Efficient degradation of lube oil by a mixed bacterial consortium

A laboratory study was performed to assess the biodegradation of lube oil in bio-reactor with 304# stainless steel as a biofilm carrier. Among 164 oil degrading bacterial cultures isolated from oil contaminated soil samples, Commaonas acidovorans Px1, Bacillus sp. Px2, Pseudomonas sp. Px3 were selected to prepare a mixed consortium for the study based on the efficiency of lube oil utilization. the percentage of oil degraded by the mixed bacterial consortium decreased slightly from 99% to 97.2% as the concentration of lube oil was increased from 2000 to 10,000 mg/L. the degradation of tDOC （total dissolved organic carbon） showed a similar tendency compared with lube oil removal, which indicated that the intermediates in degradation process hardly accumulated. selected mixed bacterial consortium showed their edge compared to activated sludge. scanning electron microscopy （sEM） photos showed that biofilms on stainless steel were robust and with a dimensional framework constructed by EPs （extracellular polymeric substances）, which could promote the biodegradation of hydrocarbons. the increase of biofilm followed first-order kinetics with rate of 0.216 μg glucose/（cm2·day） in logarithm phase. With analysis of Fourier transform infrared spectroscopy （Ft-IR） and gas chromatography-mass spectrometry （GC-Ms） combined with removal of lube oil and tDOC, mixed bacterial consortium could degrade benzene and its derivatives, aromatic ring organic matters with a percentage over 97%.

Micromagnetism simulation on effects of soft phase size on Nd2Fe14B/α-Fe nanocomposite magnet with soft phase imbedded in hard phase

In this study, micromagnetism simulation by using timte dltterence method is cameo out on the Ncl21-el415/a-Fe nanocomposite magnet with soft phase imbedded in hard phase. the effects of soft magnetic phase size （s） on the magnetic properties and magnetic reversal modes are systematically analyzed. As s increases from 1 nm to 48 nm, the remanence （Jr） increases, while the coercivity （Hci） decreases, leading to the result that the magnetic energy prod- uct [（BH）max] first increases slowly, and then decreases rapidly, peaking at s = 24 nm with the （BH）max of 72.9 MGOe （1 MGOe = 7.95775 kJ.m-3）. Besides, with the increase of s, the coercivity mechanism of the nanocomposite magnet changes from nucleation to pinning. Furthermore, by observing the magnetic moment evolution in demagnetization pro- cess, the magnetic reversal of the soft phase in the nanocomposite magnet can be divided into three modes with the increase of s： coherent rotation （s 〈 3 nm）, quasi-coherent rotation （3 nm≤s 〈 36 nm）, and the vortex-like rotation （s ≥36 nm）.

the COVID-19 pandemic caused by the severe acute respiratory syndrome coronavirus 2(sARs-CoV-2)has wreaked havoc around the globe,with no end in *** rapid emergence of viral mutants,marked by rapid transmission and effective immune evasion,has also posed unprecedented challenges for vaccine development,not least in its speed,mass production,and *** we report a versatile“plug-and-display”strategy for creating protein vaccines,including those against malaria parasites and sARs-CoV-2,through the combined use of the intrinsically disordered protein ligase spystapler and computationally designed viral-like *** resulting protein nanoparticles harboring multiple antigens induce potent neutralizing antibody responses in mice,substantially stronger than those induced by the corresponding free *** modular vaccine design enabled by spystapler furnishes us with a new weapon for combatting infectious diseases.

Autism spectrum disorder (AsD) is a neurodevelopmental disorder with considerable clinical and genetic *** this study,we identified all classes of genomic variants from whole-genome sequencing (WGs) dataset of 32 Chinese trios with AsD,including de novo mutations,inherited variants,copy number variants (CNVs) and genomic structural variants.A higher mutation rate (Poisson test,PtR regions (1.42×10^(-8)) was revealed in comparison with that of whole genome (1.05×10^(-8)).Using an integrated model,we identified 87 potentially risk genes (Ps harboring various rare deleterious variants,including CHD8 and NRXN2,implying that the disorders may be in favor to *** particular,frequent rare inherited mutations of several microcephaly-associated genes (AsPM,WDR62,and ZNF335)were found in *** chromosomal structure analyses,we found four de novo CNVs and one de novo chromosomal rearrangement event,including a de novo duplication of UBE3A-containing region at 15q11.2-q13.1,which causes Angelman syndrome and microcephaly,and a disrupted tNR due to de novo chromosomal translocation t (1;5) (q25.1;q33.2).taken together,our results suggest that abnormalities of centrosomal function and chromatin remodeling of the microcephaly-associated genes may be implicated in pathogenesis of *** of WGs as a new yet efficient technique to illustrate the full genetic spectrum in complex disorders,such as AsD,could provide novel insights into pathogenesis,diagnosis and treatment.

the search for Antimalarial Drugs and the Discovery of Artemisinin

With its novel chemical structure, artemisinin is an antimalarial component isolated from the traditional Chinese medicine qinghao(Artemisia annua L.). Nowadays, artemisinin and its derivatives are used compatibly with new synthesized chemical antimalarial compounds to create artemisinin-based combination therapies(ACts). these have become the first choice in treating malaria p.f. all over the world, providing an effective solution for the global challenge of curing drug-resistant malaria. Among the five ACts recommended by the WHO, two were initiated in China and are used as the first-line treatment of falciparum malaria in all malaria endemic areas. As the use of artemisinin-based compound drugs have made such significant contributions to rolling back malaria, regarded as one of the great achievements globally in public health of the early twenty-first century, tu Youyou, one of the most important researchers in the discovery of artemisinin, was made the first Nobel Prize laureate in Physiology or Medicine from the Chinese mainland. Artemisinin was discovered in a special social and cultural context through a combination of the exploration of traditional Chinese medical literature with the modern research approach of pharmaceutical sciences. this(Project 523) is a typical case of goal-oriented research leading to scientific advance, and the result of scientific research driven by the national needs.

目的:探讨蒽醌大黄酸、大黄素甲醚和二乙酰大黄酸对人胰腺导管腺癌的抗肿瘤作用及表观遗传学效应。方法:在生物信息学研究中,采用中位生存率分析来评估胰腺癌患者中位生存率与靶基因表达的相关性,并推断蒽醌改变靶基因表达对生存率的影响。这一步骤是通过Kaplan-Meier绘图仪(KMP)从在线数据库中检索177例胰腺癌患者的临床资料来完成的。最后,进行网络药理学和靶向预测分析,确定大黄活性成分和二乙酰大黄酸可能作用于抑制胰腺癌的潜在靶点。采用高、中、低剂量的每种药物(大黄酸/二乙酰大黄酸:80μM、40μM、20μM、大黄素甲醚:8μM、4μM和2μM)体外评价大黄酸、大黄素甲醚和二乙酰大黄酸的疗效。Mtt法测定人胰腺导管腺癌细胞株sW1990和Capan-2在24、48和72小时内的细胞存活率和毒性。用Rt-q PCR法检测sW1990中靶基因24和48小时的基因表达。靶基因是1)先前报道在胰腺癌中甲基化的肿瘤抑制基因:RAssF1A、CDKN2A、tUsC3、stK11、CDH13;2)癌基因:KRAs;3)泛素特异性蛋白酶:UsP21;4)调节性甲基化基因:DNMt1和tEt1。此外,根据Rt-q PCR结果,用甲基化特异性PCR(MsP)检测大黄酸治疗24小时和大黄素甲醚治疗48小时后抑癌基因甲基化和非甲基化的百分率。结果:中位生存预测显示KRAs基因表达的下降和tUsC3基因表达的增加与胰腺癌患者生存期的增加呈正相关。网络药理学分析阐明了从更广泛的角度分析,大黄及其有效成分和二乙酰大黄酸可通过催乳素信号途径、VEGF信号途径、HIF-1信号途径、Erb B信号途径等途径抑制胰腺癌。更重要的是,大黄酸可能通过p53信号途径靶向,因为该途径的富集倍数更大,类似地,二乙酰大黄酸可能通过过氧化物酶体增殖物激活受体(PPARs)信号途径起作用,而大黄素甲醚可能通过催乳素信号途径起作用。大黄酸和二乙酰大黄酸对sW1990和Capan-2细胞的增殖具有剂量和时间依赖性。大黄素甲醚以时间依赖的方式抑制这些细胞系的增殖。此外,3种蒽醌在sW1990中调节了目标基因的表达。大黄酸在24小时、二乙酰大黄酸48小时和大黄素甲醚48小时对基因表达调控结果相对较好。大黄酸降低了KRAs、UsP21和DNMt1的基因表达,增加了CDH13、stK11、RAssF1A和tUsC3的基因表达,但没有增加tEt1的基因表达。二乙酰大黄酸降低了KRAs、UsP21的基因表达,增加了RAssF1A和CDKN2A的基因表达,并略微增加了tEt1(低剂量)和stK11的基因表达,但没有增加CDH13的基因表达。大黄素甲醚降低KRAs、UsP21和DNMt1的基因表达,增加CDH13、RAssF1A、CDKN2A、tUsC3和tEt1的基因表达,但不增加stK11的基因表达。对于MsP,用大黄酸处理sW1990细胞24小时,使所有靶肿瘤抑制基因RAssF1A、stK11、CDKN2A、tUsC3和CDH13去甲基化,而大黄素甲醚处理细胞48小时,可以使除stK11以外的所有靶肿瘤抑制基因去甲基化。这些结果与Rt q PCR结果一致。中位生存预测显示RAssF1和tUsC3基因表达的增加与胰腺癌患者生存率的增加呈正相关。结论:***低表达和tUsC3高表达都与生存期延长有关。2.大黄和大黄素甲醚可能通过催乳素信号通路,而大黄酸和二乙酰大黄酸可能分别通过p53和PPAR信号通路作用于胰腺癌。3.大黄酸、大黄素甲醚和二乙酰大黄酸可以抑制sW1990和CAPAN-2的体外增殖,并调节靶基因RAssF1A、tUsC3、stK11、CDH13、CDKN2A、KRAs、UsP21、DNMt1和tEt1。大黄酸可使RAssF1A、tUsC3、stK11、CDH13、CDKN2A去甲基化,大黄素甲醚可使除stK11以外的上述肿瘤抑制基因去甲基化。由于大黄酸、大黄素甲醚和二乙酰大黄酸可能是治疗胰腺癌的有希望的药物,因此有必要进一步研究以确定其在临床上的疗效。

Xiayuxue Decoction(下瘀血汤)Attenuates Hepatic stellate Cell Activation and sinusoidal Endothelium Defenestration in CCI_4-Induced Fibrotic Liver of Mice

Objective： to investigate the effects of ancient Chinese medical formula Xiayuxue Decoction （下瘀血汤, XYXD） on activation of hepatic stellate cells （HsCs） and defenestration of sinusoidal endothelial cells （sECs） in CCI4-induced fibrotic liver of mice. Methods： High performance liquid chromatography was used to identify the main components of XYXD and control the quality of extraction. C57BL/6 mice were induced liver fibrosis by CCI4 exposure and administered with XYXD for 6 weeks simultaneously. Liver tissue was investigated by hematoxylin-eosin and sirius-red staining. sinusoidal fenestrations were observed by scanning electronic microscopy and fluorescent immunohistochemistry of PECAM-1 （CD31）. Whole liver lysates were detected of α-smooth muscle actin （α-sMA） and type-I collagen by Western blot. Primary rat HsCs-t6 cells were analyzed by detecting α-sMA, F-actin, DNA fragmentation through confocal microscopy, Western blot, terminal-deoxynucleoitidyl transferase mediated nick end labeling （tUNEL） assay and cellomics arrayscan, respectively. Results： Amygdalin and emodin in XYXD were identified. XYXD （993 mg/kg） inhibited sirius red positive area up to 70.1% （P〈0.01）, as well as protein levels of α-sMA and type- I collagen by 42.0% and 18.5% （P〈0.05） respectively. In vitro, XYXD （12.5 μg/mL, 50 μg/mL） suppressed the activation of HsCs and reversed the myofibroblastic HsCs into quiescent, demonstrated as inhibition of fluorescent F-actin by 32.3% and 46.6% （P〈0.05）. Besides, XYXD induced the apoptosis of HsC-t6 cells by 20.0% （P〈0.05） and 49.5% （P〈0.01）, evidenced by enhanced tUNEL positivity. Moreover, ultrastructural observation suggested XYXD inhibited defenestration of sECs, which was confirmed by 31.1% reduction of protein level of CD31 （P〈0.05）. Conclusions： XYXD inhibited both HsCs activation and sECs defenestration which accompany chronic liver injuries. these data may help to understand the underlying mechanisms of XYXD for prevetion of chronic liver diseases
