FACILE REGULATION OF GLUTARALDEHYDE-MODIFIED GRAPHENE OXIDE FOR PREPARING FREE-STANDING PAPERS AND NANOCOMPOSITE FILMS

Colloidal suspensions of glutaraldehyde （GA） crosslinked or grafted graphene oxide （GO） sheets were fabricated by simply tailoring the feed sequence. The different structures were confirmed by Fourier transform infrared spectra and X-ray diffraction. As demonstration of the utilities, the different colloidal suspensions were used to prepare free-standing papers by flow-directed filtration and poly（vinyl alcohol）（PVA）-based nanocomposite films by casting. Free-standing papers from GA crosslinked GO sheets exhibited better mechanical properties than unmodified GO paper, while nanocomposite films from GA grafted GO exhibit higher tensile strength and Young＇s modulus.

Controlling the probability density of quantum systems

Global effect of an RNA polymerase β-subunit mutation on gene expression in the radiation-resistant bacterium Deinococcus radiodurans

The β-subunit of RNA polymerase, which is involved in rifampin binding, is highly conserved among prokaryotes, and Rile mutants detected in many bacteria are the result of amino acid changes. Spontaneous rifampin resistance mutations resulting in amino acid replacement （L420R） and deletion （1258-66 9 bp deletion） have been previously isolated in the rpoB gene of Deinococcus radiodurans. In this study, a β-subunit mutation in D. radiodurans resulted in a unique effect on growth rate. We used DNA microarrays and biochemical assays to investigate how the Rif mutation in the β-subunit led to changes in growth rate via altered regulation of multiple genes. The expression of genes with predicted functions in metabolism, cellular processes and signaling, and information storage and processing were significantly altered in the 9 bp-deletion rpoB mutant. The consensus promoter sequence of up-regulated genes in the 9 bp-deletion rpoB mutant was identified as an AT-rich sequence. Greater levels of reactive oxygen species accumulated in the L420R and 9 bp-deletion rpoB mutants compared with wild type. These results provide insight into the molecular mechanism of how the β-subunit Rif mutation alters the regulation of multiple genes.

Superparamagnetic iron oxide（SPIO） nanoparticles were used more and more universally in biomedical research recently. In order to develop a suitable immuno-magnetic probe which can be applied in antigen- antibody based immunoassay, such as tumor marker or cardiovascular marker detection, SPIO capped by dextran was synthesized with chemical coprecipitation method. With sodium periodate method, hydroxyl groups of dextran were oxidated to aldehyde groups, thus conjugated with antibodies by covalent bonds. Western blot pattern illustrated that structure integrity of the most antibodies was kept well, harmful influence given by oxidation of conjugation was very finite. Both immune reactivity and antigenicity of antibodies in the SPIO based immunoprobes were retained effectively. This probe has a potentially application in SPIO based novel suspension array for its superparamagnetic property that can be separated under gradient magnetic field.
