Aim:To determine whether interferon-α(IFNα) can enhance doxorubicinsensitivity in osteosarcoma cells and its molecular ***:Cellviability was evaluated using 3-(4,5-dimethylthiazol-2.yl)-2.5-diphenyltetrazoliumbromide *** was studied using Flow cytometry analysis,Hoechst332.8 staining,DNA fragmentation assay,as well as the activation ofcaspase-3 and poly (ADP-ribose) *** expression was detectedby Western *** dependence of p53 was determined using ***:IFNα increased doxorubicin-induced cytotoxicity to a muchgreater degree through apoptosis in human osteosarcoma p53-wild U2.S cells,but not p53-mutant MG63 ***α markedly upregulated p53,Bax,Mdm2.andp2.,downregulated Bcl-2.and activated caspase-3 and PARP cleavage in re-sponse to doxorubicin in U2.S ***,the siRNA-mediated silencing ofp53 significantly reduced the IFNα/doxorubicin combination-induced cytotoxic-ity and PARP ***:IFNα enhances the sensitivity of humanosteosarcoma U2.S cells to doxorubicin by p53-dependent *** propercombination with IFNα and conventional chemotherapeutic agents may be a ra-tional strategy for improving the treatment of osteosarcoma with functional p53.

Primary small cell carcinoma (SCC) of the breast, an exceedingly rare and aggressive tumor, is often characterized by rapid progression and poor prognosis. We report a case of primary SCC of the breast that was diagnosed through pathologic and immunohistochemical examinations. Computed tomography (CT) scans failed to reveal a non-mammary primary site. Due to the scant number of relevant case summaries, this type of tumor is proved to be a diagnostic and therapeutic challenge. Therefore, we also reviewed relevant literature to share expertise in diagnosis, clinicopathologic characteristics, treatment, and prognosis of this type of tumor. Future studies with more cases are required to define more appropriate treatment indications for this disease.

Obstructive Sleep Apnea-Hypopnea Ssyndrome and Hearing in Children

Objective To explore the relationship between hypoxemia and hearing in children with obstructive sleep apnea-hypopnea syndrome. Methods Auditory brainstem responses (ABRs) were recorded in 68 ears and distortion product otoacoustic emissions (DPOAEs) in 60 ears in children with OSAHS and type "A" tym-panograms, and in 30 ears in normal children. Results ABR latencies of waves I, III and V, and I-III, III-V and I-V intervals were not statistically different between OSAHS and normal children. Wave I latency was delayed in children with OSAHS compared to normal children3 (P < 0.05). DPOAE amplitudes in children with mild OSAHS were lower than normal children at 8 kHz (P < 0.05). DPOAEs were lower at 6 kHz and 8 kHz in children with moderate/severe OSAHS than normal children (P < 0.05). Conclusion Cochlear function was affected when AHI was at or greater than 10/hour. ABR and DPOAE can be used to detect early changes in auditory function in children with OSAHS.

AIM:To investigate the prognostic value of KRAS mutation,and phosphatase and tensin (PTEN) expression in Chinese metastatic colorectal cancer metastatic colorectal cancer (mCRC) patients treated with ***:Ninety Chinese mCRC patients treated with cetuximab were evaluated for KRAS mutation and PTEN protein expression by DNA sequencing of codons 12.and 13 and immunohistochemistry,*** then selected 61 patients treated with cetuximab,either in combination with chemotherapy,or alone as a second-line or third-line regimen to assess whether KRAS mutation or PTEN protein expression is associated with the response and the survival time of mCRC patients treated with ***:KRAS mutation was found in 30 (33.3%) tumor samples from the 90 patients,and positive PTEN expression was detected in 58 (64.4%) of the 90 *** the 61 patients who were treated with cetuximab as a second-line or third-line regimen,the resistance to cetuximab was found in 2. patients with KRAS mutation and in 39 patients without KRAS mutation,with a response rate of 4.5% and 46.1% respectively (P=0.001),a shorter median progression-free survival (PFS) time of 14 ± 1.3 wk and 32.± 2.5 wk respectively (P 2.mo and 19 ± 1.8 mo respectively (P 2. patients with negative PTEN expression and in 37 patients with positive PTEN expression respectively (P 4.2. and 48.6% respectively,a shorter median PFS survival time of 17 ± 2.0 wk and 2. ± 1.9 wk respectively (P=0.07),and a median OS time of 11 ± 1.3 mo and 18 ± 1.9 mo respectively (P=0.004).Combined KRAS mutation and PTEN expression analysis showed that the PFS and OS time of patients with two favorable prognostic factors were longer than those of patients with one favorable prognostic factor or no favorable prognostic factor (P < 0.001).CONCLUSION:KRAS mutation and PTEN protein expression are significantly corr

AIM: To explore the biological behaviors of hepatic oval cells after transfused into the circulation of experimental ***: Oval cells from male SD rat were transfused into the circulation of a female rat which were treated by a 2.AAF/CCl4 program, through caudal vein. Sex-determining gene sry which located on Y chromosome was examined by PCR and in situ hybridization technique in liver, kidney and spleen of the experimental animals, ***: The results of the cell-transplant experiment showed that the srygene was detectable only in the liver but not in spleen and kidney of the experimental rats, and no signals could be detected in the control animals. It can be also morphologically proved that some exogenous cells had migrated into the parenchyma of the liver and settled ***: The result means that there are exogenous cells located in the liver of the experimental animal and the localization is specific to the liver. This indicates that some 'signal molecules' must exist in the circulation of the rats treated by 2.AAF/CCl4. These 'signal molecules' might play an important role in specific localization and differentiation of transfused oval cells.

Energy metabolism and survival of liver grafts from non-heart-beating donor rats with warm ischemia injury

BACKGROUND: The shortage of donor livers is a critical limiting factor for the use of liver transplantation in treatment of end-stage liver diseases. Organs from non- heart-beating donors seem to be an effective option to alleviate this problem. Warm ischemia injury, however, directly influences the grafts' activity and functional recovery after operation. We investigated the energy metabolism and post-transplant survival of liver grafts after different warm ischemia times (WITs) in rats and determined the maximum limit for liver grafts with warm ischemia. METHODS: Rats were randomized into 7 groups with WITs of 0 (control), 10, 15, 2., 30, 45 or 60 minutes. The indices of energy metabolism were measured by reversed- phase high performance liquid chromatograpy and all liver graft specimens were subjected to ultrastructural observation. After orthotopic liver transplantation (OLT), the recovery of energy metabolism in liver grafts after 2. and 48 hours and the survival of the rats were assessed. RESULTS: The levels of adenosine triphosphate (ATP) and energy charge (EC) decreased gradually after different WITs in a time-dependent manner, and this was especially significant within 30 minutes. The levels of ATP and EC in liver grafts with 30 minutes of warm ischemia largely recovered 2. hours after OLT, with 45 minutes of warm ischemia partially recovered 48 hours after OLT, and with 60 minutes of warm ischemia, hardly recovered even 48 hours after OLT. The survival time after OLT did not significantly change with up to 30 minutes of WIT, while long-term survival was reduced with 45 and 60 minutes of ***: The levels of ATP and EC after OLT may be important criteria for evaluating the quality of a liver graft. The WIT of a liver graft is closely related to the recovery of hepatic energy metabolism and the graft survival.
