Locust(Locusta migratoria)has a single striated muscle myosin heavy chain(Mhc)gene,which contains 5 clusters of alternative exclusive exons and 1 differently included penultimate *** alternative exons of Mhc gene encode 4 distinct regions in the myosin motor domain,that is,the N-terminal SH3-like domain,one lip of the nucleotide-binding pocket,the relay,and the ***,we investigated the role of the alternative regions on the motor function of locust muscle *** Sf9-baculovirus protein expression system,we expressed and purified 5 isoforms of the locust muscle myosin heavy meromyosin(HMM),including the major isoform in the thorax dorsal longitudinal flight muscle(FL1)and 4 isoforms expressed in the abdominal intersegmental muscle(AB1 to AB4).Among these 5 HMMs,FL1-HMM displayed the highest level of actin-activated adenosine triphosphatase(ATPase)activity(hereafter referred as ATPase activity).To identify the alternative region(s)responsible for the elevated ATPase activity of FL1-HMM,we produced a number of chimeras of FL1-HMM and *** with the relay of AB4-HMM(encoded by exon-14c)substantially decreased the ATPase activity of FL1-HMM,and conversely,the relay of FL1-HMM(encoded by exon-14a)enhanced the ATPase activity of *** showed that the exon-14a-encoded residues Gly474 and Asn509 are responsible for the elevated ATPase activity of *** results indicate that the alternative relay encoded by exon-14a/c play a key role in regulating the ATPase activity of FL1-HMM and AB4-HMM.

Effects of simulated gut pH environment on bacterial composition and pheromone production of Dendroctonus valens

Bark beetles are an economically and ecologically important insect group,with aggregation behavior and thus host colonization success depends on pheromone-mediated *** some species,such as the major invasive forest pest in China,red turpentine beetle(Dendroctonus valens),gut microbiota participates in pheromone production by converting tree monoterpenes into pheromone ***,how variation in gut microenvironment,such as pH,affects the gut microbial composition,and consequently pheromone production,is *** this study,we fed wild caught *** with 3 different pH media(main host diet with natural pH of 4.7;a mildly acidic diet with pH 6 mimicking the beetle gut pH;and highly acidic diet with pH 4),and measured their effects on the gut pH,bacterial community and production of the main aggregation and anti-aggregation pheromone(verbenone).We further tested the verbenone production capacity of 2 gut bacterial isolates in different pH environments(pH 6 and 4).Compared to natural state or main host diet,feeding on less acidic diet(pH 6)diluted the acidity of the gut,whereas feeding on highly acidic diet(pH 4)enhanced *** changes in gut pH reduced the abundance of dominant bacterial genera,resulting in decreased verbenone ***,the highest pheromone conversion rate of the bacterial isolates was observed in pH mimicking the acidity in beetle *** together,these results indicate that changes in gut pH can affect gut microbiota composition and pheromone production,and may therefore have the potential to affect host colonization behavior.

梳理是一种进化上保守的刻板行为，广泛存在于包括人类在内的各种动物中。梳理行为在动物维持体表清洁、调节体温、去觉醒、减轻压力以及社会行为等方面发挥重要功能。在啮齿类动物中，梳理行为通常可以分为四个连续的阶段，并以从头至尾顺序的方式进行重复刻板动作：从口鼻部到面部，到头部，最后以舔舐躯体结束。梳理行为的发生具有环境依赖性，说明此行为具有适应的重要性。本综述简要总结了啮齿类动物梳理行为涉及的神经基质，并探讨了异常梳理行为表型的存在与神经精神疾病和神经退行性疾病模型的相关性。此外，本文进一步强调指出，在神经精神病学模型中，啮齿类动物的梳理行为可作为测定重复刻板行为的一种可靠测量方法，有望在转化精神病学中应用。本文主要聚焦啮齿动物的自我理毛行为，因受限于篇幅，未对allogrooming（同种动物通过舔舐或以仔细啃咬的方式为另一种动物梳理毛发）与heterogrooming（在特定情况下发生的对另一种动物的梳理行为，如母性行为、性行为、攻击行为或社会行为）展开讨论。

Effect of temperature on antioxidant defense and innate immunity in Brandt’s voles

Ambient temperature is an importa nt factor in flue ncing many physiological processes, in eluding antioxidant defense and immunity. In the present study, we tested the hypothesis that antioxidant defense and immunity are suppressed by high and low temperature treatme nt in Bran dt's voles (Lasiopodomys brandtii). Thirty male voles were randomly assigned into different temperature groups (4, 23, and 32℃, n=10 for each group), with the treatment course lasting for 27 d. Results showed that low temperature in creased gross en ergy in take (GEI) and liver, heart, and kidney mass, but decreased body fat mass and dry carcass mass. With the decline in temperature, hydrogen peroxide (H2O2) concentration, which is indicative of reactive oxyge n species (ROS) levels, in creased in the liver, decreased in the heart, and was unchanged in the kidney, testis, and small intestine. Lipid peroxidation indicated by malonaldehyde (MDA) content in the liver, heart, kidney, testis, and small intestine did not differ among groups, implying that high and low temperature did not cause oxidative damage. Similarly, superoxide dismutase (SOD) and catalase (CAT) activities and total antioxidant capacity (TAOC) in the five tissues did not respond to low or high temperature, except for elevation of CAT activity in the testis upon cold exposure. Bacteria killing capacity, which is indicative of innate immunity, was nearly suppressed in the 4℃ group in contrast to the 23℃ group, whereas spleen mass and white blood cells were un affected by temperature treatment. The levels of testosterone, but not corticostero ne, were in flue need by temperature treatment, though neither were correlated with innate immunity, H2O2 and MDA levels, or SOD, CAT, and TAOC activity in any detected tissues. Overall, these results showed that temperature had different in flue nces on oxidative stress, an tioxida nt en zymes, and immunity, which depended on the tissues and parameters tested. Receptor variability-driven evolution of snake toxins

Three-finger toxins （TFTs） are well-recognized non- enzymatic venom proteins found in snakes. However, although TFTs exhibit accelerated evolution, the drivers of this evolution remain poorly understood. The structural complexes between long-chain α-neurotoxins, a subfamily of TFTs, and their nicotinic acetylcholine receptor targets have been determined in previous research, providing an opportunity to address such questions. In the current study, we observed several previously identified positively selected sites （PSSs） and the highly variable C-terminal loop of these toxins at the toxin/receptor interface. Of interest, analysis of the molecular adaptation of the toxin-recognition regions in the corresponding receptors provided no statistical evidence for positive selection. However, these regions accumulated abundant amino acid variations in the receptors from the prey of snakes, suggesting that accelerated substitution of TFTs could be a consequence of adaptation to these variations. To the best of our knowledge, this atypical evolution, initially discovered in scorpions, is reported in snake toxins for the first time and may be applicable for the evolution of toxins from other venomous animals.

Role of transient receptor potential vanilloid-1 in behavioral thermoregulation of the Mongolian gerbil Meriones unguiculatus

Ambient temperature considerably affects the physiology and behavior of *** and thermoregulatory abilities play an important role in the response to changing ambient temperature in ***,the molecular mechanisms of behavioral thermoregulation remain poorly *** receptor potential vanilloid-1(TRPV1)is activated by changes in ambient temperature and is involved in acute ***,we aimed to determine whether TRPV1 is involved in behavioral thermoregulation in wild rodents by conducting 2 *** the first,42 adult Mongolian gerbils(Meriones unguiculatus;14 per treatment)were randomly assigned to 3 housing temperatures(4,23,and 36℃for 4 *** the second,20 gerbils(10 per treatment)were randomly injected with capsaicin(TRPV1 agonist)or AMG517(TRPV1 antagonist).The results showed a significant decrease in food intake and non-shivering thermogenesis in the gerbils housed at 36℃.Additionally,there was a significant increase in the preference of gerbils housed at 4℃ to low *** expression of TRPV1 protein in the brown adipose tissue(BAT)and liver was significantly positively correlated with that of protein kinase A(PKA).The expression of TRPV1 and PKA proteins in the BAT was positively correlated with the temperature preference of the *** gerbils injected with capsaicin preferred significantly lower temperatures than the control group *** findings suggest that TRPV1 and PKA are involved in behavioral thermoregulation in Mongolian gerbils.

An intercontinental comparison of insect seed predation between introduced and native oaks

Novel interactions between introduced oaks and their natural enemies across different continents provide an opportunity to test the enemy release hypothesis(ERH)at local and global *** on the ERH,we assessed the impacts of native seed-feeding insects on introduced and native oaks within and among *** combined a common-garden experiment in China and biogeographic literature surveys to measure seed predation by insects and the proportion of acorn embryos surviving after insect infestation among 4 oak species with different geographical origins:Quercus mongolica origin from China,*** and *** from Europe,and *** from North *** supporting the ERH,oaks in introduced continents escaped seed predation compared to those in native continents and compared to other native oaks in introduced ***-garden comparisons showed that total acorn infestation rate of introduced ***(section Lobatae)was considerably lower than that of native oaks(section Quercus)in China and Europe,likely because of the differences in seed traits associated with different oak *** surveys showed that seed predation of introduced oaks was lower in the introduced continent than in the native *** survival was higher in introduced *** than native oaks in China and ***,insect seed predation of recently introduced *** in China was similar to that in Europe,which is not consistent with the *** results suggest that reduced acorn attack by native insects and higher embryo survival after acorn damage could increase the establishment success or invasion risk of introduced oaks in non-native continents.

Cuticular hydrocarbons form a barrier that protects terrestrial insects from water loss via the *** loads and transports lipids,including hydrocarbons,from one tissue to *** some insects,the lipophorin receptor(LpR),which binds to lipophorin and accepts its lipid cargo,is essential for female fecundity because it mediates the incorporation of lipophorin by developing ***,it is unclear whether LpR is involved in the accumulation of cuticular hydrocarbons and its precise role in aphid reproduction remains *** herein present the results of our molecular characterization,phylogenetic analysis,and functional annotation of the pea aphid(Acyrthosiphon pisum)LpR gene(ApLpR).This gene was transcribed throughout the *** life cycle,but especially during the embryonic stage and in the abdominal ***,we optimized the RHA interference(RNAi)parameters by determining the ideal dose and duration for gene silencing in the pea *** observed that the RNAi-based ApLpR suppression significantly decreased the internal and cuticular hydrocarbon contents as well as adult ***,a deficiency in cuticular hydrocarbons increased the susceptibility of aphids to desiccation stress,with decreased survival rates under simulated drought ***,ApLpR expression levels significantly increased in response to the desiccation *** results confirm that ApLpR is involved in transporting hydrocarbons and protecting aphids from desiccation ***,this gene is vital for aphid ***,the ApLpR gene of *** may be a novel RNAi target relevant for insect pest management.

Traumatic mating causes strict monandry in a wolf spider

DEAR EDITOR,We identified strict monandry in a wandering wolf spider species(Pardosa pseudoannulata) and revealed the underlying mechanisms of this mating structure. Evidence indicated that males damaged the inner walls of the female genital tract with the sharp part of their intromittent organ during insemination. The traumatic mating caused hemolymph hemorrhage, which mixed with seminal fluid and gradually formed an impermeable amorphous mating plug after about15 days, completely blocking the female copulatory opening.
