Azvudine(FNC)is a nucleoside analog that inhibits HIV-1.RNA-dependent RNA polymerase(RdRp).Recently,we discovered FNC an agent against SARS-CoV-2.and have taken it into Phase III trial for COVID-1. *** monophosphate analog inhibited SARS-CoV-2.and HCoV-OC43 coronavirus with an EC_(50) between 1.2.and 4.3 μM,depending on viruses or cells,and selective index(SI)in 1.-83 *** administration of FNC in rats revealed a substantial thymus-homing feature,with FNC triphosphate(the active form)concentrated in the thymus and peripheral blood mononuclear cells(PBMC).Treating SARS-CoV-2.infected rhesus macaques with FNC(0.07 mg/kg, qd,orally)reduced viral load,recuperated the thymus,improved lymphocyte profiles,alleviated in flammation and orga n damage,and lessened grou nd・glass opacities in chest *** gle-cell seque ncing suggested the promotion of thymus function by FNC.A randomized,single-arm clinical trial of FNC on compassionate use(n=31.showed that oral FNC(5 mg,qd)cured all COVID-1. patients,with 1.0%viral ribonucleic acid negative conversion in 3.2.±2.2. days(range:1.9 days)and 1.0%hospital discharge rate in 9.00±4.93 days(range:2.2. days).The side-effect of FNC is minor and transient dizziness and nausea in 1..1.0}(5/31.***,FNC might cure COVID-1. through its anti-SARS-CoV-2.activity concentrated in the thymus,followed by promoted immunity.

Seed germination and seedling establishment are important for the reproductive success of plants,but seeds and seedlings typically encounter constantly changing environmental *** inhibiting seed germination and post-germinative growth through the PYR1.PYL/RCAR ABA receptors and PP2. co-receptors,the phytohormone abscisic acid(ABA)prevents premature germination and seedling growth under unfavorable ***,little is known about how the ABA-mediated inhibition of seed germination and seedling establishment is ***,we report that ABA Signaling Terminator(ABT),a WD40 protein,efficiently switches off ABA signaling and is critical for seed germination and seedling *** is induced by ABA in a PYR1.PYL/RCAR-PP2.-dependent *** of ABT promotes seed germination and seedling greening in the presence of ABA,whereas knockout of ABT has the opposite *** found that ABT interacts with the PYR1.PYL/RCAR and PP2. proteins,interferes with the interaction between PYR1.PYL4 and ABI1.ABI2.and hampers the inhibition of ABI1.ABI2.by ABA-bound PYR1.PYL4,thereby terminating ABA *** together,our results reveal a core mechanism of ABA signaling termination that is critical for seed germination and seedling establishment in Arabidopsis.

目的:观察冠心病稳定型心绞痛牵涉痛分布区域与穴位的关系,并结合动物实验探讨局部敏化出现的规律。方法:临床研究部分,观察1.046例冠心病稳定型心绞痛患者外周压痛敏感部位、局部皮肤形态异常改变,并测量了77位患者疼痛阈值的变化。动物实验部分,以SD大鼠冠状动脉左前降支结扎4h为心肌缺血模型组(n=1.),以假手术为对照组(n=4),分别用在自然光下拍摄观察和活体荧光成像的方法,记录大鼠尾静脉注射Evans Blue后体表出现蓝染的部位,并检测痛阈变化。结果:心绞痛患者压痛部位以及皮肤局部组织结构的改变多分布在左侧胸前区、背部、肩部、上肢尺侧部;右侧也有一定数量的分布。压痛点出现在穴位上、穴位附近和穴位以外。心肌缺血的大鼠模型出现类似的变化。结论:心脏发生缺血性病变时,其相应节段(T 1.T 5)神经支配皮节区域发生规律性的牵涉性的"敏化"反应,这些敏感度改变的部位与"穴位"形成有关。

Immunomodulatory signaling imposes tight regulations on metabolic programs within immune cells and consequentially determines immune response *** the glucocorticoid receptor(GR)has been recently implicated in regulating the function of myeloid-derived suppressor cells(MDSCs),whether the dysregulation of GR in MDSCs is involved in immune-mediated hepatic diseases and how GR regulates the function of MDSCs in such a context remains ***,we revealed the dysregulation of GR expression in MDSCs during innate immunological hepatic injury(IMH)and found that GR regulates the function of MDSCs through modulating HIF1.-dependent *** modulation of GR by its agonist(dexamethasone,Dex)protects IMH mice against inflammatory ***,GR signaling suppresses HIF1.and HIF1.-dependent glycolysis in MDSCs and thus promotes the immune suppressive activity of *** studies reveal a role of GR-HIF1.in regulating the metabolism and function of MDSCs and further implicate MDSC GR signaling as a potential therapeutic target in hepatic diseases that are driven by innate immune cell-mediated systemic inflammation.

In addition to landscape changes,urbanization also brings about changes in environmental factors that can affect *** the common referral in the published literature to multiple environmental factors such as light and noise pollution,there is a gap in knowledge about their combined *** developed a multidimensional environmental framework to assess the effect of urbanization and multiple environmental factors(light,noise,and temperature)on life-history traits and breeding success of Barn Swallows(Hirundo rustica)across rural to urban gradients in four locations spanning over 2.00 km from North to South *** a single breeding season,we measured these environmental factors nearby nests and quantified landscape urbanization over a 1.m~*** then analysed the relationships between these multiple environmental factors through a principal component analysis and conducted spatially explicit linear-mixed effects models to assess their effect on lifehistory traits and breeding *** were particularly interested in understanding whether and how Barn Swallows were able to adapt to such environmental conditions associated with *** results show that there is significant variation in the exposure to environmental conditions experienced by Barn Swallows breeding across urbanization gradients in *** changes and their effects are complex due to the behavioural responses ameliorating potential negative effects by selecting nesting sites that minimize exposure to environmental ***,significant relationships between landscape urbanization,exposure to environmental factors,and life-history traits such as laying date and clutch size were ***,the impact on breeding success was,at least in our sample,negligible,suggesting that Barn Swallows are extremely adaptable to a wide range of environmental features.

BACKGROUND Myocardial bridging is a common anatomical malformation,and the milking effect is a characteristic phenomenon of myocardial bridging in coronary ***,the phenomenon is ***,this article reports an inconceivably rare myocardial bridging phenomenon that breaks through our conventional *** milking effect changed obviously in two coronary angiography examinations,which subverted the traditional deep-rooted view of the myocardial bridging phenomenon and revealed the limitations of coronary angiography in diagnosing myocardial bridging and judging the prognosis of *** SUMMARY A 63-year-old man was diagnosed with ST-segment elevation myocardial infarction and received primary percutaneous coronary intervention on December {1.,*** heart rate was 1.4 beats per minute,and blood pressure was 1..3/1..3 kPa.A severe milking effect was found in the left anterior descending coronary artery during his index coronary angiography on January 1.,*** patient was given intensive medical management,including aβ1.adrenergic receptor blocker,during hospitalization and after ***,coronary angiography showed that the previous impressive milking effect was dramatically alleviated(close to normal)at the follow-up on October 1.,*** that moment,the patient’s heart rate was 83 beats per minute,and blood pressure was 1.1.7/8.0 *** The myocardial bridging phenomenon is not invariable and,in certain circumstances,may ***,the autonomic nervous system may be involved in the myocardial bridging phenomenon.

姜黄素调节BDNF/TrkB信号通路对细菌性脑膜炎新生大鼠神经元凋亡的影响

目的:探讨姜黄素(Cur)调节脑源性神经营养因子(BDNF)/原肌球蛋白相关激酶B受体(TrkB)信号通路对细菌性脑膜炎(BM)新生大鼠神经元凋亡的影响。方法:随机选择1.只大鼠作为非感染组(NF组),其它大鼠通过额蛛网膜下腔注射肺炎链球菌ATCC4961.菌悬液构建BM大鼠模型,将构建成功的BM模型随机平分为BM组、L-Cur组(1.2. mg/kg)、M-Cur组(2.5mg/kg)、H-Cur组(5 mg/kg)、H-Cur+K2.2.组(5 mg/kg Cur+2.5μg/kg K2.2.),每组1.只大鼠,每天一次,连续注射3周。对大鼠称量体重并进行临床评分,HE染色以及尼氏染色观察神经细胞损伤;TUNEL染色评估神经元凋亡情况。免疫荧光染色检测神经胶质纤维酸性蛋白(GFAP)、脊髓钙离子接头蛋白-1.IBA-1.水平。实时荧光定量聚合酶链式反应(qRT-PCR)检测脑组织炎症因子和趋化因子水平。蛋白质印迹法(Western blot)检测脑组织凋亡蛋白以及BDNF/TrkB通路蛋白表达。结果:NF组脑组织结构正常,神经元胞质内出现大面积蓝色尼氏体。BM组蛛网膜下腔出现大量炎性渗出液和炎性细胞浸润现象,胞体增大,突起收缩,形态不规则;尼氏体数目、体重、Bcl-2.BDNF、p-TrkB/TrkB蛋白水平显著下降(P1.GFAP相对荧光强度值、趋化因子配体-2.CCL-2.、趋化因子配体-3(CCL-3)、肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)的相对表达量、神经细胞凋亡率、BCL2.Associated X蛋白(Bax)蛋白水平显著升高(P2.因(Bcl-2.、BDNF、p-TrkB/TrkB蛋白水平显著升高(P1.GFAP相对荧光强度值、CCL-2.CCL-3、TNF-α、IL-6的相对表达量、神经细胞凋亡率、Bax蛋白水平显著下降(P2.2.组与BM组效果相似,K2.2.逆转了Cur对BM大鼠神经元凋亡及损伤的改善作用。结论:Cur可能通过激活BDNF/TrkB信号通路降低BM大鼠神经元凋亡。

In fMRI experiments on object representation in visual cortex, we designed two types of stimuli: one is the gray face image and its line drawing, and the other is the illusion and its corresponding completed illusion. Both of them have the same global features with different minute details so that the results of fMRI experiments can be compared with each other. The first kind of visual stimuli was used in a block design fMRI experiment, and the second was used in an event-related fMRI experiment. Comparing and analyzing interesting visual cortex activity patterns and blood oxygenation level dependent (BOLD)- fMRI signal, we obtained results to show some invariance of global features of visual images. A plau- sible explanation about the invariant mechanism is related with the cooperation of synchronized re- sponse to the global features of the visual image with a feedback of shape perception from higher cortex to cortex V1. namely the integration of global features and embodiment of sparse representation and distributed population code.

Once-daily tadalafil administration has been well established; however, studies about tadalafil once-daily treatment in the Chinese population are lacking. In this phase 4, postmarketing study, we ascertained the long-term safety and effectiveness of tadalafil {1.5 mg and 5.0 mg once daily in Chinese men with erectile dysfunction （n = 635）. The primary endpoint of the study was safety at 1.1.months as assessed by the proportion of patients experiencing at least one treatment-emergent adverse event （serious or nonserious）. The secondary endpoints included safety and effectiveness, measured by the International Index of Erectile Function-Erectile Function （IIEF-EF） domain scores. Similar adverse events to the known safety profile of tadalafil, such as nasopharyngitis, upper respiratory tract infection, headache, and dizziness, were detected. No new cardiovascular safety concerns were observed. After 3 months of treatment, significant increases in IIEF-EF domain scores were detected for both {1.5-mg （least squares [LS] mean change： 6.3; 95% confidence interval [CI]： 5.4-7.1. P 〈 0.001. and 5.0-mg （LS mean change： 7.4; 95% CI： 6.8-7.9; P 〈 0.001. tadalafil doses, and significance was maintained up to 1.1.months. In addition, approximately 40% of patients regained normal erectile function （IIEF-EF 〉{1.） following 1.year of tadalafil once-daily treatment. The findings in this study provide evidence for the extended effectiveness and tolerability of tadalafil, demonstrating no new safety concerns, in a Chinese population and make once-daily tadalafil administration a viable option for improving sexual performance and satisfaction in Chinese men with erectile dysfunction.
