Phylogenetic Analysis and Molecular Identification of the Six Reticulitermes Species Using Different Markers

The tennite genus Reticulitermes species are serious pests of wooden structures and important quarantine insects in the world. It is very difficult to distinguish different Reticulitermes species based on their similar morphology, which brings much trouble to quarantine work for invasive Reticulitermes species. Here, we used the four molecular markers, 18 S rDNA, COI, COII and ITS genes, to carry out phylogenetic analysis and identification of the six Reticulitermes species. We found that R. labralis, R. speratus, R. aculabialis, R. chinensis and *** from china clustered together in the phylogenetic tree of COI gene, consisting with geographical distribution of them. We suggest that COI gene is suitable for interspecific phylogenetic analysis of Reticulitermes species. Subsequently, we designed and synthesized specific primer and TaqMan probe based on the COI gene of R. chinensis, and found that this primer can specifically detect R. chinensis. The minimum concentration of DNA sample is lpg/μL for the identification of R. chinensis. Our results suggest that the rapid and accurate identification of different Reticulitermes species can be achieved by fluorescent PCR technology based on COI gene.

利用一个已报道的DNA分子标记IDVGA-76作为牛特异性的遗传标记,通过聚合酶链式反应(PCR)鉴定牛DNA及制成品的牛基因组DNA.通过对目的片段的PCR扩增共检测了来自14个不同牛品种的59个样品和9个其他动物样品.所有供试牛样品均产生1条188 bp的片段,而对照动物样品的扩增结果为阴性,表明IDVGA-76可以作为牛特异性遗传标记用以检测食品和饲料中的牛源性成分.

本文提出了一种基于电流比较仪原理的高精度电流总加器,它包括6路输入绕组、反馈绕组和校验绕组,可有效校验工业电流总加测量系统。提出了最小公约数法,可方便选择各路输入绕组,有效测量总电流。实验表明,这种总加器的精度可优于5×10。

[Objectives] A simple and reliable HPLC fingerprint method was developed for the identification of dried barks of Ilex rotunda and I. godajam. [Methods] Nine batches of dried barks of I. rotunda,and seven batches of dried barks of I. godajam collected from different pharmacies and arboretums in different regions of china were used to establish fingerprints. The software Similarity Evaluation System of Chromatographic Fingerprints of Traditional Chinese Medicine( 2004 A Edition) was used to evaluate the fingerprints. [Results]The fingerprints of dried barks of I. rotunda and I. godajam were established. Methodological study met the technical requirements of fingerprints. The similarities of the fingerprints of dried barks of I. rotunda and I. godajam were all more than 0. 8 and 0. 9 respectively. There were 31 and 28 common peaks in I. rotunda and I. godajam,which could be classified into two clusters by principal component analysis( PCA) and hierarchical cluster analysis. [Conclusions] The feasibility and advantages of used HPLC fingerprints were verified,and the results indicated that the HPLC fingerprint as a characteristic distinguishing method combining similarity evaluation,principal component analysis and hierarchical cluster analysis can be successfully used to identify the authenticity of dried barks of I. rotunda and I. godajam.

Phytophthora is genus of plant-damaging Oomycetes, whose member species are capable of causing enormous economic losses on crops worldwide. In the present study, four candidate genes ITS, CO1, EF-1α and β-tubulin were tested using 123 strains of 80 species of Phytophthora to investigate the feasibility of ser- ving as DNA barcoding markers. The results showed that among the four candidate genes, ITS and CO1 had the highest success rate of PCR amplification and se- quencing, up to 100% and 96.7%. There were obvious barcode gaps in ITS, CO1 andβ-tubulin, but their frequency distributions of intra- and interspecific genetic distances were slightly overlapped. Wilcoxon rank sum test on intraspecific genetic distances of the four genes showed ITS = CO1 = β - tubulin = EF-1α indicating they bad the same effect on intraspecific discrimination, while the test on interspecific genetic distances of the four genes showed ITS 〉 C01 〉 β- tubulin 〉 EF - 1α. In summary, ITS and COl should be used in combination as the primary barcodes, β-tubulin as the complementary barcede for the identification of 11 quaran- tine Phytophthora species.

[Objective] The study aimed to discuss the application of boron-doped diamond （BDD） film electrode in fast detection of samonella in water. [ Method] Boron-doped diamond film electrode was prepared and used as the working electrode in fast detection of salmonella in water using chronoamberometry, and the oxidation mechanism of the electrode acting on salmonella was discussed. [ Result] Compared with traditional biologi- cal methods, chronoamperometry could detect the number of salmonellae in water more simply, rapidly and sensitively. [ Conclusion] The method of using BDD electrode to detect salmonella quantity will be widely applied in future.

Chemicals, due to their combustible, explosive, toxic characteristics and aptness to jeopardize the environment, human health and public safety, have long been on the top agenda of the governments throughout the world. At present china is a large country in the production, consumption and trading of chemicals with 45, 000 kinds already manufactured and in use and some 100 new kinds per year awaiting being registered to enter into the markets.1 Generally, the chemicals management in china has undergone through labor protection between 1950s and 1960s, pollution control and public safety during 1970s and 1980s, and ozone protection, climate change, recycling economy and anti-terrorism in recent decades. The focus of the legislation on chemicals has shifted from separate and scattered regulations in the form of departmental rules to national regulations which give special attention to the coordination of concerned departments and to the linkup and compatibility with relevant international treaties. In a word, chemicals management has become one of the sectors in china＇s environmental management domains where there are a large number of stringent regulations.
