The role of cholesterol metabolism in lung cancer

Elevated serum cholesterol metabolism is associated with a reduced risk of lung *** cholesterol metabolism is evident in both lung cancer patients and tumor *** tumor cell cholesterol uptake or biosynthesis pathways,through the modulation of receptors and enzymes such as liver X receptor and sterolregulatory element binding protein 2,effectively restrains lung tumor ***,promoting cholesterol excretion yields comparable *** metabolites,including oxysterols and isoprenoids,play a crucial role in regulating cholesterol metabolism within tumor cells,consequently impacting cancer *** lung cancer patients,both the cholesterol levels in the tumor microenvironment and within tumor cells significantly influence cell growth,proliferation,and *** effects of cholesterol metabolism are further mediated by the reprogramming of immune cells such as T cells,B cells,macrophages,myeloid-derived suppressor cells,among *** research is investigating drugs targeting cholesterol metabolism for clinical ***,targeting the cholesterol biosynthesis pathway,are widely employed in lung cancer treatment,either as standalone agents or in combination with other ***,drugs focusing on cholesterol transportation have shown promise as effective therapies for lung *** this review,we summarized current research regarding the rule of cholesterol metabolism and therapeutic advances in lung cancer.

Dopamine can be used to treat depression,myocardial infarction,and other ***,few reports are available on the de novo microbial synthesis of dopamine from low-cost *** this study,integrated omics technology was used to explore the dopamine metabolism of a novel marine multi-stress-tolerant aromatic yeast Meyerozyma guilliermondii ***6 was found to have the ability to biosynthesize dopamine when using glucose as the substrate.14.key genes for the biosynthesis of dopamine were identified by whole genome-wide *** and proteomic data showed that the expression levels of gene AAT2 encoding aspartate aminotransferase(regulating dopamine anabolism)were upregulated,while gene AO-I encoding copper amine oxidase(involved in dopamine catabolism)were downregulated under 10%NaCl stress compared with non-NaCl stress,thereby contributing to biosynthesis of ***,the amount of dopamine under 10%NaCl stress was 2.51-fold higher than that of zero NaCl,which was consistent with the multi-omics ***-time fluorescence quantitative PCR(RT-qPCR)and high-performance liquid chromatography(HPLC)results confirmed the metabolic model of ***,by overexpressing AAT2,AST enzyme activity was increased by 24.89%,the expression of genes related to dopamine metabolism was enhanced,and dopamine production was increased by 56.36%in recombinant *** conclusion,Meyerozyma guilliermondii GXDK6 could utilize low-cost carbon source to synthesize dopamine,and NaCl stress promoted the biosynthesis of dopamine.

Heart valve diseases threaten human health. One reliable way to save lives of such patients is to replace the pathologically changed heart valves by artificial ones. Over 2 million patients have received LTI-carbon heart valve’s implantation. However, the thrombosis after the implantation is one of the difficulties that need to be solved. In order to improve the blood compatibility and security of mechanical heart valves, ion beam technology was used to modify the surface properties of the materials. The investigation results have been summarized in this paper.

Aim:Extrapyramidal syndrome (EPS) is most commonly affected by typical antip-sychotic drugs that have a high affinity with the D2 ***,manyresearch groups have reported on the positive relationship between the geneticvariations in the DRD2 gene and the therapeutic response in schizophrenia pa-tients as a result of the role of variations in the receptor in modulating *** this study,we evaluate the role DRD2 plays in chlorpromazine-induced EPS in schizophrenic ***:We identified seven SNP(singlenucleotide polymorphism) (-14.Cins>del,TaqIB,TaqID,Ser311Cys,rs6275,rs6277and TaqIA) in the DRD2 gene in 14. schizophrenic inpatients (59 with EPS and 87without EPS according to the Simpson-Angus Scale) treated with chlorpromazineafter 8 *** alleles of all loci were determined by PCR (polymerase chainreaction).Results:Polymorphisms TaqlD,Ser311Cys and rs6277 were not poly-morphic in the population recruited in the present *** statistical signifi-cance was found in the allele distribution of-14.Cins>del,TaqIB,rs6275 andTaqIA or in the estimated haplotypes (constituted by TaqIB,rs6275 and TaqIA) inlinkage disequilibrium between the two ***:Our results did notlend strong support to the view that the genetic variation of the DRD2 gene playsa major role in the individually variable adverse effect induced by chlorpromazine,at least in Chinese patients with *** results confirmed a previousstudy on the relationship between DRD2 and EPS in Caucasians.

Comparative studies of D_2 receptors and brain perfusion inhemi-parkinsonism rats

The relationship between dopamine D2 receptors and brain perfusion in hemi-parkinsonism rats was studied. Hemi-parkinsonism rats were made by stereotaxic 6-hydroxy dopamine (6-OH-DA) lesions in substantia nigra(SN) and ventral tegmental area(VTA), apomorphine (Apo) which could induced the successful model rat rotates toward the intact side was used to select the rats, 125I-IBZM ex-vivo autoradiography analysis and 99mTc-HM-PAO regional cerebral biodistribution were used to evaluate D2 receptors and cerebral blood flow. The HPLC-ECD were used to measure striatum DA and its metabolites content. The lesioned side striatum DA and its metabolites HVA DOPAC reduced significantly than that of the intact side and seudo-operated group, striatum/cerebellum 125I-IBZM uptake ratio was 8.04.-0.71 in lesioned side of hemi-parkinsonism rats, significantly increased compared with the intact side and the seudo-operated group(p 30.11+-4.53% enhancement as compared to the intact side, and also show good correlation with 30 min Apo induced rotation numbers (r=0.98), the regional cerebral blood flow study didn’t show significant difference between bilateral brain cortex area(p >0.05). These results indicated that in the 6- OH-DA lesioned side DA content decreased significantly and an up-regulation of striatum D2 receptor binding sites was induced in hemi-parkinsonism rats, which showed good correlation with rotation behavior induced by Apo. Comparing with cerebral blood flow, D2 receptor reflected by IBZM seems to be more specific and earlier to detect the cerebral functional impairment in experimental bend-parkinsonism.

AIM: To investigate the presence of glutamate transporters and receptors in mouse testis. METHODS: Glutamate uptake analysis was performed to study the function of glutamate transporters in mouse testis. Comparative RT-PCR technique and sequencing analysis were used to study the expression of glutamate receptors and transport- ers in mouse testis. RESULTS: Mouse testis possessed glutamate uptake capacity with sodium-dependence. Vmax value of glutamate uptake was (1.60 ± 0.21) pmol/min per mg protein and Km value of glutamate uptake was (11.0±1.6) μmol/L in mouse testis according to saturation analysis. Furthermore, the uptake activity could be inhibited by DHK (GLT1 selective inhibitor) and THA (glutamate uptake inhibitor). In addition, RT-PCR results revealed that glutamate transporters (GLT1 and EAAC1) and ionotropic glutamate receptors (NR1, NR2B, GluR6 and KA2) were expressed in mouse testis. CONCLUSION: Glutamate transporters and receptors do exist in mouse testis.

Audiofrequency electrotherapy(AFET)isused in 14. intestinal adhesion(IA)cases re-gardless of patient’s age,duration of illness,severity of symptoms,number of operations andrecurrences and failure of other methods oftreatment with 72.8% cures,18.6% marked im-provement(over 50% improvement)and only8.6% *** up observations on 4. casesshowed that 29 remained well,7 had occasionalmild attacks of abdominal discomfort and/or painwhich did not interfere with work and only 4.ad *** treatment relieved signs,symptoms and complications including pelvicinflammation,peritonitis,intestinal obstruction,abdominal masses,lumbar pain,diarrhea,noe-turia,general anasarca and obstipation and is thetreatment of choice in this hospital.

Circadian expression of clock genes and angiotensin Ⅱ type 1 receptors in suprachiasmatic nuclei of sinoaortic-denervated rats

Aim:To investigate whether the circadian expression of central clock genes andangiotensin Ⅱ type 1 (AT1) receptors was altered in sinoaortic-denervated (SAD)***:Male Sprague-Dawley rats underwent sinoaortic denervation or asham operation at the age of 12 *** weeks after the operation,bloodpressure and heart period were measured in the conscious state in a group ofsham-operated (n=10) and SAD rats (n=9).Rest SAD and sham-operated ratswere divided into 6 groups (n=6 in each group).The suprachiasmatic nuclei (SCN)tissues were taken every 4.h throughout the day from each group for the determi-nation of the mRNA expression of clock genes (Per2 and Bmal1) and the AT1receptor by RT-PCR;the protein expression of Per2 and Bmal1 was determined byWestern ***:Blood pressure levels in the SAD rats were similar tothose of the sham-operated ***,blood pressure variabilities signifi-cantly increased in the SAD rats compared with the sham-operated *** variation of clock genes in the SCN of the sham-operated rats was char-acterized by a marked increase in the mRNA and protein expression during ***2 and Bmal1 mRNA levels were significantly lower in the SAD rats,especially during dark *** blot analysis confirmed an attenuation ofthe circadian rhythm of the 2 clock proteins in the SCN of the SAD ***1receptor mRNA expressions in the SCN were abnormally upregulated in the lightphase,changed to a 12-h cycle in the SAD ***:The circadian varia-tion of the 2 central clock genes was attenuated in the SAD *** baroreflexdysfunction also induced a disturbance in the expression of AT1 receptors in theSCN.

Identification of genes responsive to apoptosis in HL-60 cells

AIM: To identify genes responsive to apoptosis in HL-60 cells treated by homoharringtonine. METHODS: cDNA microarray technology was used to detect gene expression and the result of microarrays for genes (TIEG and VDUP1) was confirmed by Northern analysis. RESULTS: Seventy-five individual mRNAs whose mass changed significantly were identified. Among these genes (25 were up-regulated and 50 were down-regulated), most are known related to oncogenes and tumor suppressor. Some genes were involved in apoptosis signaling pathways. CONCLUSION: TGFβ and TNF apoptosis signaling pathways were initiated during apoptosis in HL-60 cells. TIEG and VDUP1 play important roles in mediating apoptosis.
