Identification of Quantitative Trait Loci Affecting Hemicellulose Characteristics Based on Cell Wall Composition in a Wild and Cultivated Rice Species

Cell wall hemicellulosic polysaccharides are structurally complex and diverse. Knowledge about the synthesis of cell wall hemicelluloses and their biological roles is limited. Quantitative trait loci （QTL） mapping is a helpful tool for the dissection of complex phenotypes for gene identification. In this study, we exploited the natural variation in cell wall monosaccharide levels between a common wild rice, Yuanj, and an elite indica cultivar, Teqing, and performed QTL map- ping with their introgression lines （ILs）. Chemical analyses conducted on the culms of Yuanj and TeqJng showed that the major alterations are found in glucose and xylose levels, which are correlated with specific hemicellulosic polymers. Gly- cosidic linkage examination revealed that, in Yuanj, an increase in glucose content results from a higher level of mixed linkage I^-glucan （MLG）, whereas a reduction in xylose content reflects a low level of xylan backbone and a varied arabi- noxylan （AX） structure. Seventeen QTLs for monosaccharides have been identified through composition analysis of the culm residues of 95 core ILs. Four major QTLs affecting xylose and glucose levels are responsible for 19 and 21% of the phenotypic variance, respectively. This study provides a unique resource for the genetic dissection of rice cell wall forma- tion and remodeling in the vegetative organs.

With an upsurge of biodegradable metal implants,the research and application of Mg alloys in the gastrointestinal environment of the digestive tract have been of great *** enzymes,mainly pepsin in the stomach and pancreatin in the small intestine,are widespread in the gastrointestinal tract,but their effect on the degradation of Mg alloys has not been well *** this study,we investigated the impacts of pepsin and pancreatin on the degradation of Mg-2Zn alloy *** results showed that the pepsin and pancreatin had completely different even the opposite effects on the degradation of Mg,although they both affected the degradation product *** degradation rate of Mg wire declined with the addition of pepsin in simulated gastric fluid(SGF)but rose with the addition of pancreatin in simulated intestinal fluid(SIF).The opposite trends in degradation rate also resulted in completely different degradation morphologies in wires surface,where the pitting corrosion in SGF was inhibited because of the physical barrier effect of pepsin *** contrast,the adsorption of pancreatin affected the integrity of magnesium hydrogen phosphate film,causing a relatively uneven degraded *** results may help us to understand the role of different digestive enzymes in the degradation of magnesium and facilitate the development and clinical application of magnesium alloy implanted devices for the digestive tract.

Screening of hepatocyte proteins binding to F protein of hepatitis C virus by yeast two-hybrid system

AIM： To investigate the biological function of F protein by yeast two-hybrid system. METHODS： We constructed F protein bait plasmid by cloning the gene of F protein into pGBKT7, then recombinant plasmid DNA was transformed into yeast AH109 （a type）. The transformed yeast AH109 was mated with yeast Y187 （α type） containing liver cDNA library plasmid in 2×YPDA medium. Diploid yeast was plated on synthetic dropout nutrient medium （SD/-Trp-Leu-HisAde） containing X-α-gal for selection and screening. After extracting and sequencing plasmids from positive （blue） colonies, we underwent sequence analysis by bioinformatics. RESULTS： Thirty-six colonies were selected and sequenced. Among them, 11 colonies were zymogen granule protein, 5 colonies were zinc finger protein, 4 colonies were zinc-α-2-glycoprotein, 1 colony was sialyltransferase, 1 colony was complement control protein factor I, 1 colony was vitronectin, and 2 colonies were new genes with unknown function. CONCLUSION： The yeast two-hybrid system is an effective method for identifying hepatocyte proteins interacting with F protein of hepatitis C virus. Comparison of FP-7 and S-2 Ahmed glaucoma valve implantation in refractory glaucoma patients for short-term follow-up

Background Ahmed glaucoma valves （AGV） has been used for decades, but there is no detailed report about the efficacy of AGV in Chinese glaucoma patients. This study aimed to compare the intraocular pressure （IOP） lowering efficacy and side effects of S-2 polypropylene and PF-7 silicone AGV implantation in Chinese refractory glaucoma *** Patients were divided into S-2 model AGV group and FP-7 model AGV group. The complete and qualified surgical success rate, change of IOP, number of anti-glaucoma medications used and postoperative complications were recorded and *** Average follow-up time was comparable between two groups. IOP was reduced from （37.9±12.7） mmHg preoperatively to （17.3±5.3） mmHg at the last follow-up in S-2 group and reduced from （39.9±14.4） mmHg to （17.7±4.9）mmHg in FP-7 group. Anti-glaucoma medications were reduced from 3.8±0.2 to 1.5±0.2 in S-2 group, and 3.5±0.2 to 0.7±0.2 in FP-7 groups. The cumulative success rates were comparable in two groups, which were 61.2% and 72.1% in S-2 group and FP-7 group respectively. When IOP reduction criteria was used, complete success rates were 30.6% and 51.2% for S-2 and FP-7 groups, and qualified success rates were 86.1% and 92.7% separately. In both groups, the major complication was hypotony, and the previous trabeculectomy of patients was the major risk factor for surgery *** In this short-term retrospective study, S-2 AGV is showed at least as effective as FP-7 AGV in IOP reduction, but associated with higher rate of complications. Previous trabeculectomy is a principle risk factor for AGV implantation failure. These clinical outcomes are important for converting use of the FP-7 silicon AGV in Chinese refractory glaucoma patients.

目的基于特征离子分析川陈皮素在大鼠体内代谢产物。方法10只大鼠随机分为给药组和空白组,分别灌胃给予川陈皮素0.5%CMC-Na混悬液(250 mg/kg)和0.5%CMC-Na溶液,采集血浆、尿液和粪便,固相萃取法前处理,进行UHPLC-HRMS分析。根据诊断产物离子、色谱保留时间、精确分子量和中性丢失碎片对代谢物进行系统描述,以-CH_(3)(m/z 15)特征离子为诱饵,在代谢物数据集中获得准确的代谢物。结果共鉴定出64个代谢物,主要代谢途径为葡萄糖醛酸化、硫酸化、氢化及其复合反应。结论本实验阐明了川陈皮素在大鼠体内代谢产物,为其进一步开发提供了新的借鉴。

Lignans and diterpenes isolated from Tirpitzia ovoidea and their biological activities

A new lignan, tirpitzin A(17) together with 20 known compounds(1-16, and 18-21) were isolated from the ethyl acetate soluble fraction of ethanol extract of the aerial parts of Tirpitzia ovoidea. The structure of new compound was elucidated by means of spectroscopic analysis. Of the known compounds, 7-21 were isolated from Linaceae family for the first time. The pharmacological activity of the crude extracts was tested using a mouse inflammation model induced by dimethyl benzene. The results demonstrated that the ethyl acetate soluble fraction had anti-inflammatory activity. Moreover, the cytotoxic and anti-inflammatory activities of some compounds were studied. The new compound 17 showed moderate cytotoxic effect against Bx PC-3 cell line(IC_50 = 19.51μmol·L^(-1)) and Compound 10 showed significant cytotoxicity against Hep G2, HL-60, U87 and Bx PC-3 cell lines with IC_50 values in the range 4.2-8.3μmol·L^(-1). Additionally, Compounds 2, 10, 11, and 13 exhibited potent inhibitory effects on LPS-induced nitric oxide production in RAW 264.7 macrophages at the concentration of 50μmol·L^(-1).

Oximes derivatives have been vastly used in organic synthesis. In this review, C(sp^(3))-H bond functionalization of oximes derivatives via iminyl radical induced 1,5-hydrogen atom transfer was discussed. According to the different type of products, this review was divided into three parts: (1) C(sp^(3))-H bond functionalization for C-C bond formation. (2) C(sp^(3))-H bond functionalization for C-N bond formation. (3) C(sp^(3))-H bond functionalization for C-S, C-F bond formation.

Screening of genes of proteins interacting with p7 protein of hepatitis C virus from human liver cDNA library by yeast two-hybrid system

AIM： To investigate the biological function of p7 protein and to look for proteins interacting with p7 protein in hepatocytes. METHODS： We constructed p7 protein bait plasmid by cloning the gene of p7 protein into pGBKTT, then transformed it into yeast AH109 （a type）. The transformed yeast was mated with yeast Y187 （α type） containing liver cDNA library plasmid, pACT2 in 2×YPDA medium. Diploid yeast was plated on synthetic dropout nutrient medium （SD/- Trp-Leu-His-Ade） containing x-α-gal for selection and screening. After extracting and sequencing of plasmids from blue colonies, we performed sequence analysis by bioinformatics. RESULTS： Fifty colonies were selected and sequenced. Among them, one colony was Homo sapiens signal sequence receptor, seven colonies were Homo sapiens H19, seven colonies were immunoglobulin superfamily containing leucine-rich repeat, three colonies were spermatid peri-nuclear RNA binding proteins, two colonies were membrane-spanning 4-domains, 24 colonies were cancer-associated antigens, four colonies were nudeoporin 214 ku and two colonies were CLL-associated antigens. CONCLUSION： The successful cloning of gene of protein interacting with p7 protein paves a way for the study of the physiological function of p7 protein and its assodated protein.

Comparative Toxicity of Nanomaterials to Air-blood Barrier Permeability Using an In Vitro Model

Objective To comparatively study the toxicity of four metal-containing nanoparticles(MNPs) and their chemical counterparts to the air-blood barrier(ABB) permeability using an in vitro *** ABB model, which was developed via the co-culturing of A549 and pulmonary capillary endothelium, was exposed to spherical CuO-NPs(divided into CuO-40, CuO-80, and CuO-100 based on particle size), nano-Al2O3(sheet and short-rod-shaped), nano-ZnO, nano-Pb S, CuSO4, Al2(SO4)3, Zn(CH3COO)2, and Pb(NO3)2 for 60 *** 10 min following exposure, the cumulative cleared volume(ΔTCL) of Lucifer yellow by the model was calculated.A clearance curve was established using linear regression analysis of ΔTCL versus *** coefficient(P) was calculated based on the slope of the curve to represent the degree of change in the ABB *** The results found the increased P values of CuO-40, CuO-80, sheet, and short-rod-shaped nano-Al2O3, Al2(SO4)3, and Pb(NO3)*** them, small CuO-40 and CuO-80 were stronger than CuO-100 and CuSO4;no difference was observed between Al2(SO4)3 and sheet and short-rod-shaped nano-Al2O3;and nano-Pb S was slightly weaker than Pb(NO3)*** clearly the MNPs possess diverse *** ABB permeability abnormality means pulmonary toxicity *** studies are warranted to understand MNPs toxicity and ultimately control the health hazards.

Uncovering the dynamic evolution of nucleotidebinding site-leucine-rich repeat(NBS-LRR) genes in Brassicaceae

Plant genomes harbor dozens to hundreds of nucleotide-binding site-leucine-rich repeat （NBS-LRR） genes; however, the long-term evolutionary history of these resistance genes has not been fully understood, This study focuses on five Brassicaceae genomes and the Carica papaya genome to explore changes in NBS-LRR genes that have taken place in this Rosid II lineage during the past 72 million years. Various numbers of NBS-LRR genes were identified from Arabidopsis lyrata （198）, A. thaliana （165）, Brassica rapa （204）, Capsella rubella （127）, Thellungiella salsuginea （88）, and C. papaya （51）. In each genome, the identified NBS-LRR genes were found to be unevenly distributed among chromosomes and most of them were clustered together. Phylogenetic analysis revealed that, before and after Brassicaceae speciation events, both toll/interleukin-1 receptor-NBS-LRR （TNL） genes and non-toll/interleukin-1 receptor-NBS-LRR （nTNL） genes exhibited a pattern of first expansion and then contraction, suggesting that both subclasses of NBS-LRR genes were responding to pathogen pressures synchronically. Further, by examining the gain/loss of TNL and nTNL genes at different evolutionary nodes, this study revealed that both events often occurred more drastically in TNL genes. Finally, the phylogeny of nTNL genes suggested that this NBS-LRR subclass is composed of two separate ancient gene types： RPW8-NBS-LRR and Coiled-coiI-N BS-LRR.
