AIM: To Study the interaction of macrophage colony-stimulating factor (M-CSF) and interleukin-10 (IL-10) in productions of IL-12 and IL-18 and expressions of CD14, CD23, and CD64 by human monocytes. METHODS: Purified adherent human monocytes were cultured with M-CSF or IL-10 alone, or with M-CSF+IL-10 and 2-3d later, the culture supernatants and cells were separated and collected. IL-12P40 and IL-18 levels in the superna-tants were determined by ELISA and the percentages of CD14, CD23, and CD64 positive cells were examined by flow cytometry. RESULTS: (1) IL-10 decreased M-CSF-induced IL-18 levels, while M-CSF further reduced IL-12P40 level in the culture supernatants of IL-10-treated monocytes; (2) IL-10 alone had no effect on the percentage of CD14-positive cells, but further increased the percentage of CD14-positive cells induced by M-CSF; M-CSF alone had no effect on the percentage of CD64-positive cells, but further increased the percentage of CD64-positive cells induced by IL-10; (3) IL-10 decreased

AIM:To assess the efficiency and toxicities of irinotecan (CPT-11)-involved regimens in patients with advanced gastric ***:Randomized phases Ⅱ and Ⅲ clinical trials on chemotherapy for advanced gastric cancer were searched from MEDLINE,EMbase,Cochrane Controlled Trials Register,and *** abstracts were manually searched.A total of 657 patients were analyzed for their overall response rate (ORR),time to treatment failure (TTF),overall survival (OS) rate,and *** survival rate,reported as hazard ratio (HR) with 95% CI,was used as the primary outcome ***:Four randomized controlled trials on chemotherapy for advanced gastric cancer were *** CPT-11-containing combination chemotherapy was not significantly advantageous over the non CPT-11containing combination chemotherapy for OS rate (HR =1.12,95% CI:0.92-1.36,P=0.266) and ORR [risk ratio (RR)=1.23,95% CI:0.71-2.14,P=0.458].However,the CPT-11-containing combination chemotherapy was significantly advantageous over the non CPT-11-containing combination chemotherapy for TTF (HR=1.35,95% CI:1.12-1.64,P=0.002).Grade 3/4 haematological toxicity (thrombocytopenia:RR=0.20,95% CI:0.09-0.48;P 3,P < 0.001) were lower in patients with advanced gastric cancer after CPT11-containing combination chemotherapy than after non CPT-11-containing combination ***:CPT-11-containing combination chemotherapy is advantageous over non CPT-11-containing combination chemotherapy for TTF with no significant ***-11-containing combination chemotherapy can be used in treatment of advanced gastric cancer.

AIM: To kill CEA positive colorectal carcinoma cells specifically using the E coli cytosine deaminase (CD) suicide gene, a new replication-deficient recombinant adenoviral vector was constructed in which CD gene was controlled under CEA promoter and its in vitro cytotoxic effects were evaluated. METHODS: Shuttle plasmid containing CD gene and regulatory sequence of the CEA gene was constructed and recombined with the right arm of adenovirus genome DNA in 293 cell strain. Dot blotting and PCR were used to identify positive plaques. The purification of adenovirus was performed with ultra-concentration in CsCl step gradients and the titration was measured with plaque formation assay. Cytotoxic effects were assayed with MTT method, The fifty percent inhibition concentration (IC(50)) of 5-FC was calculated using a curve-fitting parameter. The human colorectal carcinoma cell line, which was CEA-producing, and the CEA-nonproducing Hela cell line were applied in cytological tests. An established recombinant adenovirus vector AdCMVCD, in which the CD gene was controlled under CMV promoter, was used as virus control. Quantitative results were expressed as the mean +/- SD of the mean. Statistical analysis was performed using ANOVA test. RESULTS: The desired recombinant adenovirus vector was named AdCEACD. The results of dot blotting and PCR showed that the recombinant adenovirus contained CEA promoter and CD gene. Virus titer was about 5.0 X 10(14)pfu/L(-1) after purification. The CEA-producing Lovo cells were sensitive to 5-FC and had the same cytotoxic effect after infection with AdCEACD and AdCMVCD (The IC(50) values of 5-FC in parent Lovo cells, Lovo cells infected with 100 M.O.I AdCEACD and Lovo cells infected with 10 M.O.I AdCMVCD were >15000, 216.5+/-38.1 and 128.8+/-25.4 micromol.L(-1), P<0.001, respectively), and the cytotoxicity of 5-FC increased accordingly when the m.o.i of adenoviruses were enhanced (The value of IC(50) of 5-FC was reduced to 27.9+/-4.2 micr

We report observations in a 7 year old girl with manifest right accessory pathway (AP) and depressed cardiac function. Her Echocardiogram showed LV dilatation with asynchronous ventricular wall motion and LVEF reduction. The electrophysiological study demonstrated a para-his AP. She underwent successfully ablation and without any complications. The Echo validated that asynchronous ventricular wall motion disappeared after ablation. Her cardiac size and function was normal after follow up of three month. Our findings suggested that manifest AP leading to asynchronous ventricular motion was a possible cause of left ventricular dilation and dysfunction. Catheter ablation could reverse cardiac remodeling in such patient. Case report A seven year old girl was referred to our hospital for fainting episode in last six month. To exclude cephalic syncope, she underwent electroencephalogram, the result was negative. The electrocardiogram (ECG) showed right AP. An apparent delta wave looked like LBBB pattern with short PR interval (80ms) and long QRS duration (180ms) (figure 1 left panel). Echocardiogram (Echo) revealed left ventricle dilatation and left ventricular ejection fraction reduction (LVDd=52mm, LVEF=46%). Asynchronous ventriclular wall motion and moderate mitral valve regurgitation also observed on Tissue Doppler Imaging (TDI). She underwent electrophysiology study (EPS) and ablation procedure on the 5th hospital day. The intracardiac electrogram showed a para-His AP with antegrade conduction. The orthodromic SVT (201 bpm) was induce right ventricular apex programmed stimulation. During SVT episode, her symptom was similar to spontaneous one. After successful ablation, delta wave on ECG disappeared and SVT can not be induced with or without isoproterenol. Normal atrial-ventricular conduction was kept without any complications. AIM: To study the relationship between genotype of CYP2D6* 10B and pharmacokinetics of propafenone enantiomers. METHODS: Genotype of 17 healthy Chinese HAN subjects was determined by an allele specific amplification method. The blood samples (0-15 h) of the subjects were taken after oral administration of a single dose (400 mg) of propafenone hydrochloride. Concentrations of propafenone enantiomers in plasma were measured by a reverse-phase HPLC with precolumn derivatization. RESULTS: Seventeen subjects characterized for CYP2D6* 10B genotype included (*1/*1) (n=4), (*1/*10) (n=5) and (*10/*10) (n=8). The metabolic ratios (lg MR) of the three genotypes were-2.68±0.23, -2.2±0.7, and -1.1±0.5, respectively. The AUC of the three groups were (1534±334), (1891±793), (3171±1075) μg·h·L-1for S-enantiomer and (1136±345), (1467±817), (2277±745) μg·h·L-1for R-enantiomer, respectively. Hypertrophic cardiomyopathy （HCM） is one of the diseases damaging people health most badly and some mutations of exons in cardiac troponin I （cTnI） gene are closely associated with family hypertrophic cardiomyopathy （FHCM）.A microarray was fabricated to screen mutations in exons 3,5,7,and 8 in cTnI *** were designed for the PCR （polymerase chain reaction） to amplify the target DNA fragments from fresh blood *** order to simplify the PCR process,multiplex PCR technology was investigated in *** concentration of Mg2+played an important role in multiplex PCR process,a properly low concentration of Mg2+submitted a better speciality of PCR *** speciality was also favored when the annealing temperature was reasonably enhanced and 64℃is the optimal annealing temperature for the multiplex PCR *** applying the fabricated gene-chip to detect the target fragments from PCR mixture,the signal intensity sequence is in accordance with that from theoretic estimate.

AIM:To investigate the association between hepatocel-lular carcinoma (HCC) susceptibility and a 12-bp inser-tion/deletion polymorphism (rs6147150) in the 3'UTR of ***:Using a case-control design,the rs6147150 genotypes in 270 patients with HCC and 270 healthy controls were determined by direct polymerase chain reaction and polyacrylamide gel *** regression was used to analyze the association between the polymorphism and cancer ***:Computational modeling suggested that rs6147150 was located in the seed region of hsa-let-7c,a potential target sequence in ErbB4 3'*** re-gression analysis showed that,compared with individu-als homozygous for wild-type,heterozygotes [adjusted odds ratio (OR)=1.48,95% confidence interval (CI)= 1.03-2.17,P=0.034] and individuals homozygous for 12-bp del/del (OR=2.50,95% CI=1.37-4.56,P=0.001) were at significantly higher risk of ***-riers of the "del" allele of rs6147150 had a 1.59-fold increased risk for HCC (95% CI=1.22-2.07,P=0.003).CONCLUSION:rs6147150 may be associated with HCC risk,in part through let-7c-mediated regulation,and may be involved in the pathogenesis of HCC in Chi-nese populations.

BACKGROUND: Heat shock protein (HSP) gp96 is a member of the HSP90 family and presumably overexpresses as a result of stimulation by mutated or abnormal proteins. Its abnormal expression correlates with carcinogenesis, progression and prognosis of hepatocellular carcinoma (HCC). In this study, we investigated the pathological characteristics of liver gp96 expression and its relationship with hepatitis B virus (HBV) replication in HCC patients. METHODS: Tumor specimens were prospectively collected from 30 HCC patients undergoing liver resection. Total RNAs were extracted from HCC or their noncancerous tissues. The distribution of gp96 expression in hepatocytes was investigated by streptavidin peroxidase (S-P) immunohistochemistry and tissue HBV-DNA was detected by the in situ molecular hybridization technique. The association of gp96 expression with HBV replication, and the histopathological characteristics of HCC were analyzed. RESULTS: The gp96 was strongly expressed in HCC (73.3%, 22 of 30) and weakly (46.7%, 14 of 30) in non-cancerous tissues. The gp96 expression in HCC tissues was correlated with degree of tumor differentiation and tumor size, but not with tumor number (P>0.05). Immunohistochemical analysis showed that 17 of 19 HCC patients with HBVDNA -positive were strongly expressed for gp96, whereas only 5 of 11 patients with HBV-DNA-negative were positive for gp96. A significant difference was found between the two groups (89.5% vs. 45.5%, P<0.05). CONCLUSIONS: The abnormal expressions of HSP gp96 in HCC tissues are associated with HBV replication. This finding indicates that HBV infection plays an important role in the development of HCC.
