Curcumin Alleviates Hyperandrogenism and Promotes Follicular Proliferation in Polycystic Ovary Syndrome Rats:Insightson IRS1/PI3K/GLUT4 and PTEN Modulations

Objective:To explore the effect of curcuminon the insulin receptor substrate1(IRS1)/phosphatidylinositol-3-kinase(PI3K)/endometrial expression of glucose 4(GLUT4)signalling pathway and its regulator,phosphatase and tensin homolog(PTEN),in a rat model of polycystic ovarian syndrome(PCOS).Methods:PcoS model was induced by letrozole intragastric ***-Dawleyrats were randomized into 4groups according to a random number table:(1)control group;(2)PcoS group,which was subjected to PCOS and received vehicle;(3)curcumin group,which was subjected to PCoS and treated with curcumin(200 mg/kg for 2 weeks);and(4)curcumin+LY294002 group,which was subjected to PCOS,and treated with curcumin and LY294002(a specific PI3K inhibitor).Serum hormone levels(17β-estradiol,follicle stimulating hormone,luteinizinghormone,progesterone,and testosterone)were measured by enzyme linked immunosorbentassay,and insulin resistance(IR)was assessed using the homeostasismodel assessment of *** tissues were stained with haematoxylin and eosin for pathological and apoptosis *** levels of key transcriptional regulators and downstream targets,including IRS1,Pl3K,protein kinase B(AKT),GLUT4,and PTEN,were measured via reverse transcription polymerase chain reaction and Western blot,***:The Pcos group showed impaired ovarian morphology and *** with the PCoS group,curcumin treatment exerted ovarioprotective effects,down-regulated serum testosterone,restored IR,inhibited inflammatory cell infiltration in ovarian tissues,decreased IRS1,PI3K,and AKT expressions,and up-regulated GLUT4 and PTEN expressions in PCOS rats(P0.05).Conclusion:The beneficial effects of curcumin on PCOS rats included the alteration of serum hormone levels and recovery of morphological ovarian lesions,in which,PTEN,a new target

目的探讨低频脉冲电磁场对骨质疏松性胸腰椎骨折内固定术后康复的促进作用。方法将118例骨质疏松性胸腰椎骨折内固定术后患者采用随机数字表分两组。对照组给予唑来膦酸治疗,观察组给予低频脉冲电磁场联合唑来膦酸治疗。对比两组患者疼痛程度,骨折愈合时间,治疗前后的骨密度,骨代谢指标水平与腰椎功能评分变化。结果111例患者均获得完整随访,随访率94.07%,随访时间6~12个月,平均(9.00±1.08)个月。其中观察组男20例、女37例,年龄50~83岁,平均(64.50±5.65)岁;对照组男18例、女36例,年龄50~83岁,平均(64.05±5.82)岁。观察组和对照组不同时刻疼痛评分进行重复测量设计的方差分析显示,观察组和对照组组间差异有统计学意义[观察组:(8.51±0.79)分、(5.72±0.65)分、(2.86±0.41)分,对照组:(8.96±0.91)分、(6.38±0.72)分、(4.77±0.53)分](F=12.451,P<0.001),组别与时间点间存在交互作用(F=11.338,P<0.001),各时间点两组差异均有统计学意义(F=2.786、5.074、21.301,P=0.006、<0.001、<0.001);观察组在各时间点间差异均有统计学意义(F=18.967,P<0.001);观察组骨折愈合时间短于对照组[(12.31±2.07)周比(14.76±2.25)周](t=6.125,P<0.001);观察组和对照组治疗前与治疗6个月后在腰椎(L1~L4)骨密度、N-MID、β-CTx和PINP水平及腰椎功能障碍评分[观察组:(0.65±0.07)g/cm2与(0.71±0.08)g/cm2、(15.58±3.01)ng/mL与(8.82±1.23)ng/mL、(0.32±0.06)ng/mL与(0.13±0.02)ng/mL、(32.89±4.33)ng/mL与(54.56±7.37)ng/mL、(60.18±5.20)分与(15.88±3.01)分;对照组:(0.64±0.06)g/cm2与(0.67±0.07)g/cm2、(15.43±2.96)ng/mL与(11.24±1.74)ng/mL、(0.31±0.06)ng/mL与(0.19±0.04)ng/mL、(33.02±4.41)ng/mL与(41.23±6.06)ng/mL、(59.25±5.43)分与(24.35±4.20)分],进行重复测量设计的方差分析显示,观察组和对照组组间差异有统计学意义(F=12.401、15.468、18.742、16.229、18.924,均P<0.001),组别与时间点间存在交互作用(F=8.065、12.224、18.965、22.467、14.881,均P<0.001),治疗前两组差异均无统计学意义,治疗6个月后两组差异均有统计学意义(F=2.894、8.750、10.318、10.738、10.330,均P<0.05);观察组在治疗前与治疗6个月后差异均有统计学意义(F=2.924、19.286、22.569、36.385、12.387,均P<0.05)。结论低频脉冲电磁场在骨质疏松性胸腰椎骨折内固定术后能够有效缓解患者骨质疏松疼痛,缩短骨折愈合时间,提高骨密度,改善骨代谢,还可提高腰椎功能。

The diffusion and chemical reactions inside the catalyst particles and the heterogeneous flow structure in the computational cells are key factors to affect the accuracy of the coarse-grid simulation in circulating fluidized bed(CFB)methanation *** this work,a particle-scale model is developed to calculate the effective reaction rate considering the transient diffusion and chemical reactions in the particle scale,i.e.,the scale of the single catalyst particle.A modified sub-grid drag model is proposed to consider the effects of the meso-scale and chemical reactions on the heterogeneous gas-solid interaction,where the meso-scale is between the single particle and the whole reactor and featured with the particle ***,a coupled model is developed by integrating the particle-scale and modified sub-grid drag models into ***,the coupled model is validated to achieve accurate predictions on the CO methanation process in a CFB ***,the coupled model can be performed with a coarse grid(∼58 times particle diameter)and a large time step(0.005 s)to accelerate the *** simply changing the reaction kinetics,different gas-solid catalytic reaction systems can be simulated by using the coupled model.

目的探索土木香内酯体内外的抗病毒作用,并基于Ⅰ型干扰素(typeⅠinterferon,IFN-Ⅰ)通路探究其抗病毒作用机制。方法CCK-8法检测土木香内酯对A549细胞活力的影响;利用表达绿色荧光蛋白的水疱性口炎病毒(vesicular stomatitis virus expressing green fluorescent protein,VSV-GFP)感染细胞模型,结合流式细胞术检测土木香内酯对病毒复制的影响;qRT-PCR检测土木香内酯对甲型流感病毒(influenza A virus,H1N1)、脑心肌炎病毒(encephalomyocarditis virus,EMCV)复制的影响;Western blotting检测土木香内酯对VSV病毒G蛋白和H1N1病毒NP蛋白表达的影响;利用生物信息学初步分析土木香内酯的抗病毒分子机制;qRT-PCR检测药物处理后干扰素α1(interferonα1,Ifna1)、干扰素β1(interferonβ1,Ifnb1)、干扰素诱导蛋白1(interferon-induced protein with tetratricopeptide repeats 1,Ifit1)、干扰素刺激基因15(interferon stimulated gene 15,Isg15)的mRNA表达变化;流式细胞术和qRT-PCR检测Ⅰ型干扰素受体1(typeⅠinterferon receptor 1,IFNAR1)敲除(Ifnar1^(-/-))细胞中,土木香内酯的抗病毒作用与干扰素信号通路的关系;构建小鼠H1N1病毒滴鼻感染模型探究土木香内酯的体内抗病毒作用。结果土木香内酯体外抑制VSV、H1N1和EMCV病毒复制;土木香内酯激活IFN-Ⅰ信号通路;在Ifnar1^(-/-)细胞中土木香内酯抗病毒作用被削弱;土木香内酯提高H1N1感染小鼠的存活率,降低脏器H1N1病毒载量。结论土木香内酯激活基于IFN-Ⅰ信号通路的抗病毒天然免疫反应,发挥抵抗多种病毒复制的作用。

HetNets Under Decoupled Uplink and Downlink Access with UE Random Discontinuous Transmission: Local Delay and Energy Efficiency

Due to the limited uplink capability in heterogeneousnetworks (HetNets), the decoupled uplinkand downlink access (DUDA) mode has recently beenproposed to improve the uplink performance. In thispaper, the random discontinuous transmission (DTX)at user equipment (UE) is adopted to reduce the interferencecorrelation across different time slots. By utilizingstochastic geometry, we analytically derive themean local delay and energy efficiency (EE) of an uplinkHetNet with UE random DTX scheme under theDUDA mode. These expressions are further approximatedas closed forms under reasonable *** results reveal that under the DUDA mode, there isan optimal EE with respect to mute probability underthe finite local delay constraint. In addition, with thesame finite mean local delay as under the coupled uplinkand downlink access (CUDA) mode, the HetNetsunder the DUDA mode can achieve a higher EE witha lower mute probability.

集装箱码头在不发达国家、地区,在供电基础设施条件不满足运营需要的情况下,多采用自建柴油机电站供电的形式。岸桥起重机在作业过程中负荷特性频繁波动,在极端情况下会有很大的逆功率反馈到供电系统中,给柴油机组运行带来挑战。根据所依托工程的情况,对比研究不同储能技术特点,提出以解决负荷侧波动对供电系统的影响为重点考虑、采用高速电气飞轮和BESS两组储能装置研究的建议,并且给出开展微网控制策略研究、预留新能源接口及微网协同、微配协同接口的研究思路建议,形成模块化、标准化、智能化的集装箱码头微电网配置方案。

Deep geothermal resources in the Fujian-Guangdong-Hainan region,China,offer significant potential for sustainable *** diverse igneous rock formations along the southeast coast present intricate geological challenges that impede exploration and evaluation *** this study,we address critical concerns related to the Fujian-Guangdong-Hainan region's deep geothermal resources,encompassing heat source composition,formation conditions,strategic favorable areas,and exploration *** methods involve the analysis of regional geothermal reservoirs and cap *** findings include:the primary heat sources in the Fujian-Guangdong-Hainan region consist of the radioactive heat generation from granites in the crust,heat conduction in the mantle,and,in specific areas like Yangjiang and Shantou,melts within the middle and lower crust;the deep,high-temperature geothermal resources in the region predominantly reside in basins'depressed *** areas are characterized by the confluence of triple heat sources:heat from the Earth's crust,mantle,and other supplementary sources;our analysis led to the identification of three strategic areas favorable for deep geothermal resources in the Fujian-Guangdong-Hainan *** are the Beibu Gulf Basin's continental area,the Yuezhong Depression,and the Fuzhou-Zhangzhou area.
