Evaluation of antioxidants and argpyrimidine in normal and cataractous lenses in north indian population

AIM： To assess the level of glutathione, thioltransferase, and argpyrimidine in nuclear and cortical cataractous lenses as well as in the clear lenses in the north indian ***： Human cataractous lenses were collected from the patients who underwent extracapsular cataract extraction surgery; clear lenses were collected from the freshly donated eye bank eyes. Antioxidant molecules such as glutathione and thioltransferase enzyme activity were measured; simultaneously in these lenses a blue fluorophore argpyrimidine, an advanced glycation end（AGE） product level was assessed using high performance liquid chromatography（HPLC）.RESULTS： The protein concentration was found to be present at higher levels in the control lenses compared to cataract lenses. A significant decrease in the glutathione level was observed in the nuclear cataractous lenses compared to cortical cataractous（P=0.004） and clear lenses（P≤0.005）, but no significant change in the level of antioxidant enzyme thioltransferase was observed. Further, argpyrimidine a blue fluorophore（AGE） was found to be significantly higher in the nuclear cataract（P=0.013） compared to cortical cataract ***： Antioxidants such as glutathione significantly decrease in age-related nuclear and cortical cataract and an AGE, argpyrimidine are present at significantly higher levels in nuclear cataract.

Objective:The objective of this study was to evaluate the in vitro antiplasmodial properties against malaria parasite in 15 plants mentioned in indian traditional medicine ***:In vitro antiplasmodial activity of methanolic extracts obtained from indian traditional medicinal plants was evaluated on Plasmodium falciparum of FCK2 and INDO strains using schizont maturation inhibition assay and parasite lactate dehydrogenase inhibition ***:Methanolic extracts of Adhatoda zeylanica,Embelia ribes,Piper nigrum and Plumbago zeylanica exhibited more than 50%inhibition in both the stains in schizont maturation inhibition *** extracts of seven medicinal plants exhibited antiplasmodial activity at half maximal inhibitory concentration(IC50)<100 mg/m L,and methanolic extracts of five medicinal plants exhibited antiplasmodial activity at IC50<50 mg/m L in *** lactate dehydrogenase(PfLDH)inhibition ***,*** and *** exhibited promising antiplasmodial activity in PfLDH inhibition *** and *** exhibited improved activity against resistant in comparison to sensitive ***:*** and *** were the most promising extracts from this study and deserve further investigation of their antiplasmodial properties.

Evaluation of the anti-dyslipidemic effect of spice fixed oils in the in vitro assays and the high fat diet-induced dyslipidemic mice

Dyslipidemia is a condition in which the lipid levels are altered in the *** fixed oils(FOs)from spices,Alpinia galanga,Cinnamomum zeylanicum,Trigonella foenum-graecum,Foeniculum vulgare,and Myristica fragrans were evaluated as potential sources of anti-dyslipidemic *** nutraceutical compound fractions such as polyphenols,tocopherols,γ-oryzanol,and phytosterols were extracted from spice FOs,tested individually and in combination for cytotoxicity and their effect on lipid accumulation in 3T3-L1 and Hep-G2 cell ***,the best performing FOs in the in vitro studies were fed to high fat diet-induced dyslipidemic C57BL/6 mice and studied its activity in the liver and adipose *** in vitro assays showed no cytotoxicity in the studied FOs up to 200μg/mL *** of all the studied nutraceutical compound fractions,theγ-oryzanol and phytosterols fractions showed a reduction in the accumulation of lipid droplets in 3T3-L1 cell ***,the reconstituted nutraceutical fractions from ***,***-graecum,and *** FOs exhibited a dose dependent inhibition of lipid accumulation compared to the fractions of *** and *** *** in vivo studies confirmed the anti-dyslipidemic activity of *** FO that showed an improved lipid profile,antioxidant enzymes,and reduced lipid droplets in the liver and adipose *** histopathological images of the liver and adipose tissue showed a comparable visualisation in Atorvastatin,and *** fed group ***,the spice FOs are potentially efficacious anti-dyslipidemic functional food ingredients and could be safer than statins.

Curcumin reverses T cell-mediated adaptive immune dysfunctions in tumor-bearing hosts

Immune dysfunction is well documented during tumor progression and likely contributes to tumor immune ***81 cytotoxic T lymphocytes(CTLs)are involved in antigen-specific tumor destruction and CD41 T cells are essential for helping this CD81 T cell-dependent tumor *** often target and inhibit T-cell function to escape from immune *** dysfunction includes loss of effector and memory T cells,bias towards type 2 cytokines and expansion of T regulatory(Treg)*** has previously been shown to have antitumor activity and some research has addressed the immunoprotective potential of this plant-derived polyphenol in tumor-bearing *** we examined the role of curcumin in the prevention of tumor-induced dysfunction of T cell-based immune *** observed severe loss of both effector and memory T-cell populations,downregulation of type 1 and upregulation of type 2 immune responses and decreased proliferation of effector T cells in the presence of ***,in turn,prevented this loss of T cells,expanded central memory T cell(TCM)/effector memory T cell(TEM)populations,reversed the type 2 immune bias and attenuated the tumor-induced inhibition of T-cell proliferation in tumor-bearing *** investigation revealed that tumor burden upregulated Treg cell populations and stimulated the production of the immunosuppressive cytokines transforming growth factor(TGF)-b and IL-10 in these ***,however,inhibited the suppressive activity of Treg cells by downregulating the production of TGF-b and IL-10 in these *** importantly,curcumin treatment enhanced the ability of effector T cells to kill cancer ***,our observations suggest that the unique properties of curcumin may be exploited for successful attenuation of tumor-induced suppression of cell-mediated immune responses.

Objective Calcium Glucarate (Cag), Ca salt of D-glucaric acid is a naturally occurring non-toxic compound present in fruits, vegetables and seeds of some plants, and suppress tumor growth in different models. Due to lack of knowledge about its mode of action its uses are limited in cancer chemotherapy thus the objective of the study was to study the mechanism of action of Cag on mouse skin tumorigenesis. Methods We have estimated effect of Cag on DMBA induced mouse skin tumor development following complete carcinogenesis protocol. We measured, epidermal transglutaminase activity (TG), a marker of cell differentiation after DMBA and/or Cag treatment and [3H] thymidine incorporation into DNA as a marker for cell proliferation. Results Topical application of Cag suppressed the DMBA induced mouse skin tumor development. Validity of subjective assessment as screening tool for dry eye disease and its association with clinical tests

AIM: To determine the role of subjective assessment using Mc Monnies dry eye questionnaire in diagnosing dry eye disease and its association with clinical ***: There were 500 patients screened for dry eye using Mc Monnies dry eye questionnaire between May to October 2013 at the outpatient Department of Ophthalmology of a medical college hospital. All 500 patients were subjected to clinical tests. Dry eye was defined as having one or more symptoms often or all the time. Positive signs were if one or both eyes revealed tear film breakup time(TBUT) of ≤10s, a Schirmer test score of ≤10 mm, a Rose Bengal staining score of ≥1, a Lissamine green staining score of ≥1 or existence of meibomian gland disease(≥grade 1). Statistical analysis was performed to describe the distribution of symptoms and signs, to assess the correlations between Mc Monnies score(MS) and variable clinical signs of dry eye, and to explore the association between dry eye symptoms and variable clinical signs. Analysis was performed using software package Epi info. A Probability(P) value using Chi-square test of 20), 75.86% had a low TBUT(<10s), 58.62% had a low Schirmer’s I test(≤10 mm),86.20% had Rose Bengal staining score of ≥1, 79.31%had Lissamine green staining score of ≥1. We found statistically significant associations between positive Schirmer test and arthritis(P <0.002), dryness elsewhere(P <0.001), contact lens use(P < 0.002), systemic medication(P <0.0001), sleeping with eyes partly open(P <0.002), history of dry eyes treatment(P <0.0001),environmental factors(P <0.001), swimming(P <0.001).· CONCLUSION: Subjective assessment plays an important role in diagnosing dry eye disease. Comparing acid steatocrit and faecal elastase estimations for use in M-ANNHEIM staging for pancreatitis

AIM To compare two tests for exocrine pancreatic function(EPF) for use in M-ANNHEIM staging for pancreatitis. METHODS One hundred and ninety four consecutive patients with acute pancreatitis(AP; n = 13), recurrent acute pancreatitis(RAP; n = 65) and chronic pancreatitis(CP; n = 116) were enrolled. EPF was assessed by faecal elastase-1(FE-1) estimation and stool fat excretion by the acid steatocrit method. Patients were classified as per M-ANNHEIM stages separately based on the results of the two tests for comparison. Independent Student's t-test, χ~2 test, Kruskal-Wallis test, Mann-Whitney U test and Mc Nemar's test were used as appropriate. RESULTS Sixty-one(52.5%) patients with CP had steatorrhoea when assessed by the acid steatocrit method; 79 (68.1%) with CP had exocrine insufficiency by the FE-1 test(χ~2 test, P < 0.001). The results of acid steatocrit and FE-1 showed a significant negative correlation(Spearman's rho =-0.376, P < 0.001). A statistically significant difference was seen between the M-ANNHEIM stages as classified separately by acid steatocrit and the FE-1. Thirteen(6.7%), 87(44.8%), 89(45.8%) and 5(2.5%) patients were placed in M-ANNHEIM stages 0,?Ⅰ, Ⅱ, and Ⅲ respectively, with the use of acid steatocrit as against 13(6.7%), 85(43.8%), 75(38.6%), and 21(10.8%) respectively by FE-1 in stages 0,?Ⅰ, Ⅱ, and Ⅲ thereby altering the stage in 28(14.4%) patients(P < 0.001, Mc Nemar's test). CONCLUSION FE-1 estimation performed better than the acid steatocrit test for use in the staging of pancreatitis by the M-ANNHEIM classification since it diagnosed a higher proportion of patients with exocrine insufficiency.

Comprehensive Application of Graphene: Emphasis on Biomedical Concerns

Graphene, sp^2 hybridized carbon framework of one atom thickness, is reputed as the strongest material to date. It has marked its impact in manifold applications including electronics, sensors, composites, and catalysis. Current state-of-the-art graphene research revolves around its biomedical applications. The two-dimensional(2D) planar structure of graphene provides a large surface area for loading drugs/biomolecules and the possibility of conjugating fluorescent dyes for bioimaging. The high near-infrared absorbance makes graphene ideal for photothermal therapy. Henceforth, graphene turns out to be a reliable multifunctional material for use in diagnosis and treatment. It exhibits antibacterial property by directly interacting with the cell membrane. Potential application of graphene as a sca old for the attachment and proliferation of stem cells and neuronal cells is captivating in a tissue regeneration scenario. Fabrication of 2D graphene into a 3D structure is made possible with the help of 3D printing, a revolutionary technology having promising applications in tissue and organ engineering. However, apart from its advantageous application scope, use of graphene raises toxicity concerns. Several reports have confirmed the potential toxicity of graphene and its derivatives, and the inconsistency may be due to the lack of standardized consensus protocols. The present review focuses on the hidden facts of graphene and its biomedical application, with special emphasis on drug delivery, biosensing, bioimaging, antibacterial, tissue engineering, and 3D printing applications.

Contraception with RISUG and functional reversal through DMSO and NaHCO3 in male rabbits

The study aimed to evaluate reversal of short- and long-term vas occlusion with reversible inhibition of sperm under guidance （RISUG） using dimethyl sulfoxide （DMSO） and sodium bicarbonate （NaHCO3） in male rabbits （Oryctolagus cuniculus）. Animals were divided into seven groups containing five animals each. Fortnightly, semen analysis revealed that sperm concentration and output steadily declined after vas occlusion and complete azoospermia was attained at 30-60 days postinjection. Spermatozoa reappeared at 60-75 days of reversal and normozoospermia was noticed between 135 days and 150 days in the reversal groups. All spermatozoa were found nonmotile prior to azoospermia and a gradual recovery in sperm motility was observed between 105 days and 135 days of reversal. A significant decline in viability of sperms was noticed during vas occlusion up to 30-60 days which recovered at 60-75 days postreversal and normalized by 75-105 days in the reversal groups. A significant enhancement in the sperm abnormalities was recorded in all vas occluded animals as well as those in initial periods of reversal. Other parameters, namely, semen volume, ejaculation time, pH, color, and consistency, remained unaltered during all phases of the study. Fertility test, at the intervals of 15 days, demonstrated that animals exhibited complete sterility during the entire period of vas occlusion. A gradual recovery in fertility was observed with the appearance of spermatozoa following vas occlusion reversal and 100% fertility was observed following 135-150 days of reversal. F, progeny of reversed animals was found normal. The results suggest that reversal with DMSO or NaHCO3 is feasible, with normal progeny, following short- and long-term contraception.
