Oral leukoplakia is the most common type of oral potentially malignant disorders and considered a precursor lesion to oral squamous cell ***,a predictor of oral leukoplakia prognosis has not yet been *** investigated whether copy number alteration patterns may effectively predict the prognostic outcomes of oral leukoplakia using routinely processed paraffin *** of copy number alteration patterns between oral leukoplakia with hyperplasia(HOL,n=22)and dysplasia(DOL,n=21)showed that oral leukoplakia with dysplasia had a higher copy number alteration rate(86%)than oral leukoplakia with hyperplasia(46%).Oral leukoplakia with dysplasia exhibited a wider range of genomic variations across all chromosomes compared with oral leukoplakia with *** also examined a retrospective cohort of 477 patients with oral leukoplakia with hyperplasia with detailed follow-up *** malignant transformation(MT,n=19)and leukoplakia recurrence(LR,n=253)groups had higher frequencies of aneuploidy events and copy number loss rate than the free of disease(FD,n=205)***,our results revealed the association between the degree of copy number alterations and the histological grade of oral leukoplakia and demonstrated that copy number alteration may be effective for prognosis prediction in oral leukoplakia patients with hyperplasia.

Salvianolic Acid B Inhibits the TLR4-NFκ B-TNFα Pathway and Attenuates Neonatal Rat Cardiomyocyte Injury Induced by Lipopolysaccharide

Objective： To investigate the role of the TLR4-NF K B-TNFa inflammation pathway on lipopolysaccharide （LPS）-induced neonatal rat cardiomyocyte injury and the possible protective effects of salvianolic acid B （Sal B）. Methods： Wistar rat （1-2 days old） cardiomyocytes were isolated and cultured. Sal B 10-5mol/L, 10-6mol/L and 10-7mol/L were pro-treated for 6 h in the culture medium. LPS （1 μg/mL） was added to the culture medium and kept for 6 h to induce inflammation injury. The concentration of lactate dehydrogenase （LDH） in the supernatant was detected by spectrophotometry. The concentrations of tumor necrosis factor （TNF a） and heat shock protein 70 （HSP70） in the supernatant were detected by enzyme linked immunosorbent assay. The protein expressions of toll, such as receptor 4 （TLR4） and nuclear factor kappa B （NF K B） were detected by immunohistochemistry. The mRNA expressions of TLR4 and NF K B were detected by realtime reverse transcription polymerase chain reaction （RT-PCR）. Results：（1） The concentrations of LDH and TNF a in the LPS control group were significantly higher than those in the control group （561.41 ± 67,39 U/L and 77.94± 15.08 pg/mL, versus 292.13± 26.02 U/L and 25.39 ±16.53 pg/mL, respectively, P〈0.01, P〈0.05）. Compared with the LPS control group, the concentrations of LDH and TNF α were significantly decreased in the Sal B 10-5mol/L pro-treated group （451.76 ± 83.96 U/L and 34.00± 10.38 pg/mL, respectively, P〈0.05）. （2） The TLR4 and NF K B protein expression area in the LPS control group were significantly higher than those in the control group （1712.41 ± 410.12 μm2 and 2378.15 ± 175.29 μm2, versus 418.62 ± 24.42 μ m2 and 1721.74 ± 202.87μ m2, respectively, P〈0.01）. The TLR4 and NF K B protein expression internal optical density （IOD） values in the LPS control group were also significantly higher than those in the control group （3.06 ±0.33 and 7.20± 1.04, versus 0.91 ±0.21 and 4.24±0.48, respectively, P〈0.05 and P〈0.01）. Trends of incidence rate and age at diagnosis for cervical cancer in China,from 2000 to 2014

Objective： To analyze the trends of incidence rate and age at diagnosis for cervical cancer incidence in China using population-based cancer registration data from 2000 to 2014. Methods： Data were from National Central Cancer Registry of China. Crude incidence rates （CIRs）, age-specific incidence rate, age-standardized incidence rates （ASIRs）, age percentage distribution, standardized age percentage distribution, mean age at diagnosis and standardized mean age at diagnosis for cervical cancer in all areas of China, urban China and rural China were calculated separately. The world Segi＇s population was applied to remove the age structure influence. Joinpoint regression was performed to obtain average annual percent change （AAPC） and age- period-cohort analysis was used to examine the incidence trends. Results： CIRs and ASIRs for cervical cancer increased in China from 2000 to 2014. The AAPC of ASIRs in China was at 9.2% [95% confidence interval （95% CI）： 7.0%-I 1.5%, P〈0.05], and the AAPC in rural areas was relatively high. The age-specific incidence rate in groups aged 0-69 years have significantly increased over time. Groups aged 40-69 years showed the highest incidence risk, and the annual percent changes （APCs） of incidence rate in groups aged 40-59 years in urban China and groups aged 0-49 years in rural China were more than 10%. For each age group, the urban-to-rural incidence rate ratios （IRRs） got close to 1 over time. There were clear birth cohort effects in successive generations born from 1940 to 1970 in China. In rural China, the standardized mean age at diagnosis had significantly declined by 5.18 years. In China, the main peak and secondary peak of standardized age percentages appeared in the groups aged 45-49 and 40-44 years, respectively. A novel missense mutation in obscurin gene in a chinese consanguineous family with left ventricular noncompaction

BACKGROUND Left ventricular noncompaction(LVNC) is an increasingly recognised cardiomyopathy of which a significant percentage are genetic in origin. The purpose of the present study was to identify potential pathogenic mutation leading to disease in a chinese LVNC *** A 3-generation family affected by LVNC was recruited. Clinical assessments were performed on available family members, with clinical examination, ECG, echocardiography and cardiac MRI. The proband(Ⅰ-2), the proband’s daughter(Ⅱ-1, affected) and mother(Ⅲ-1, unaffected) were selected for WGS. Sanger sequencing were performed in all of the 4 surviving family *** Combined whole genome sequencing with linkage analysis identified a novel missense mutation in the giant protein obscurin(OBSCN NM_001098623, c.C19063T), as the only plausible disease-causing variant that segregates with disease among the four surviving individuals, with interrogation of the entire genome excluding other potential causes. This c.C19063T missense mutation resulted in p.R6355W in the encoded OBSCN protein. It affected a highly conserved residue in the C terminus of the obscurin-B-like isoform between the PH and STKc domains, which was predicted to affect the function of the protein by different bioinformatics *** Here we present clinical and genetic evidence implicating the novel R6355W missense mutation in obscurin as the cause of familial LVNC. This expands the spectrum of obscurin’s roles in cardiomyopathies. It furthermore highlights that rare obscurin missense variants, currently often ignored or left uninterpreted, should be considered to be relevant for cardiomyopathies and can be identified by the approach presented here. This study also provided new insights into the molecular basis of OBSCN mutation positive LVNC.

Treatment of Lung Cancer with Orally Administered chinese Herbal Medicine: An Evidence Map between 1970–2020

Objective: Through showing the full picture of double-arm controlled clinical research and systematic review evidence in the field of orally administrated chinese herbal medicine(CHM) for treatment of lung cancer, to provide a reference for future clinical research and to indicate a direction for future systematic reviews. Methods: A comprehensive search of clinical controlled studies was performed regarding orally administered CHM treatment for lung cancer published from January 1970 to September 2020. The language was restricted to chinese and English. Relevant data were extracted, the quality of systematic reviews was evaluated, and the research evidence was visually displayed. Results: Randomized controlled trials were the most common type of research design. The research sample sizes were typically small. Oral CHM showed certain curative advantages in treating lung cancer. The key stages in oral CHM intervention for lung cancer are chemotherapy, radiotherapy, and late palliative treatment. The advantageous outcomes of oral CHM treatment of lung cancer are the short-term efficacy, quality of life, and adverse reactions. The perioperative stage, overall survival, pharmacoeconomic evaluation, and chinese medicine decoctions are weak research areas. Conclusions: CHM has staged and therapeutic advantages in treating lung cancer. The overall methodological quality is poor, and the level of evidence requires improvement. It is necessary to carry out large-scale, standardized, and higherquality research in the superior and weak areas of CHM treatment of lung cancer.

Effects and mechanism of chinese medicine Jiawei Yupingfeng in a mouse model of allergic rhinitis

Objective: chinese medicine has the potential to modulate allergic rhinitis(AR). There have been studies investigating the treatment efficacy of Yupingfeng San, alone or in combination with other ingredients, in AR, though few have studied the potential mechanisms of these drugs. In the present study, we measured the effects of Jiawei Yupingfeng(JWYPF), a traditional chinese medicine formula, on mice with ovalbumin-induced AR and explored its underlying mechanism of ***: Forty BALB/c mice were randomly divided into normal control, allergy control and two treatment groups of ten mice each. In the normal control group, mice were sensitized and challenged with saline. The mice in the allergy control and treatment groups were sensitized and challenged with ovalbumin and aluminum hydroxide gel. The treatments of JWYPF and Nasonex were administered intranasally in the AR mice for one week. Several signs of allergic inflammation, such as nasal eosinophils and inflammatory cytokines, were measured to determine the underlying ***: Mice in the JWYPF and Nasonex groups had significantly lower AR symptom scores than those in the allergy control group(the mean differences between JWYPF and the allergy control, and Nasonex and the allergy control were -2.00 ± 0.35 and -2.40 ± 0.32). After treatment with JWYPF and Nasonex, the levels of ovalbumin-specific Ig E and histamine were significantly reduced, as were the levels of interlukin-4 and transforming growth factor-b, while interferon-c levels were increased(all P < 0.0001,vs. allergy control). These two treatments also significantly inhibited eosinophil and mast cell infiltration into the nasal cavity but were not statistically different from ***: JWYPF has a potential therapeutic effect on AR via adjusting the rebalance of T helper 1 and T helper 2.

Controlled clinical trials of integrative therapies available to patients with rheumatoid arthritis （RA） improved dramatically in the past 20 years, largely because of the growing need and the methodologies improvement. Tripterygium wilfordii Hook. F., a typical example of popular use herb, has been extensively studied in trials. However, clear and convincing evidence of integrative therapy, effectiveness and safety, remains insufficient to make decision. Many research efforts are hampered by standing problems with ＇syndrome＇ recruitment failure. In addition, the outcome multiplicity induces the findings inefficiency to generalize to RA patients at large. Development of validated syndrome outcomes and methodologies has also been critical. Current efforts to enhance the understanding of integrative treatment options for patients with RA include the development of drug-specific rather than disease-specific strategies, studies in predictive biomarkers, and development of peer-review trial protocol for regular clinical trials.

A novel classification method for aid decision of traditional chinese patent medicines for stroke treatment

Traditional chinese patent medicines are widely used to treat stroke because it has good efficacy in the clinical environment. However, because of the lack of knowledge on traditional chinese patent medicines, many Western physicians, who are accountable for the majority of clinical prescriptions for such medicine, are confused with the use of traditional chinese patent medicines. Therefore, the aid-decision method is critical and necessary to help Western physicians rationally use traditional chinese patent medicines. In this paper, Manifold Ranking is employed to develop the aid-decision model of traditional chinese patent medicines for stroke treatment. First, 115 stroke patients from three hospitals are recruited in the cross-sectional survey. Simultaneously, traditional chinese physicians determine the traditional chinese patent medicines appropriate for each patient. Second, particular indicators are explored to characterize the population feature of traditional chinese patent medicines for stroke treatment. Moreover, these particular indicators can be easily obtained by Western physicians and are feasible for widespread clinical application in the future. Third, the aid-decision model of traditional chinese patent medicines for stroke treatment is constructed based on Manifold Ranking. Experimental results reveal that traditional chinese patent medicines can be differentiated. Moreover, the proposed model can obtain high accuracy of aid decision.
