Syntheses of amphiphilic biodegradable copolymers of poly(ethyl ethylene phosphate) and poly(3.hydroxybutyrate) for drug delivery

Biodegradable and amphiphilic triblock copolymers poly(ethyl ethylene phosphate)-poly(3.hydroxybutyrate)-poly(ethyl ethylene phosphate) (PEEP-b-PHB-b-PEEP) have been successfully synthesized through ring-opening polymerization. The structures are confirmed by gel permeation chromatography and NMR analyses. Crystallization investigated by X-ray diffraction reveals that the block copolymer with higher content of poly(ethyl ethylene phosphate) (PEEP) is more amorphous, showing decreased crystallizability. The obtained copolymers self-assemble into biodegradable nanoparticles with a coreshell micellar structure in aqueous solution, verified by the probe-based fluorescence measurements and transmission electronic microscopy (TEM) observation. The hydrophobic poly(3.hydroxybutyrate) (PHB) block serves as the core of the micelles and the micelles are stabilized by the hydrophilic PEEP block. The size and size distribution are related to the compositions of the copolymers. Paclitaxel (PTX) has been encapsulated into the micelles as a model drug and a sustained drug release from the micelles is observed. MTT assay also demonstrates that the block copolymers are biocompatible, rendering these copolymers attractive for drug delivery.

The acceleration of transferring the extensive economy to the intensive one in province (cities and districts) and the relation of coordinated growth of science and technology, economy, society are of momentous current significance.

Effects of Nitrogen Doping on Microstructures and Irradiation Resistance of Ti-Zr-Nb-V-Mo Refractory High-Entropy Alloy

Interstitial strengthening with nitrogen(N)is one of the effective ways to improve the mechanical properties of HEAs,but the effects of N on the microstructures and mechanical properties of the irradiated HEAs have not been studied ***,the microstructures and mechanical properties of N-free and N-doped Ti_(2)ZrNbV_(0.5)Mo_(0.2)HEAs before and after He irradiation were *** results showed that the solid solution strengthening caused by interstitial N improved the yield strength at room temperature and 1023.K without significantly reducing plasticity.N doping significantly promoted the growth,aggregation and wider spatial distribution of He bubbles by enhancing the mobility of He atoms/He-vacancy complexes,with the average size of He bubbles increasing from 10.4 nm in N-free HEA to 3..0 nm in N-doped *** addition,N-doped HEA showed a much higher irradiation hardness increment and hardening fraction than N-free *** to conventional materials doped with N,the introduction of N into Ti_(2)ZrNbV_(0.5)Mo_(0.2)HEA had adverse effects on its resistance to He bubble growth and irradiation *** results of this study indicated that N doping may not improve the irradiation resistance of HEAs.

cDNA cloning,sequence analysis,and recombinant expression of akitonin beta,a C-type lectin-like protein from Agkistrodon acutus

AIM: To clone the cDNA of a new member of snake venom C-type lectin-like proteins, to study its structure- function relationships and to achieve its recombinant production. METHODS: PCR primers were designed based on the homology and cDNA was amplified by RT-PCR using total RNA from snake venom gland as the template. The PCR products were cloned into the plasmid pGEM-T and sequenced. The deduced protein sequence was analyzed with some bioinformatic programs. A recombinant expression plasmid was constructed using pBAD- TOPO as vector and transformed into *** TOP10 competent cells. RESULTS: A novel cDNA sequence encod- ing akitonin β was found and accepted by GenBank (accession number AF3.7100). Akitonin β consists of a typical carbohydrate recognition domain (CRD) of C-type lectins, and it is homologous with other snake venom C-type lectin-like proteins. It was predicted to be a platelet antagonist. Upon induction with arabinose rAkitonin β express- ing in E coli was achieved at a high level (superior to 150 mg/L). The recombinant fusion protein exhibited inhibitory activities on rat platelet aggregation in vitro. CONCLUSION: A new member of snake venom C-type lectin-like proteins was discovered and characterized, and an efficient recombinant expression system was estab- lished for its production.

In the process of accelerating industrialization, urbanization is inevitable and important to chinas modernization drive. While people are cherishing the advantages of living in cities, the increasing negative impact of urbanization has to be put on the top of priority list. As urbanization proceeds, the existing pollution problem aggravates with not only increasing quantity of pollutants, but diversity as well. science and technology have been contributing to urbanization process in both positive and negative ways. Nonetheless, the role of science and technology in promoting sustainable urbanization is increasingly important. china has been conducting environmental research and development since the early 1970s, and now is capable of supplying most technologies needed for urban environmental protection. To keep pace with the rapid urbanization process in china, environmental research and technology development should be strengthened. This is mostly dependent on domestic resources with introduction of advanced yet cost\|effective technologies from the rest of the world. With a survey of current urban environmental R & D in china and on\|going activities of the Chinese Academy of sciences (CAS), recommendations on future UCL(University College London)\|CAS collaboration in the domain of urban environment are made.

Alterations in circadian rhythms of melaton in and cortisol in patients with bronchial asthma

AIM: To investigate the possible relationships between alterations in circadian rhythm of melatonin, cortisol andbronchial asthma. METHODS: Salivary melatonin and cortisol were measured simultaneously by radioimmunoas-say in 10 mild intermittent or persistent patients, 11 moderate-to-severe persistent asthma patients, and 15 controlsubjects. Twelve salivary samples were collected in a series during a 24-h period in each subject. RESULTS: Theresults showed overall lower levels of salivary melatonin in asthma patients compared with control subject(P5) and moderate-to-severe persistent asthma patients (P5, P<0.01) compared with control subject. CONCLUSION: Disordered circadian rhythms of salivary melato-nin and cortisol were found in asthma patients, which may be involved in the pathogenesis of bronchial asthma.

It has been revealed that poly I：C is a potent stimulator for NK cells, which can induce NK cell rapid activation and preferential accumulation into liver. However, the process mediating the influx of NK cells remains obscure. In this study, we found that poly I：C administration increased the portion and absolute number of NK cells in liver, but largely decreased those in spleen. There were no obvious changes of these lymphocytes in other immune organs. The results from splenic adoptive transfer and splenectomy showed that the recruited spleen NK cells contributed to the accumulation of NK cells in liver, and this process was regulated by the production of chemokines and the presence of T cells. This investigation will help to understand the enhanced immune cell recruitment in liver upon viral infection. Cellular ＆ Molecular Immunology. 2005;2（6）：449-453.

Antisense-Induced Blockade of GATA-3.Expression Could Inhibit Th2 Excursion of Tumor Cells in vitro and in vivo

Previous studies have shown that tumor cells predominantly express Th2 type cytokines and transcription factors. GATA-3. as a Th2-specific transcription factor, plays a central role in positive-regulating Th2 development. So whether the expression of GATA-3.in tumor cells has any effect on tumor development is a question of interest. In the present study, we inhibited the expression of GATA-3.in tumor cells through antisense RNA blockade technique, and observed its effects on tumor in vitro and in vivo. Our results showed that antisense GATA-3.treatment could inhibit the expression of TNF-α and Th2 cytokines in tumor cells, and antisense-induced blockade of GATA-3.could also depress tumor growth in tumor-bearing mice. We suggest that the ratio of T-bet/GATA-3.can be evaluated as a more important marker of the status of Thl/Th2 type. Anisotropic propagation of flare-induced shocks

It is demonstrated that the fastest propagation direction of a flare-induced shock deviates eastwards by about 4° at 1 AU relative to the flare normal if the shock introduced at 0.1 AU propagates along the local magnetic field, whereas it deviates westwards by about 1° if the introduced shock propagates in the radial direction instead.
