Anticancer activity of sodium caffeate and its mechanism

Aim: To study the anticancer activity of sodium caffeate （SC）. Methods: A nucleoside transport assay was used to analyze the inhibitory effects of SC on nucleoside rescue. The MTT assay was used to measure cell proliferation. Flow cytometrywas used to measure the apoptosis of BEC-7402 induced by SC and the cell cycledistribution change. Western blotting analysis was employed to investigate Bcl2, caspase and Bax expression. Intracellular Ca2+and mitochondrial membranepotential were determined by flow cytometry. In vivo anti-tumor activity wasmeasured using a tumor transplantation model in mice. Results: SC inhibited thenucleoside transport of BEL-7402 cells with an IC50of 1.02 mg/mL. SC inhibitedtumor cell proliferation with an IC50between 100 μg/mL and 200μg/mL. SC induced BEL-7402 cell apoptosis in a time-and dose-dependent manner, which wasinduced by arresting cells in S phase. The in vivo study showed that tumorgrowth was inhibited in a dose-dependent manner. Activated caspase-3 and Baxexpression were up-regulated after treatment with SC, while Bcl-2 expression wasdown-regulated. Intracellular Ca2+was increased while mitochondrial membranepotential was decreased by SC. Conclusion: SC is a new anticancer agent withpromising potential.

Chemopreventive effect of dimethyl dicarboxylate biphenyl on malignant transformation of WB-F344 rat liver epithelial cells

Aim: To study the potential chemopreventive effect of dimethyl dicarboxylate biphenyl (DDB), an anti-hepatitis drug, on hepatocarcinogenesis in vitro. Methods: The anti-carcinogenesis effect of DDB was assessed on a two-stage chemical oncogenesis model induced by 3-methylcholanthrene and 12-0-tetradecanoyl phorbol 13-acetate (TPA) with WB-F344 rat liver epithelial cells (WB-F344 cells) in vitro. A soft-agar colony formation assay was used to determine the tumorigenic potential of the transformed WB-F344 cells. The gap junctional intercellular communication (GJIC) was detected using the scrape loading/ dye transfer technique. Results: DDB at 1 μmol/L, 2 μmol/L, and 4 μmol/L significantly prevented the malignant transformation of WB-F344 cells induced by 3-methylcholanthrene and TPA. The average number of transformed foci decreased dramatically by 10.0%, 37.2%, and 47.4%, respectively. In soft agar, a remarkable decrease in colony numbers was observed in transformed cells treated with 2 μmol/L and 4 μmol/L DDB. DDB at 1 μmol/L, 2 μmol/L, and 4 μmol/L inhibited the downregulation of GJIC induced by TPA in a dose-dependent manner. The GJIC recovered to 25.6%, 34.6%, and 44.9%, respectively, of the control WB-F344 cells by DDB. Conclusion: DDB has a potential chemopreventive effect on hepatocarcinogenesis induced by carcinogens in vitro.

Cytokines Expression in Lymphocytes From Rats With Allergic Asthma Induced by Pleurotus sapidus besidiospores

Allergic asthma caused by mushroom spores (Pleurotus sapidus besidiospores) is a common health problem among mushroom-cultivating workers in china. An animal model of allergic asthma through the challenge of Pleurotus sapidus besidiospores in primed rats was developed for investigating the role of cytokines in the pathogenesis of the disease. In the study a series of related cytokines and their receptors, including their activity and mRNA levels of spleen lymphocytes isolated from asthmatic rats, were measured. Determined by 3H-TdR incorporation assay and NAG microcolorimetric assay, Con A-induced spleen lymphocyte proliferation and IL-2 activity in culture supernatants of spleen lymphocytes from 7-day challengedrats with allergic asthma increased significantly by 261% and 208%, respectively, as compared with those in the control. Cytokines and their receptor expression at mRNA levels were determined by RNA/cDNA hybridization, using (α-32P-dCTP radiolabeled cDNA probes for different cytokines and their receptors in vitro. The results showed that mRNA expression of IL-4, GM-CSF, IL-6, IL-2 and IL-2R, except IL-6R, in lymphocytes of 7-day-challenged asthma-suffering rats, increased significantly by 54%, 45%, 170%, 83% and 76%, respectively. In 2-day challenged-rats, mRNA levels of IL-4, IL-6 and IL-2 increased by 37%,58% and 125%, respectively, whereas mRNA leve1s of GM-CSF, IL-2R and IL-6R remained unchanged. Thus, the experimental results suggested a significant increase in TH2type cytokines in the pathogenesis of Pleurotus sapidus besidiospore-induced allergic asthma and IL-4 may play an essential role.

Editorial comment Translational medicine is a new discipline which aims to eliminate the barrier between preclinical and clinical medicine. Here, Dr. Li discusses the application of translational medicine in the research, teaching and clinical work of Prof. Liu Shih-Hao, the founder of endocrinology in china, who was particularly renowned for his early work in calcium and phosphorus metabolism. This well-known success story can be traced back to an early appreciation of translational medicine by Prof. Liu Shih-Hao, and serves as an important and revelatory lesson for us all.

Reference ranges and age-related changes of peripheral blood lymphocyte subsets in chinese healthy adults

This study was performed to build region-specific reference ranges of peripheral blood lymphocyte subsets for chinese healthy adults from the young to the elderly and analyze the trends of changes in lymphocyte subsets for evaluating the impact of age on the values.151 healthy adults aged 19-86 were recruited based on the SENIEUR *** sets of reference ranges were finally built applicable for the healthy young(19-44 years),middle-aged(45-64 years) and elder adults(≥65).Comparisons in parameters among the three cohorts showed that a statistically significant increase in CD16CD56+ NK cell was observed between the middle-aged and elder cohorts,whereas for the majority of the parameters,a significant decline was observed between the young and the middle-aged *** results showed that inverse correlations were observed between the age and CD19+ B,CD3+ T,CD3+CD4+ T,CD4+CD45RA+CD62L+ nave T cell and CD4+CD28+/CD4+,while the positive one was identified between the age and the NK *** significant changes of the most of immune parameters provided evidence for ***,T cell activation markers of CD8+CD38+ and CD8+HLA-DR+ showed reverse trends of association with age,which provides a clue for further researches on the mechanisms underlying the paradoxical clinical presentation of the elder patients.

Background: The incidence and mortality of colorectal cancer (CRC) in china are increasing in recent years. The clarified pathogenesis and detectable precancerous lesions of CRC make it possible to prevent, screen, and diagnose CRC at an early stage. With the development of endoscopic and surgical techniques, the choice of treatment for early CRC is also worth further discussion, and accordingly, a standard follow-up program after treatment needs to be ***: This clinical practice guideline (CPG) was developed following the recommended process of the World Health Organization, adopting Grading of Recommendations Assessment, Development and Evaluation (GRADE) in assessing evidence quality, and using the Evidence to Decision framework to formulate clinical recommendations, thereby minimizing bias and increasing transparency of the CPG development process. We used the Reporting Items for practice Guidelines in HealThcare (RIGHT) statement and Appraisal of Guidelines for Research and Evaluation II (AGREE II) as reporting and conduct guides to ensure the guideline’s completeness and ***: This CPG comprises 46 recommendations concerning prevention, screening, diagnosis, treatment, and surveillance of CRC. In these recommendations, we have indicated protective and risk factors for CRC and made recommendations for chemoprevention. We proposed a suitable screening program for CRC based on the chinese context. We also provided normative statements for the diagnosis, treatment, and surveillance of CRC based on existing clinical evidence and ***: The 46 recommendations in this CPG are formed with consideration for stakeholders’ values and preferences, feasibility, and acceptability. Recommendations are generalizable to resource-limited settings with similar CRC epidemiology pattern as china.

High-throughput fluorescence polarization method for identifying ligands of LOX-1

Aim: To develop and optimize a competitive fluorescence polarization(FP)method,and use it as a high-throughput screening(HTS)assay for drug ***: Human lectin-like oxidized low-density lipoprotein receptor-1(hLOX-1)and oxidized low-density lipoprotein(oxLDL)were used to establish a high-throughput fluorescence polarization assay to screen ligands of human LOX-1. A96-well plate assay was performed with a fast plate reader. Three fluoresceinisothiocyanate-labeled hLOX-1 concentrations(100, 200, and 400 nmol/L)wereselected to be titrated by oxLDL(from 0.05 nmol/L to 100 μmol/L)in order to obtainoptimal reactive concentrations. The concentration of Me2SO used(0%, 1%, 3%,5%) and incubation time(15 min, 30 min, 1 h, 2 h)were optimized. The Z’factor wascalculated to estimate the quality of FP-based HTS. Results: Concentrations of200 nmol/L for human LOX-1 and 50 μmol/L for oxLDL were used in the actualassay. Concentrations of 0% to 5% Me2SO and different reaction times did notaffect the FP-based HTS. The Z’value was 0.66. By using this detection andscreening system, 12 700 compounds were screened and 3 ligands with an IC50ofless than 4.5 μmol/L were found. Conclusion: The established competitive FP-based assay is sensitive, stable, highly reproducible and robust, and suitable forHTS for ligands of the hLOX-1 receptor.

Cloning and Characterization of a Novel Gene SRG-L Expressed in Late Stages of Mouse Spermatogenic Cells

A cDNA expressed specifically in late stages of mouse spermatogenic cells during spermato- genesis was cloned by using overlapping RT-PCR and RACE. The cDNA contained an open reading frame (ORF) of 2625 base pairs that encoded an 874 amino acids protein. Comparison analysis of amino acid sequence showed 91% and 80% identity to a rat homologue XP-226242 and a monkey homologue BAB63115 respectively. The expression of the mRNA was only observed in pachytene spermatocytes, round, and elongating spermatids. We named this gene as SRG-L (spermatogenesis related gene expressed in the late stages of spermatogenic cells, GenBank accession No. AY352586). The tissue-specific analysis showed that the SRG-L was highly expressed in spleen and testis. The results suggested that SRG-L might play special roles during spermatogenesis, particularly related to meiosis and spermiogenesis. Analysis of the amino acid sequence showed there was a coiled-coil region near the N-terminal region and rich phosphorylation sites, suggesting SRG-L might function as a transmembrane protein mediating signal transduction. This study also demonstrated that gene cloning by RT-PCR was applicable and convenient when its homologous gene was known.
