Inhibition of histone deacetylase 2 mitigates profibrotic TGF-β1 responses in fibroblasts derived from Peyronie＇s plaque

Epigenetic modifications, such as histone acetylation/deacetylation, have been shown to play a role in the pathogenesis of fibrotic disease. Peyronie＇s disease （PD） is a localized fibrotic process of the tunica albuginea, which leads to penile deformity. This study was undertaken to determine the anti-fibrotic effect of small interfering RNA （siRNA）-mediated silencing of histone deacetylase 2 （HDAC2） in primary fibroblasts derived from human PD plaque. PD fibroblasts were pre-treated with HDAC2 siRNA and then stimulated with transforming growth factor-p1 （TGF-β1）. Protein was extracted from treated fibroblasts for Western blotting and the membranes were probed with antibody to phospho-Smad2/Smad2, phospho-Smad3/Smad3, smooth muscle α-actin and extracellular matrix proteins, including plasminogen activator inhibitor-β 1, fibronectin, collagen I and collagen IV. We also performed immunocytochemistry to detect the expression of extracellular matrix proteins and to examine the effect of HDAC2 siRNA on the TGF-β1-induced nuclear translocation of Smad2/3 in fibroblasts. Knockdown of HDAC2 in PD fibroblasts abrogated TGF-β1-induced extracellular matrix production by blocking TGF-β1-induced phosphorylation and nuclear translocation of Smad2 and Smad3, and by inhibiting TGF-β1-induced transdifferentiation of fibroblasts into myofibroblasts. Decoding the individual function of the HDAC isoforms by use of siRNA technology, preferably siRNA for HDAC2, may lead to the development of specific and safe epigenetic therapies for PD.

A modified method for simulating non-stationary multi-point earthquake ground motion

A spectral-representation-based algorithm is proposed to simulate non-stationary and stochastic processes with evolutionary power,according to a prescribed non-stationary cross-spectral density matrix. Non-stationary multi-point seismic ground motions at different locations on the ground surface are generated for use in engineering applications. First,a modified iterative procedure is used to generate uniformly modulated non-stationary ground motion time histories which are compatible with the prescribed power spectrum. Then,ground motion time histories are modeled as a non-stationary stochastic process with amplitude and frequency modulation. The characteristic frequency and damping ratio of the Clough-Penzien acceleration spectrum are considered as a function of time in order to study the frequency time variation. Finally,two numerical examples are presented to validate the efficiency of the proposed method,and the results show that this method can be effectively applied to the dynamic seismic analysis of long and large scale structures.

Revision of seismic design codes corresponding to building damagesin the “5.12” Wenchuan earthquake

A large number of buildings were seriously damaged or collapsed in the "5.12" Wenchuan earthquake. Based on field surveys and studies of damage to different types of buildings, seismic design codes have been updated. This paper briefly summarizes some of the major revisions that have been incorporated into the "Standard for classification of seismic protection of building constructions GB50223-2008" and "Code for Seismic Design of Buildings GB50011-2001." The definition of seismic fortification class for buildings has been revisited, and as a result, the seismic classifications for schools, hospitals and other buildings that hold large populations such as evacuation shelters and information centers have been upgraded in the GB50223-2008 Code. The main aspects of the revised GB50011-2001 code include: (a) modification of the seismic intensity specified for the Provinces of Sichuan, Shanxi and Gansu; (b) basic conceptual design for retaining walls and building foundations in mountainous areas; (c) regularity of building configuration; (d) integration of masonry structures and precast RC floors; (e) requirements for calculating and detailing stair shafts; and (f) limiting the use of single-bay RC frame structures. Some significant examples of damage in the epicenter areas are provided as a reference in the discussion on the consequences of collapse, the importance of duplicate structural systems, and the integration of RC and masonry structures.

Collision tumor of the rectum: A case report of metastatic gastric adenocarcinoma plus primary rectal adenocarcinoma

Collision tumors are thought to arise from the accidental meeting and interpenetration of two independent tumors. We report here a highly unusual case of a 61-year old man who had a unique tumor that was composed of a metastatic adenocarcinoma from the stomach to the rectum, which harbored a collision tumor of primary rectal adenocarcinoma. The clonalities of the two histologically distinct lesions of the rectal mass were confirmed by immunohistochemical and molecular analysis. Although histologic examination is the cornerstone in pathology, immunohistochemical and molecular analysis can provide evidence regarding whether tumors originate from the same clone or different clones. To the best of our knowledge, this is the fi rst reported case of such an occurrence.

Experimental and Numerical Investigation on Impact Performance of Carbon Reinforced Aluminum Laminates

It is known that fiber metal laminates （FML） as one of hybrid materials with thin metal sheets and fiber/epoxy layers have the characteristics of the excellent damage tolerance, fatigue and impact properties with a relatively low density. Therefore, the mechanical components using FML can contribute the enhanced safety level of the sound construction toward the whole body. In this study, the impact performance of carbon reinforced aluminum laminates （CARAL） is investigated by experiments and numerical simulations. Drop weight tests are carried out with the weight of 4.7 kg at the speed of 1 and 2 m/s, respectively. Dynamic non-linear transient analyses are also accomplished using a finite element analysis software, ABAQUS. The experiment results and numerical results are compared with impact load-time histories. Also, energy-time histories are applied to investigate the impact performance of CARAL.

A BOUNDARY INTEGRAL METHOD FOR COMPUTING ELASTIC MOMENT TENSORS FOR ELLIPSES AND ELLIPSOIDS

The concept of elastic moment tensor occurs in several interesting contexts, in particular in imaging small elastic inclusions and in asymptotic models of dilute elastic composites. In this paper, we compute the elastic moment tensors for ellipses and ellipsoids by using a systematic method based on layer potentials. Our computations reveal an underlying elegant relation between the elastic moment tensors and the single layer potential.

AIM: This case-control study investigated the effects of kimchi,soybean paste, fresh vegetables,nonfermented alliums, nonfermented seafood, nonfermented soybean foods, and the genetic polymorphisms of some metabolic enzymes on the risk of gastric cancer in koreans. METHODS: We studied 421 gastric cancer patients and 632 age- and sex-matched controls. Subjects completed a structured questionnaire regarding their food intake pattern. Polymorphisms of cytochrome P450 1A1 (CYP1A1), cytochrome P450 2E1 (CYP2E1), glutathione S-transferase mu 1 (GSTM1),glutathione S-transferase theta 1 (65777) and aldehyde dehydrogenase 2 (ALDH2) were investigated. RESULTS: A decreased risk of gastric cancer was noted among people with high consumption of nonfermented alliums and nonfermented seafood. On the other hand, consumption of kimchi, and soybean pastes was associated with increased risk of gastric cancer. Individuals with the CYP1A1 Ile/Val or Val/Val genotype showed a significantly increased risk for gastric cancer. Increased intake of kimchi or soybean pastes was a significant risk factor for the CYP1A1 lie/lie, the CYP2E1 c1/c1,the GSTM1 non-null, the GSTT1 non-null, or the ALDH2 *1/*1 *** addition, eating soybean pastes was associated with the increased risk of gastric cancer in individuals with the GSTM1 null type. Nonfermented alliums were significant in individuals with the CYP1A1 lie/lie, the CYP2E1 c1/c2 or c2/c2, the GSTT1 null, the GSTT1 non-null, or the ALDH2 *1/*2 or *2/*2 genotype,nonfermented seafood was those with the CYP1A1 lie/lie,the CYP2E1 c1/c1, the ALDH2 *1/*1 genotype or any type of GSTM1 or GSTT1. In homogeneity tests, the odds ratios of eating kimchi for gastric cancer according to the GSTM1 or 65777 genotype were not homogeneous. CONCLUSION: Kimchi, soybean pastes, and the CYP1A1 Ile/Val or Val/Val are risk factors,and nonfermented seafood and alliums are protective factors against gastric cancer in koreans. In this paper we present a systematic way of computing the polarization tensors, anisotropic as well as isotropic, based on the boundary integral method. We then use this method to compute the anisotropic polarization tensor for ellipses and ellipsoids. The computation reveals the pair of anisotropy and ellipses which produce the same polarization tensors.

Preparation of Highly Crystalline Graphitic Nanocarbon for the Electro-Oxidation of Methanol

Highly crystalline graphitic nanocarbons （GNC） have been prepared by the wet-air treatment of hydrothermally- derived graphitic porous carbon. The materials were characterized by scanning electron microscopy, X-ray diffraction, Raman spectroscopy, and electrochemical methods. The experimental results revealed that the treatment temperature has a significant effect on the morphology and degree of graphitic crystallinity. When GNC was treated at 450 ~C under a wet-air atmosphere, the product （GNC-450） consisted of aggregates of silkworm-shaped carbon nanoparticles with enhanced graphitic characteristics. GNC-450 was evaluated as a catalyst support in the electro-oxidation of methanol. The Pt/GNC-450 catalyst contained smaller Pt particles and had a higher electrochemically active surface area than a commercial carbon black-supported Pt catalyst. In the electro-oxidation of methanol, the Pt/GNC-450 catalyst showed the highest performance among the Pt catalysts examined in this study. The superior catalytic performance appears to be closely related to the enhanced graphitic characteristics with highly dispersed Pt nanoparticles on the graphitic layers, which have a positive effect on the electrochemical performance.
