Arrhythmogenic cardiomyopathy(ACM),a fatal heart disease characterized by fibroadipocytic replacement of cardiac myocytes,accounts for 20%of sudden cardiac death and lacks effective *** is often caused by mutations in desmosome proteins,with Desmoglein-2(DSG2)mutations as a common ***,the mechanism underlying the accumulation of fibrofatty in ACM remains unknown,which impedes the development of curative *** we investigated the fat accumulation and the underlying mechanism in a mouse model of ACM induced by cardiac-specific knockout of Dsg2(CS-Dsg2^(-/-)).Heart failure and cardiac lipid accumulation were observed in CSDsg2^(-/-)*** demonstrated that these phenotypes were caused by decline of fatty acid(FA)β-oxidation resulted from impaired mammalian target of rapamycin(m TOR)*** worsened while overexpression of m TOR and 4EBP1 rescued the FAβ-oxidation pathway in CS-Dsg2^(-/-)*** of PPARa by fenofibrate or AAV9-Ppara significantly alleviated the lipid accumulation and restored cardiac *** results suggest that impaired m TOR-4EBP1-PPARa-dependent FAβ-oxidation contributes to myocardial lipid accumulation in ACM and PPARa may be a potential target for curative treatment of ACM.

The stone chip resistance performance of automotive coatings has attracted increasing attention in academic and industrial *** though traditional gravelometer tests can be used to evaluate stone chip resistance of automotive coatings,such experiment-based methods suffer from poor repeatability and high *** main purpose of this work is to develop a CFD-DEM-wear coupling method to accurately and efficiently simulate stone chipbehaviorof automotive coatings inagravelometer *** this end,an approach coupling an unresolved computational fluid dynamics(CFD)method and a discrete element method(DEM)are employed to account for interactions between fluids and large *** order to accurately describe large particles,a rigid connection particle method is *** doing so,each actual non-spherical particle can be approximately described by rigidly connecting a group of non-overlapping spheres,and particle-fluid interactions are simulated based on each component *** erosion wear model is used to calculate the impact damage of coatings based on particlecoating *** spherical particle tests are performed to demonstrate the feasibility of the proposed rigid connection particle method under various air pressure ***,the developed CFD-DEM-wear model is applied to reproduce the stone chip behavior of two standard tests,i.e.,DIN 55996-1 and SAE-J400-2002 *** results are found to be in good agreement with experimental data,which demonstrates the capacity of our developed method in stone chip resistance ***,parametric studies are conducted to numerically investigate the influences of initial velocity and test panel orientation on impact damage of automotive coatings.

Acute lung injury(ALI),as a common clinical emergency,is pulmonary edema and diffuse lung infiltration caused by *** lack of non-invasive alert strategy,resulting in failure to carry out preventive treatment,means high mortality and poor *** of interferon genes(STING)is a key molecular biomarker of innate immunity in response to inflammation,but there is still a lack of STING-targeted *** this study,a novel STING-targeted PET tracer,[~(18)F]FBTA,was labeled with high radiochemical yield(79.7±4.3%)and molar activity(32.5±2.9 GBq/μmol).We confirmed that[~(18)F]FBTA has a strong STING binding affinity(K_d=26.86±6.79 nmol/L)and can be used for PET imaging in ALI mice to alert early lung inflammation and to assess the efficacy of drug *** STING-targeted strategy also reveals that[~(18)F]FBTA can trace ALI before reaching the computed tomography(CT)diagnostic criteria,and demonstrates its better specificity and distribution than[~(18)F]fluorodeoxyglucose([~(18)F]FDG).

A Smartphone-assisted Paper-based Analytical Device for Fluorescence Assay of Hg2+

Rapid,efficiency and portable detection systems in low-resource settings with limited laboratory equipment and technical expertise are urgently ***,an integrated platform composed of a paper-based analytical device and a smartphone detection system for Hg^2+ onsite testing was ***-doped carbon dots(N-CDs)were synthesized by a simple hydrothermal method using citric acid as the carbon source and ethanediamine as the nitrogen source,which gave out bright blue fluorescence under the excitation at 350 nm UV light and the absolute fluorescence quantum yield was 17.1%.The fluorescence of the prepared N-CDs can be effectively quenched by Hg^2+.In addition,an external attachment of smartphone for illumination and external light interference was designed to trace the fluorescence signals,and a software application of Android system with simple operation program was developed to perform snapshot and image *** smartphone-assisted detection system was combined with the N-CDs decorated paper chip to achieve the sensitive detection of Hg^2+ in water *** integrated method for reliable sensing of Hg^2+ shows a good linear detection range of 10–800μmol/L(R2=0.9595)with detection limit of 1.07×10^?8 mol/L.

Risk factors for very low-level viremia in patients with chronic hepatitis B virus infection: A single-center retrospective study

Two decades of suspect evidence for adaptive molecular evolution——negative selection confounding positive-selection signals

There has been a large literature in the last two decades affirming adaptive DNA sequence evolution between *** main lines of evidence are from(i) the McDonald-Kreitman(MK) test,which compares divergence and polymorphism data,and(ii) the phylogenetic analysis by maximum likelihood(PAML) test,which analyzes multispecies divergence ***,we apply these two tests concurrently to genomic data of Drosophila and *** our surprise,the> 100 genes identified by the two tests do not overlap beyond random *** the non-concordance could be due to low powers leading to high false negatives,we merge every 20-30 genes into a 'supergene'.At the supergene level,the power of detection is large but the calls still do not *** rule out methodological reasons for the *** particular,extensive simulations fail to find scenarios whereby positive selection can only be detected by either MK or PAML,but not *** molecular evolution is governed by positive and negative selection concurrently,a fundamental assumption for estimating one of these(say,positive selection) is that the other is ***,in a broad survey of primates,birds,Drosophila and Arabidopsis,we found that negative selection rarely stays constant for long in *** a consequence,the variation in negative selection is often misconstrued as a signal of positive *** conclusion,MK,PAML and any method that examines genomic sequence evolution has to explicitly address the variation in negative selection before estimating positive *** a companion study,we propose a possible path forward in two stages—first,by mapping out the changes in negative selection and then using this map to estimate positive *** now,the large literature on positive selection between species has to await reassessment.

Extensive gene flow in secondary sympatry after allopatric speciation

Temporal and vertical distribution of microphytobenthos biomass in mangrove sediments of Zhujiang (Pearl River) Estuary

Being a primary producer, Microphytobenthos （MPB） play an important role in the benthic ecosystem of mangrove swamps. The temporal and vertical distribution of MPB biomass, expressed as chlorophyll a （Chl a）, was investigated in mangrove swamps on the Qi＇ao island and the Futian Mangrove Nature Reserve, Guangdong Province, South China. Both locations are situated in the Zhujiang （Pearl River） Estuary. For each location, bi-monthly sampling was carried out at four sites during low tide period. Except one site on the Qi＇ao Island, which was in Phragrnites australis marsh, all sites were in mangrove swamps. The Chl a concentration in surface （0-1 cm） sediment ranged from 0.2/2g/cm3 to 8.3 #g/cm3 in the Qi＇ao Island and from 2.1/zg/cm3 to 15.6/2g/cm3 in the Futian Mangrove Reserve. The peak Chl a concentration occurred in winter or early spring, while the lowest Chl a concentration, with a value of about one quarter of the peak, was observed in summer or early autumn. The vertical distribution of Chl a concentration exhibited an exponential decline with depth, which indicated a muddy sediment with high organic matter, and the slope of the curve was positively related to Chl a concentration in the surface sediment. The MPB biomass of the Qi＇ao Island was significantly lower than that of the Futian Mangrove Reserve. Our results suggest that the overlaying water quality might influence the MPB biomass in surface sediments.

Imparting Pulley Effect and Self-healability to Cathode Binder of Li-S Battery for Improvement of the Cycling Stability

To construct structurally stable sulfur cathode of Li-S battery with improved cycling performance,poly(acrylic acid)(PAA)crosslinked by cationic hydroxypropyl polyrotaxane(HPRN+)via dynamically reversible boronic ester bonds is synthesized and serves as the cathode *** smart polymer networks offer multifunction including buffering the volume change of the cathode during charge/discharge through the pulley effect of polyrotaxanes(PR),suppressing the shuttle effect by adsorption of polysulfide using the plentiful carboxyl,hydroxyl and quaternary ammonium cationic groups,and self-healing the micro-damages to ensure stable conduction pathways of the *** a result,the Li-S batteries based on this novel multifunctional binder and simple commercial sulfur/carbon composites cathode exhibit excellent specific capacity and cycling *** particular,the specific capacity decay per cycle of the cell is only 0.064%along with high Coulombic efficiency after 550 cycles at 1.0 C,which is superior to most of the reported *** under high sulfur loading,moreover,the cathode can deliver superior areal capacity and cycling *** proposed binder provides a new way for the design of high-stability sulfur cathodes.

Effect of trace yttrium on the microstructure,mechanical property and corrosion behavior of homogenized Mg-2Zn-0.1Mn-0.3Ca-xY biological magnesium alloy

The effects of trace yttrium(Y)element on the microstructure,mechanical properties,and corrosion resistance of Mg-2Zn-0.1Mn-0.3Ca-xY(x=0,0.1,0.2,0.3)biological magnesium alloys are *** show that grain size decreases from 310 to 144µm when Y content increases from 0wt%to 0.3wt%.At the same time,volume fraction of the second phase increases from 0.4%to 6.0%,yield strength of the alloy continues to increase,and ultimate tensile strength and elongation decrease initially and then *** the Y content increases to 0.3wt%,Mg_(3)Zn_(6)Y phase begins to precipitate in the alloy;thus,the alloy exhibits the most excellent mechanical *** this time,its ultimate tensile strength,yield strength,and elongation are 119 MPa,69 MPa,and 9.1%,*** addition,when the Y content is 0.3wt%,the alloy shows the best corrosion resistance in the simulated body fluid(SBF).This investigation has revealed that the improvement of mechanical properties and corrosion resistance is mainly attributed to the grain refinement and the precipitated Mg_(3)Zn_(6)Y phase.
