甲状腺乳头状癌的临床分期、病理类型及有无淋巴结转移的关系。结果:实验组患者血清中IL-1α、IL-1β和COX-2的蛋白表达水平明显高于正常对照组(P1);甲状腺乳头状癌患者癌组织中IL-1α、IL-1β和COX-2的蛋白表达水平、蛋白阳性表达强度及mRNA表达水平明显高于癌旁正常组织(P1)。在甲状腺乳头状癌组织中IL-1α与IL-1β、IL-1α与COX-2、IL-1β与COX-2均呈正相关关系(r=0.64,P=0.035;r=0.71,P=0.042;r=0.69,P=0.038)。结论:甲状腺乳头状癌组织中IL-1α、IL-1β和COX-2表达水平上调,提示炎性因子参与甲状腺乳头状癌浸润和转移,其应用有望成为新的肿瘤治疗策略和手段。

目的探讨补阳还五汤对脑缺血后大鼠脑组织白细胞介素-1β（IL-1β）和肿瘤坏死因子-α（TNF-α）表达的影响.方法按随机数字表法将SD大鼠分为正常对照组、假手术组、模型组、补阳还五汤组,后3组再按给药后1、3和7 d分为亚组,每组5只.采用大脑中动脉闭塞法（MCAO）建立右侧局灶性脑缺血大鼠模型;补阳还五汤组于术后2 h起灌服补阳还五汤10 ml/kg（14.2 g/kg）、每日1次.分别于相应时间点处死大鼠后取脑组织,采用酶联免疫吸附法（ELISA）和逆转录-聚合酶链反应（RT-PCR）测定IL-1β和TNF-α的蛋白及mRNA表达.结果正常对照组和假手术组有低水平的IL-1β和TNF-α蛋白及mRNA表达.模型组IL-1β、TNF-α蛋白及mRNA表达于给药后1 d开始升高,IL-1β蛋白及mRNA表达在3 d时达高峰后开始下降,TNF-α蛋白及mRNA表达于7 d达高峰.与模型组比较,补阳还五汤组给药后1、3和7 d脑组织中IL-1β和TNF-α蛋白及mRNA表达均明显下降[IL-1β蛋白（ng/L）,1 d：90.290±8.693比102.556±13.934,3 d：129.632±11.050比150.117±8.552,7 d：66.185±9.020比91.362±9.901;TNF-α蛋白（ng/L）,1 d：210.341±19.247比236.887±20.137,3 d：267.503±21.006比322.659±15.068,7 d：299.637±17.717比386.678±16.297;IL-1β mRNA,1 d：0.54±0.09比0.64±0.11,3 d：0.80±0.06比0.89±0.07,7 d：0.70±0.09比0.78±0.08;TNF-α mRNA,1 d：0.64±0.09比0.73±0.11,3 d：0.74±0.13比0.85±0.07,7 d：0.82±0.07比0.93±0.08],差异均有统计学意义（P＜0.05或P＜0.01）.结论补阳还五汤可能通过下调缺血脑组织IL-1β和TNF-α的蛋白及mRNA表达来调控炎症因子的释放,从而起到脑保护作用.

Hyponeurotrophinemia in Serum of Women with Polycystic Ovary Syndrome as a Low Grade Chronic Inflammation

Introduction: This study aimed to discuss about neuroimmune-pathophysiology of polycystic ovary syndrome (PCOS). Nerve growth factor (NGF) as modulator of neuro-endocrine-immune (NEI) system plays a critical role in tissue survival and homeostasis maintenance. It is a strong marker for sympathetic nerve activity. Women with PCO have enhanced ovarian NGF production. Over the past few decades, PCOS has been discussed as a low grade chronic inflammatory disease. In this study, we investigated the neuromodulator role of serum NGF and inflammatory cytokines (IL-1α, β, 17A and TNF-α) in women with PCOS. Material and Methods: 171 patients were investigated in two groups: study (n = 85 PCO) and control (n = 86). Serum levels of NGF-α and interleukins IL-1α, IL-1β, IL-17A and TNFα were determined in both groups by ELISA kit. Results: Data of this study showed that the level of NGF-α and IL-17A in serum of PCO women were lower than control group (p α, IL-1β were higher than control group in PCO women (p 1). Conclusion: Probably inflammatory background in PCOS is the main cause of increased serum levels of IL-1α, IL-1β. Reduction of IL-17A and NGF in serum of PCO patients might be under the direct influence of the hyperactivity of sympathetic nervous system (SNS).
