Effects of Longyanshen polysaccharides on free radical metabolism in senescence accelerated-prone mice

BACKGROUND： Along with aging, antioxidase activity decreases and oxygen-derived free radicals greatly accumulate, resulting in cellular senescence, or even cell death. This is manifested by hypomnesia and disordered metabolism of free radicals. Studies have reported that Longyanshen polysaccharides have the function of antioxidation and improved brain memory. OBJECTIVE： To observe the effects of Longyanshen polysaccharides on free radical metabolism in brain tissue to verify the anti-aging mechanisms in senescence accelerated-prone （SAMP8） mice. DESIGN, TIME AND SETTING： The randomized, controlled, biochemical experiment was performed in the Department of Pharmacology and scientific Experimental Center of guangxi Medical University （China） from September 2005 to January 2008. MATERIALS： Forty SAMP8 mice were randomized into four groups： SAMP8 control group, as well as low-, mid-, and high-dose polysaccharide, with 10 mice in each group. Ten senescence accelerated-resistantprone （SAMR 1） mice served as the normal control group. Longyanshen polysaccharides, extracted from the medical plant Longyanshen, were supplied by the Department of Pharmacology, guangxi Medical University Superoxide dismutase （SOD）, glutathione peroxidase （GSH-Px）, malonaldehyde （MDA）, nitric oxide （NO）, and total protein test kitwere purchased from Nanjing Jiancheng Bioengineering Institute （China）. METHODS： SAMP8 mice were used to establish a dementia animal model. SAMP8 and SAMR1 control mice were administered 30 mL/kg saline. The low-, middle-, and high-dose polysaccharide groups were administered 45, 90, and 180 mg/kg Longyanshen polysaccharides, respectively. Each group was treated by intragastric administration, once daily, for 50 continuous days. MAIN OUTCOME MEASURES： One hour after the last administration, mouse brain tissues were collected, and retro orbital blood sampling was performed. BACKGROUND： During onset and development of Alzheimer＇s disease, β-amyloid （Aβ） precursor protein （APP）, β-site amyloid precursor protein cleaving enzyme （BACE）, and β-amyloid are key genes and proteins in the Aβ pathway, and over-expression of these genes can lead to Aβ deposit/on in the brain. OBJECTIVE： To observe the influence of Longyanshen polysaccharides on expression of BACE, APP, and Aβ in the senescence-accelerated mouse prone/8 （SAMP8） brain, and to compare these effects with huperzine A treatment. DESIGN, TIME AND SETTING： A randomized, controlled, neurobiochemical experiment was performed at the Department of Pharmacology and scientific Experimental Center of guangxi Medical University from September 2005 to January 2008. MATERIALS： Longyanshen polysaccharfdes powder was extracted from the dried slices of the medicinal plant Longyanshen. The active component, Longyanshen polysaccharides, was provided by the Department of Pharmacology, guangxi Medical University; huperzine A was purchased from Yuzhong Drug Manufactory, China. METHODS： Healthy SAMP8 mice were used to establish a model of Alzheimer＇s disease. A total of 50 SAMP8 mice were randomly assigned to 5 groups （n = 10）： SAMP8, huperzine A, low-, middle-, and high-dose polysaccharides. In addition, 10 senescence-accelerated mouse resistant 1 （SAMR1） mice were selected as normal controls. SAMP8 and SAMR1 mice were administered 30 mL/kg normal saline; the huperzine A group was administered 0.02 mg/kg huperzine A; the low-, middle-, and high-dose polysaccharides groups were respectively administered 45, 90, and 180 mg/kg Longyanshen polysaccharides. Each group was treated by intragastric administration, once per day, for 50 consecutive days. Degradation kinetics and mechanism of trace nitrobenzene by granular activated carbon enhanced microwave/hydrogen peroxide system

The kinetics of thedegradation of trace nitrobenzene （NB） by a granular activated carbon （GAC） enhanced microwave （MW）/hydrogen peroxide （H202 ） systemwas studied. Effects of pH, NB initial concentration and tert-butyl alcohol on the removal efficiencywere examined. Itwas found that the reaction rate fitswell to first-order reaction kinetics in the MW/GAC/H202 process. Moreover, GAC greatly enhanced thedegradation rate of NB inwater. Under a given condition （MW power300 W, H202dosage 10 mg/L, pH 6.85 and temperature （60 ± 5）°C）, thedegradation rate of NBwas 0.05214 min 1when4 g/L GACwas added. In general, alkaline pHwas better for NBdegradation; however, the optimum pHwas 8.0 in the tested pH value range of4.0-12.0. At H202dosage of 10 mg/L and GACdosage of4 g/L, the removal of NBwasdecreasedwith increasing initial concentrations of NB, indicating that a low initial concentrationwas beneficial for thedegradation of NB. These results indicated that the MW/GAC/H202 processwas effective for trace NBdegradation inwater. Gas chromatography-mass spectrometry analysis indicated that a hydroxyl radical addition reaction anddehydrogenation reaction enhanced NBdegradation.

Characterization of precipitation in the background of atmospheric pollutants reduction in Guilin: Temporal variation and source apportionment

Rainfall samples were collected from three observation sites in Guilin from 2013 to 2017, and the chemical composition characteristics of precipitation and the contribution made by different ion sources were analyzed when atmospheric pollutants levels were reduced. The results showed that acid gas emissions and atmospheric pollutant concentrations continued to decline during the study period. However, the change in the volume-weighted mean p H at the three sites suggested that acid rain pollution was not alleviated and began to deteriorate after 2015. The continuing downward trend for alkaline neutralizing ions(Ca^(2+), NH_4^+) in precipitation indicated that the reduction in alkaline neutralizing substances in the atmosphere was an important factor that led to the deterioration in acid rain across Guilin. The principal component analysis and spearman correlation analysis indicated five sources of ions in precipitation. Quantitative assessment of these five sources indicated that fossil fuel combustion contributed the most ions concentration in precipitation at the three sites, followed by agriculture, terrestrial(crustal) sources, marine sources, and biomass burning. Long-distance airflow might affect the acidity, the electrical conductivity(EC), and ion concentrations in precipitation across Guilin. The airflow trajectory from the west and southeast directions corresponded to higher acidity and ion concentrations. According to the current air pollution control strategy planned by Guilin, reducing atmospheric coarse particles and NH_3 at the same time may potentially lead to further deteriorations in acid rain contents. Therefore, Guilin needs to develop more reasonable pollution prevention measures that synergistically control atmospheric pollutants and acid rain pollution.

HMGB 1 contributes to allergen-induced airway remodeling in a murine model of chronic asthma by modulating airway inflammation and activating lung fibroblasts

The pro-inflammation factor high-mobility group box protein 1 （HMGB1） has been implicated in the pathogenesis of asthma. In this study, we used a murine model of chronic asthma to evaluate the effects of HMGB 1 on airway remodeling. Female BALB/c mice were randomly divided into four groups： control, ovalbumin （OVA） asthmatic, OVA＋ isotype antibody and OVA＋anti-HMGB 1 antibody. Anti-HMGB 1 antibody therapy was started on day 21 and was administered three times per week for 6 weeks before intranasal challenge with OVA. In this mouse model, HMGB1 expression is significantly elevated. The anti-HMGB1 antibody group exhibited decreased levels of immunoglobulin E （IgE） and inflammatory mediators and reduced inflammatory cell accumulation, airway hyperresponsiveness （AHR）, mucus synthesis, smooth muscle thickness and lung collagen content compared with the OVA groups. Treatment with HMGB1 increased proliferation, migration, collagen secretion and a-smooth muscle actin （SMA） expression in MRC-5 ceils. Treatment with the HMGB1/IL-1β complex significantly increased the expression and secretion of transforming growth factor （TGF-βl）, matrix metalloproteinase （MMP）-9 and vascular endothelial growth factor （VEGF）. Altogether, these results suggest that blocking HMGB1 activity may reverse airway remodeling by suppressing airway inflammation and modulating lung fibroblast phenotype and activation.

Cartilage defects may lead to severe degenerative joint *** engineering based on type I collagen hydrogel that has chondrogenic potential is ideal for cartilage ***,the underlying mechanisms of chondrogenic differentiation driven by type I collagen hydrogel have not been fully ***,we explored potential collagen receptors and chondrogenic signaling pathways through bioinformatical analysis to investigate the mechanism of collagen-induced *** showed that the super enhancer-related genes induced by collagen hydrogel were significantly enriched in the TGF-b signaling pathway,and integrin-b1(ITGB1),a receptor of collagen,was highly expressed in bone marrow mesenchymal stem cells(BMSCs).Further analysis showed genes such as COL2A1 and Tenascin C(TNC)that interacted with ITGB1 were significantly enriched in extracellular matrix(ECM)structural constituents in the chondrogenic induction *** of ITGB1 led to the downregulation of cartilage-specific genes(SOX9,ACAN,COL2A1),SMAD2 and TNC,as well as the downregulation of phosphorylation of SMAD2/*** of TNC also resulted in the decrease of cartilage markers,ITGB1 and the SMAD2/3 phosphorylation but overexpression of TNC showed the opposite ***,in vitro and in vivo experiments confirmed the involvement of ITGB1 and TNC in collagen-mediated chondrogenic differentiation and cartilage *** summary,we demonstrated that ITGB1 was a crucial receptor for chondrogenic differentiation of BMSCs induced by collagen *** can activate TGF-SMAD2/3 signaling,followed by impacting TNC expression,which in turn promotes the interaction of ITGB1 and TGF-SMAD2/3 signaling to enhance *** may provide concernful support for cartilage tissue engineering and biomaterials development.

Highly time-resolved aerosol characteristics during springtime in Weizhou Island

A single particle aerosol mass spectrometer was deployed to measure the changes of single particle species and sizes during March 2015 in Weizhou Island of the Beibu Gulf, guangxi province, South China. In this campaign, a total of 3,100,597 particles were sized, and 25.8%particles with both positive and negative mass spectrum were collected and 24.8%characterized in combination with the ART-2 a neural network algorithm. The distribution of sized particles was mainly in from 520 to 600 nm, and the diameters ranging from 340 to1000 nm accounted for above 90%. Eight types of particles were classified: Elemental Carbon containing(EC), Organic Carbon containing(OC), EC and OC combined containing particles,Na containing particles, K containing particles(K), Levoglucosan containing particles,mineral containing particles, and Heavy Metal containing particles(HM). EC, OC and K were the major containing particles, which accounted for 84.3% in the eight types particles. The relative ratio and size distribution of the three types were EC(48.1%, 620 nm), OC(12.7%,440 nm), and K(23.5%, 600 nm), respectively. The three types of particles were a bit increasing ratios compared with those in clean periods during haze pollution *** with the back-trajectory results from the Hysplit-4 model and local pollution sources revealed that the ambient air quality on the Weizhou Island may be influenced by biomass burning in the Indochina Peninsula(biomass burning in the Indochina Peninsula)from the transportation on higher level atmospheric layer and by mainland of south China located northeast of Weizhou Island on the ground.

It is well-known that the electrolytes can influence the electrochemical reduction of carbon dioxide(ERCO2)in aqueous *** this work,we explore the effects of alkali metal cations and anions(Li^+,Na^+,K^+,Rb^+,Cs^+,HCO3^-,Cl^-,Br^-,I^-)on the current density and product selectivity for the ERCO2 into formic acid(HCOOH)on the SnO2/carbon paper(Sn O2/C)*** of the ERCO2 experiments show that for the cations,the promotion effects on current density and faradaic efficiencies(FEs)are in the order of Li^+b Na^+b K^+b Cs^+b Rb^+.For the anions,the current density values are in the order of Na HCO3 b NaClb Na Br b Na I and KHCO3 b KCl≈KI b KBr,respectively,and that on the FEs for the formation of the HCOOH(FEHCOOH)is HCO3-b Cl-b Br-b I-.Based on this result,the effects of alkali metal cations and anions on ERCO2 are discussed.

The title complexes based on 4（3H）-quinazolinone ligand have been synthesized and characterized by elemental analysis, IR spectrum and single-crystal X-ray diffraction *** 1 belongs to the triclinic system, space group P1, with a = 6.5392（3）, b = 7.4402（5）, c =10.3397（7）？, α = 83.204（5）, β = 75.465（5）, γ = 86.075（5）° and V = 483.88（5）？~3. Complex 2 is of orthorhombic system, space group Pbca, with a = 10.1294（3）, b = 7.6747（5）, c = 23.8171（15）？ and V = 1849.1（4）？~3. Complex 3 belongs to the orthorhombic system, space group Pbca, with a =12.7188（2）, b = 6.66582（9）, c = 25.6987（6）？ and V = 2178.77（5）？~3. The complex structures are affected by the synthesis conditions.
