Activation of LONP1 by 84-B10 alleviates aristolochic acid nephropathy via re-establishing mitochondrial and peroxisomal homeostasis

Pharmaceutical formulations derived from Aristolochiaceae herbs, which contain aristolochic acids(AAs), are widely used for medicinal purposes. However, exposure to these plants and isolated AAs is linked to renal toxicity, known as AA nephropathy(AAN). Currently, the mechanisms underlying AAN are not fully understood, leading to unsatisfactory treatment strategies. In this study, we explored the protective role of 84-B10(5-[[2-(4-methoxyphenoxy)-5-(trifluoromethyl) phenyl] amino]-5-oxo-3-phenylpentanoic acid) against AAN. RNA-seq analysis revealed that the mitochondrion and peroxisome were the most affected cellular components following 84-B10 treatment in AAN mice. Consistently, 84-B10 treatment preserved mitochondrial ultrastructure, restored mitochondrial respiration, enhanced the expression of key transporters(carnitine palmitoyltransferase 2) and enzymes(acylCoenzyme A dehydrogenase, medium chain) involved in mitochondrial fatty acid β-oxidation, and reduced mitochondrial ROS generation in both aristolochic acid I(AAI)-challenged mice kidneys and cultured proximal tubular epithelial cells. Additionally, 84-B10 treatment increased the expression of key transporters(ATP binding cassette subfamily D) and rate-limiting enzymes(acyl-CoA oxidase 1) involved in peroxisomal fatty acid β-oxidation and restored peroxisomal redox balance. Knocking down LONP1 expression diminished the protective effects of 84-B10 against AAN, suggesting LONP1-dependent protection. In conclusion, our study provides evidence that AAN is associated with significant disturbances in both mitochondrial and peroxisomal functions. The LONP1 activator 84-B10 demonstrates therapeutic potential against AAN, likely by maintaining homeostasis in both mitochondria and peroxisomes.

Physics-informed deep learning for fringe pattern analysis

Recently,deep learning has yielded transformative success across optics and photonics,especially in optical *** neural networks (DNNs) with a fully convolutional architecture (e.g.,U-Net and its derivatives) have been widely implemented in an end-to-end manner to accomplish various optical metrology tasks,such as fringe denoising,phase unwrapping,and fringe ***,the task of training a DNN to accurately identify an image-to-image transform from massive input and output data pairs seems at best naive,as the physical laws governing the image formation or other domain expertise pertaining to the measurement have not yet been fully exploited in current deep learning *** this end,we introduce a physics-informed deep learning method for fringe pattern analysis (PI-FPA) to overcome this limit by integrating a lightweight DNN with a learning-enhanced Fourier transform profilometry (Le FTP) *** parameterizing conventional phase retrieval methods,the Le FTP module embeds the prior knowledge in the network structure and the loss function to directly provide reliable phase results for new types of samples,while circumventing the requirement of collecting a large amount of high-quality data in supervised learning *** by the initial phase from Le FTP,the phase recovery ability of the lightweight DNN is enhanced to further improve the phase accuracy at a low computational cost compared with existing end-to-end *** results demonstrate that PI-FPA enables more accurate and computationally efficient single-shot phase retrieval,exhibiting its excellent generalization to various unseen objects during *** proposed PI-FPA presents that challenging issues in optical metrology can be potentially overcome through the synergy of physics-priors-based traditional tools and data-driven learning approaches,opening new avenues to achieve fast and accurate single-shot 3D imaging.

Numerical analysis of cavitation shedding flow around a three-dimensional hydrofoil using an improved filter-based model

The cavitation shedding flow around a 3-D Clark-Y hydrofoil is simulated by using an improved filter-based model （FBM） and a mass transfer cavitation model with the consideration of the maximum density ratio effect between the liquid and the vapor. The unsteady cloud cavity shedding features around the Clark-Y hydrofoil are accurately captured based on an improved FBM model and a suitable maximum density ratio. Numerical results show that the predicted cavitation patterns and evolutions compare well with the experimental visualizations, and the prediction errors of the time-averaged lift coefficient, drag coefficient and Strouhal number St for the cavitation number o- = 0.8, the angle of attack a= 8°at a Reynolds number Re = 7 x 10^5 are only 3.29%, 2.36% and 9.58%, respectively. It is observed that the cavitation shedding flow patterns are closely associated with the vortex structures identified by the Q- criterion method. The predicted cloud cavitation shedding flow shows clearly three typical stages：（1） Initiation of the attached sheet cavity, the growth toward the trailing edge. （2） The formation and development of the re-entrant jet flow. （3） Large scale cloud cavity sheds downstream. Numerical results also indicate that the non-uniform adverse pressure gradient is the main driving force of the re-entrant jet, which results in the U-shaped cavity and the 3-D bubbly structure during the cloud cavity shedding.

Effect of blade tip geometry on tip leakage vortex dynamics and cavitation pattern in axial-flow pump

A series of blade tip geometries, including original plain tip, rounded tip on the pressure side and diverging tip towards the suction side, were adopted to investigate the effect of blade geometry on tip leakage vortex dynamics and cavitation pattern in an axial-flow pump. On the basis of the computation, it clearly shows the flow structure in the clearance for different tip configurations by the detailed data of axial velocity and turbulent kinetic energy. The in-plain trajectory, in aspects of the angle between the blade suction side and vortex core and the initial point of tip leakage vortex, was presented using the maximum swirling strength method. The most striking feature is that the inception location of tip leakage vortex is delayed for chamfered tip due to the change of blade loading on suction side. Some significant non-dimensional parameters, such as pressure, swirling strength and turbulent kinetic energy, were used to depict the characteristics of tip vortex core. By the distribution of circumferential vorticity which dominates the vortical flows near the tip region, it is observed that the endwall detachment as the leakage flow meets the mainstream varies considerably for tested cases. The present study also indicates that the shear layer feeds the turbulence into tip leakage vortex core, but the way is different. For the chamfered tip, high turbulence level in vortex core is mainly from the tip clearance where large turbulent kinetic energy emerges, while it is almost from a layer extending from the suction side corner for rounded tip. At last, the visualized observations show that tip clearance cavitation is eliminated dramatically for rounded tip but more intensive for chamfered tip, which can be associated with the vortex structure in the clearance.

Materials based on group IVA elements for alloying-type sodium storage

There are five elements in group IVA of the periodic table, i.e., carbon(C), silicon(Si), germanium(Ge), tin(Sn) and lead(Pb),of which Si, Ge, and Sn can be used as alloying-type electrode materials for sodium-ion batteries. Pb is also capable of alloying with sodium, but it is generally ruled out as the cause of toxicity. In recent years, materials based on Si, Ge, and Sn have been intensively exploited as sodium anodes because of their abundant resource and large capacity with reasonable working ***, successful deployment of these anode materials needs to overcome kinetic and thermodynamic issues related to poor electrochemical activity, particle pulverization associated with large volume swelling, and formation of unstable solid-electrolyte interphase. A diversity of material strategies has been employed to address these difficulties, mainly leveraging on the knowledge recently advanced for lithium anodes. This review highlights such issues and provides valuable insights for possible solutions, which serves as a guide and inspiration for future material innovation for rechargeable batteries.

DAMAGE DETECTION STRATEGY FOR RETICULATED STRUCTURES BASED ON INCOMPLETE STRAIN MODE

Damage detection based on strain responses of vibration is highly attractive for monitoring long-span reticulated ***,there are a lot of structure members in reticulated structures and it is impossible to install strain sensors in each ***,how to locate and quantify damages with the incomplete mode shapes obtained from few strain sensors is a challenge topic.A new strategy,named incomplete strain mode damage detection（ISMDD） strategy,is proposed in this *** the strategy,the distribution of the strain sensors in the reticulated structures can be optimized through sensitive analysis on strain mode perturbation matrix,which can be obtained by perturbation *** assurance criterion（MAC） value is applied in damage location,and the members with relative large MAC values are defined as damage *** addition,damage index obtained by solving the perturbation equation is used for damage *** analysis on a long-span reticulated structure,including damage location and quantification for single-and multi-member damages,detection for different damage quantity,the effect analysis of sensor quantity,are performed to verify the effectiveness of the proposed ISMDD *** can be shown from the analysis that the ISMDD strategy is effective in damage location and quantification for both single-and multi-member *** the quantity of strain sensors has effect on damage location,but has no obvious influence on damage ***,the anti-noise pollution ability analysis of the ISMDD strategy is carried out,which shows that the ISMDD strategy has excellent anti-noise pollution ability for both single-and multi-damaged members.

Application of materials based on group VB elements in sodium-ion batteries:A review

Owing to their high redox activity, abundant resource, and low cost, compounds based on group VB elements（V, Nb, and Ta） are promising electrode materials for an array of energy storage devices. Moreover,their open structure benefits transportation and accommodation of alkali Na＋ions, rending them particularly appealing for cost-effective sodium-ion batteries（SIBs）. In the last decade, these materials have evoked intensive investigations in SIBs, serving as either cathode, anode, or both depending on their sodiation degree and redox potential. Exciting progress has been made, but significant gaps in our knowledge still remain. Their practical application in SIBs remains unclear and thus calls for further efforts to settle several technical challenges in terms of poor conductivity, unstable cycling, and inefficient charge storage. In this perspective, a brief overview is given of recent progress on how to solve these technical issues and envision their bright future in SIBs, which may serve as a source to inspire researches on relevant topics.

Cardiac-resident macrophages(CRMs)play important roles in homeostasis,cardiac function,and *** CRMs play critical roles in cardiac regeneration of neonatal mice,their roles are yet to be fully ***,this study aimed to investigate the dynamic changes of CRMs during cardiac ontogeny and analyze the phenotypic and functional properties of CRMs in the promotion of cardiac *** mouse cardiac ontogeny,four CRM subsets exist successively:CX3CR1+CCR2-Ly6C-MHCII-(MP1),CX3CR1lowCCR2lowLy6C-MHCII-(MP2),CX3CR1-CCR2+Ly6C+MHCII-(MP3),and CX3CR1+CCR2-Ly6C-MHCII+(MP4).MP1 cluster has different derivations(yolk sac,fetal liver,and bone marrow)and multiple functions *** and neonatal-derived-MP1 directly promoted cardiomyocyte proliferation through Jagged-1-Notch1 axis and significantly ameliorated cardiac injury following myocardial ***2/3 subsets could survive throughout ***4,the main population in adult mouse hearts,contributed to *** ontogeny,MP1 can convert into MP4 triggered by changes in the cellular redox *** findings delineate the evolutionary dynamics of CRMs under physiological conditions and found direct evidence that embryonic and neonatal-derived CRMs regulate cardiomyocyte *** findings also shed light on cardiac repair following injury.
