Efficacy and safety of sofosbuvir and daclatasvir in treatment of kidney transplantation recipients with hepatitis C virus infection

AIM To assess the efficacy and safety of sofosbuvir and daclatasvir regimens for kidney transplantation(KT) patients with hepatitis C virus(HCV) *** This study enrolled a prospective cohort of consecutive Chinese KT patients with HCV infection. They were given sofosbuvir combined with daclatasvir, with or without ribavirin. They were monitored regularly during and after the treatment. RESULTS Six patients were recruited in our prospective study cohort. All patients were male and naive to directacting antiviral treatment. The treatment duration was 12 wk. Most patients(4/6) were infected with HCV genotype 1b. HCV RNA was undetectable at week 4 after treatment and at the end of treatment in all patients. Sustained virological response rate at 12 wk was 100%(6/6). Two patients had to accept a half dose of sofosbuvir due to serum creatinine elevation during treatment. Kidney function in the remaining patients was stable. No serious adverse events(AEs) were observed. No patient discontinued antiviral therapy due to side effects. CONCLUSION Sofosbuvir and daclatasvir for treatment of KT recipients with HCV infection are highly efficient and safe. Patients tolerated the medications well, and no serious AEs were observed. Larger prospective cohort studies are needed to validate these results.

Intermittent tri-weekly docetaxel plus bicalutamide in patients with castration-resistant prostate cancer： a single-arm prospective study using a historical control for comparison

Whether continuous docetaxel （DTX） chemotherapy offers an advantage over intermittent therapy for castration-resistant prostate cancer （CRPC） is unknown. In this study, we evaluated the efficacy, toxicity and quality of life （QoL） of intermittent tri-weekly DTX with bicalutamide in CRPC. Forty-two patients （group A） with CRPC were enrolled. The patients received intravenous DTX （75 mg m^-2） once tri-weekly with oral bicalutamide （50 mg） once daily. Patients had a DTX holiday when the prostate-specific antigen （PSA） level declined ≥ 50%. DTX was restarted in patients with a PSA increase ≥ 25%. Sixty patients （group B） who had matching characteristics and had continuously received DTX without bicalutamide for 10-12 cycles were also enrolled. There were no statistically significant differences in progression-free survival （8 months vs. 9 months, P=0.866） or overall survival （19 months vs. 21 months, P=0.753） between groups A and B; however, the proportions of patients in group A with all grades of neutropenia （33% vs. 58%, P=0.013） and nausea/vomiting （11% vs. 29%, P=0.024） were significantly less compared to group B. A significant improvement in the global health and fatigue scores was recorded for group A post-chemotherapy compared to pre-chemotherapy （P〈0.05）. The fatigue, nausea/ vomiting and appetite loss scores in group B were increased post-chemotherapy compared to pre-chemotherapy （P〈0.05）. In conclusion, intermittent tri-weekly DTX plus bicalutamide is well tolerated and has the potential to achieve comparable disease control with an improvement in QoL for oatients with CRPC.

Value of Adjuvant Radiotherapy for Thymoma with Myasthenia Gravis after Extended Thymectomy

Background：The co-existence of myasthenia gravis （MG） and thymoma makes the surgical treatment more complicated and adjuvant radiation more controversial. The aim of this study was to investigate adjuvant radiotherapy for thymoma with MG after extended ***：A total of 181 patients with both MG and thymoma were recruited between 2003 and 2014 at Tongren Hospital, China. Among all the patients, 157 patients received radiation therapy after surgery （Group A）; whereas the other 24 patients did not receive radiation therapy （Group B）. According to the time that patients started mediastinal radiation therapy, we subdivided the 157 patients in Group A into subgroups （1-month subgroup, n = 98; 2-month subgroup, n = 7; and 3-month subgroup, n = 52）. We then compared the effect of the mediastinal radiation therapy across these different groups using the survival rate, the rate of postoperative myasthenic crisis, and the complete stable remission （CSR） rate as the primary ***：There was a significant difference in the occurrence of postoperative myasthenic crisis between 1-month subgroup and Group B （χ2 = 4.631, P = 0.031）. The rates of reaching CSR were 32.6% in 1-month subgroup, 25% in 3-month subgroup, and 22.7% in Group B, respectively. The overall survival rates of 1-month subgroup, 3-month subgroup, and Group B were 88.8%, 83.3%, and 77.3%, respectively. Analysis on the Kaplan-Meier survival curves demonstrated that within 8 years after surgery, there was no significant difference in aspects of overall survival and disease-free survival between 1-month subgroup and Group B, and between 3-month subgroup and Group B; over 8 years after surgery, the disease-free survival rates in 1-month subgroup, 3-month subgroup and Group B were 79.4%, 70.6%, and 55.3%, ***：Adjuvant radiation within 1 month after extended thymectomy may be helpful in controlling postoperative MG, such as decreasing the possibility of postoperative myasthenic

The increasing attention on deep learning has tremendously spurred the design of intelligence processing hardware. The variety of emerging intelligence processors requires standard benchmarks for fair comparison and system optimization （in both software and hardware）. However, existing benchmarks are unsuitable for benchmarking intelligence processors due to their non-diversity and nonrepresentativeness. Also, the lack of a standard benchmarking methodology further exacerbates this problem. In this paper, we propose BENCHIP, a benchmark suite and benchmarking methodology for intelligence processors. The benchmark suite in BENCHIP consists of two sets of benchmarks： microbenchmarks and macrobenchmarks. The microbenchmarks consist of single-layer networks, They are mainly designed for bottleneck analysis and system optimization. The macrobenchmarks contain state-of-the-art industrial networks, so as to offer a realistic comparison of different platforms. We also propose a standard benchmarking methodology built upon an industrial software stack and evaluation metrics that comprehensively reflect various characteristics of the evaluated intelligence processors, BENCHIP is utilized for evaluating various hardware platforms, including CPUs, GPUs, and accelerators. BENCHIP will be open-sourced soon.

2-(2-Benzofuranyl)-2-imidazoline treatment within 5 hours after cerebral ischemia/reperfusion protects the brain

We previously demonstrated that administering 2-（2-benzofuranyl）-2-imidazolin（2-BFI）, an imidazoline I2 receptor agonist, immediately after ischemia onset can protect the brain from ischemic insult. However, immediate administration after stroke is difficult to realize in the clinic. Thus, the therapeutic time window of 2-BFI should be determined. Sprague-Dawley rats provided by Wenzhou Medical University in China received right middle cerebral artery occlusion for 120 minutes, and were treated with 2-BFI（3 mg/kg） through the caudal vein at 0, 1, 3, 5, 7, and 9 hours after reperfusion. Neurological function was assessed using the Longa＇s method. Infarct volume was measured by 2,3,5-triphenyltetrazolium chloride assay. Morphological changes in the cortical penumbra were observed by hematoxylin-eosin staining under transmission electron microscopy. The apoptosis levels in the ipsilateral cortex were examined with terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling（TUNEL） assay. The protein expression of Bcl-2 and BAX was detected using immunohistochemistry. We found the following： Treatment with 2-BFI within 5 hours after reperfusion obviously improved neurological function. Administering 2-BFI within 9 hours after ischemia/reperfusion decreased infarct volume and alleviated apoptosis. 2-BFI administration at different time points after reperfusion alleviated the pathological damage of the ischemic penumbra and reduced the number of apoptotic neurons, but the protective effect was more obvious when administered within 5 hours. Administration of 2-BFI within 5 hours after reperfusion remarkably increased Bcl-2 expression and decreased BAX expression. To conclude, 2-BFI shows potent neuroprotective effects when administered within 5 hours after reperfusion, seemingly by up-regulating Bcl-2 and down-regulating BAX expression. The time window provided clinical potential for ischemic stroke by 2-BFI.

Lunar Penetrating Radar （LPR） onboard the rover that is part of the Chang＇e-3 （CE-3） mission was firstly utilized to obtain in situ measurements about geological structure on the lunar surface and the thickness of the lunar regolith, which are key elements for studying the evolutional history of lunar crust. Because penetra- tion depth and resolution of LPR are related to the scientific objectives of this mission, a series of ground-based experiments using LPR was carried out, and results of the experimental data were obtained in a glacial area located in the northwest region of China. The results show that the penetration depth of the first channel antenna used for LPR is over 79 m with a resolution of 2.8 m, and that for the second channel antenna is over 50.8 m with a resolution of 17.1 cm.
