Obj.ctive: To explore th. feasibility of remotely obtaining complex information on traditional Ch.nese medicine (TCM) pulse conditions th.ough.voice signals. Meth.ds: We used multi-label pulse conditions as th. entry point and modeled and analyzed TCM pulse diagnosis by combining voice analysis and mach.ne learning. Audio features were extracted from voice recordings in th. TCM pulse condition dataset. Th. obtained features were combined with.information from tongue and facial diagnoses. A multi-label pulse condition voice classification DNN model was built using 10-fold cross-validation, and th. modeling meth.ds were validated using publicly available datasets. Results: Th. analysis sh.wed th.t th. proposed meth.d ach.eved an accuracy of 92.59% on th. public dataset. Th. accuracies of th. th.ee single-label pulse manifestation models in th. test set were 94.27%, 96.35%, and 95.39%. Th. absolute accuracy of th. multi-label model was 92.74%. Conclusion: Voice data analysis may serve as a remote adj.nct to th. TCM diagnostic meth.d for pulse condition assessment. © 2024 Beij.ng University of Ch.nese Medicine

Obj.ctive: To reveal th. distribution ch.racteristics and demograph.c factors of traditional Ch.nese medicine (TCM) constitution among elderly individuals in Ch.na. Meth.ds: Elderly individuals from seven regions in Ch.na were selected as samples in th.s study using a multistage cluster random sampling meth.d. Th. basic information questionnaire and Constitution in Ch.nese Medicine Questionnaire (Elderly Edition) were used. Descriptive statistical analysis, ch.-squared tests, and binary logistic regression analysis were used. Results: Th. single balanced constitution (BC) accounted for 23.9%. Th. results of th. maj.r TCM constitution types sh.wed th.t BC (43.2%) accounted for th. largest proportion and unbalanced constitutions ranged from 0.9% to 15.7%. East Ch.na region (odds ratio [OR] = 2.097; 95% confidence interval [CI], 1.912 to 2.301), married status (OR = 1.341; 95% CI, 1.235 to 1.457), and managers (OR = 1.254; 95% CI, 1.044 to 1.505) were significantly associated with.BC. Age >70 years was associated with.qi-deficiency constitution and blood stasis constitution (BSC). Female sex was significantly associated with.yang-deficiency constitution (OR = 1.646; 95% CI, 1.52 to 1.782). South.est region was significantly associated with.ph.egm-dampness constitution (OR = 1.809; 95% CI, 1.569 to 2.086). North.Ch.na region was significantly associated with.inh.rited special constitution (OR = 2.521; 95% CI, 1.569 to 4.05). South.Ch.na region (OR = 2.741; 95% CI, 1.997 to 1.3.763), Central Ch.na region (OR = 8.889; 95% CI, 6.676 to 11.835), senior middle sch.ol education (OR = 2.442; 95% CI, 1.932 to 3.088), and managers (OR = 1.804; 95% CI, 1.21 to 2.69) were significantly associated with.BSC. Conclusions: Th.s study defined th. distribution ch.racteristics and demograph.c factors of TCM constitution in th. elderly population. Obj.ctive: To investigate th. effect of Guangdong Sh.nqu (GSQ) on intestinal flora structure in mice with.food stagnation th.ough.16S rDNA sequencing. Meth.ds: Mice were randomly assigned to control, model, GSQ low-dose (GSQL), GSQ medium-dose (GSQM), GSQ h.gh.dose (GSQh., and lacidoph.lin tablets (LAB) groups, with.each.group containing 10 mice. A food stagnation and internal h.at mouse model was establish.d th.ough.intragastric administration of a mixture of beeswax and olive oil (1:15). Th. control group was administered normal saline, and th. model group was administered beeswax and olive oil to maintain a state. Th. GSQL (2 g/kg), GSQM (4 g/kg), GSQh.(8 g/kg), and LAB groups (0.625 g/kg) were administered corresponding drugs for 5 d. After administration, 16S rDNA sequencing was performed to assess gut microbiota in mouse fecal samples. Results: Th. model group exh.bited significant intestinal flora ch.nges. Following GSQ administration, th. abundance and diversity index of th. intestinal flora increased significantly, th. number of bacterial species was regulated, and α and β diversity were improved. GSQ administration increased th. abundance of probiotics, including Clostridia, Lach.ospirales, and Lactobacillus, wh.reas th. abundance of conditional path.genic bacteria, such.as Allobaculum, Erysipelotrich.ceae, and Bacteroides decreased. Functional prediction analysis indicated th.t th. path.genesis of food stagnation and GSQ intervention were primarily associated with.carboh.drate, lipid, and amino acid metabolism, among oth.r metabolic path.ays. Conclusion: Th. digestive mech.nism of GSQ may be attributed to its role in restoring diversity and abundance with.n th. intestinal flora, th.reby improving th. composition and structure of th. intestinal flora in mice and subsequently influencing th. regulation of metabolic path.ays. © 2024 Beij.ng University of Ch.nese Medicine

h.rpesviruses antagonize h.st antiviral responses th.ough.a myriad of molecular strategies culminating in th. death.of th. h.st cells. Pseudorabies virus (PRV) is a significant veterinary path.gen in pigs, causing neurological sequalae th.t ultimately lead to th. animal's demise. PRV is known to trigger apoptotic cell death.during th. late stages of infection. Th. virion h.st sh.tdown protein (Vh.) encoded by UL41 plays a crucial role in th. PRV infection process. In th.s study, we demonstrate th.t UL41 inh.bits PRV-induced activation of inflammatory cytokine and negatively regulates th. cGAS-STING-mediated antiviral activity by targeting IRF3, th.reby inh.biting th. translocation and ph.sph.rylation of IRF3. Notably, mutating th. conserved amino acid sites (E192, D194, and D195) in th. RNase domain of UL41 or knocking down UL41 inh.bits th. immune evasion of PRV, suggesting th.t UL41 may play a crucial role in PRV's evasion of th. h.st immune response during infection. Obj.ctive: To find a viable alternative to reduce th. number of doses required for th. patients with.post-traumatic stress disorder (PTSD), and to improve efficacy and patient compliance. Meth.ds: In th.s study, we used ginger oil, a ph.toch.mical with.potential th.rapeutic properties, to prepare ginger oil patch.s. h.gh.performance liquid ch.omatograph. (h.LC) was used to quantify th. main active component of ginger oil, 6-gingerol. Transdermal absorption experiments were conducted to optimize th. various pressure-sensitive adh.sives and permeation enh.ncers, including th.ir type and concentration. Subsequently, th. ginger oil patch.s were optimized and subj.cted to content determination and property evaluations. A PTSD mouse model was establish.d using th. foot-sh.ck meth.d. Th. th.rapeutic effect of ginger oil patch.s on PTSD was assessed th.ough.path.logical sections, beh.vioral tests, and th. evaluation of biomarkers such.as tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), brain-derived neurotroph.c factor (BDNF), and melatonin (MT). Results: Th. results demonstrated th.t ginger oil patch.s exerted th.rapeutic effects against PTSD by inh.biting inflammatory responses and modulating MT and BDNF levels. Ph.rmacokinetic experiments revealed th.t ginger oil patch.s maintained a stable blood drug concentration for at least one day, addressing th. rapid metabolism drawback of 6-gingerol and enh.ncing its th.rapeutic efficacy. Conclusions: Ginger oil can be prepared as a transdermal drug patch.th.t meets th.se requirements, and th. bioavailability of th. prepared patch.is better th.n th.t of oral administration. It can improve PTSD with.good patient compliance and ease of administration. Th. h.0 subtype avian influenza virus (AIV) poses an ongoing th.eat to both.birds and h.mans. Notably, fatal h.man cases of h.0N3 and h.0N8 infections h.ve drawn public attention. In 2022, we isolated two h.0N3 viruses (A/ch.cken/Sh.ndong/0101/2022 and A/ch.cken/Sh.ndong/0603/2022) from diseased ch.ckens in Ch.na. Genome analysis revealed th.t th.se viruses were genetically associated with.h.man-origin h.0N3 virus, with.internal genes originating from local h.N2 viruses. Compared to th. h.0N8 virus (A/ch.cken/j.angxi/102/2013), th. h.0N3 viruses exh.bited enh.nced th.rmostability, increased viral release from eryth.ocytes, and accumulation of h.magglutinin (h.) protein. Additionally, we evaluated th. path.genicity of both.h.0N3 and h.0N8 viruses in mice. We found th.t viral titers could be detected in th. lungs and nasal turbinates of mice infected with.th. two h.0N3 viruses, wh.reas h.0N8 virus titers were detectable in th. lungs and brains of mice. Notably, th. proportion of double h. Q222R and G228S mutations in h.0N3 viruses h.s increased since 2019. h.wever, th. functional roles of th. Q222R and G228S double mutations in th. h. gene of h.0N3 viruses remain unknown and warrant furth.r investigation. Our study h.gh.igh.s th. potential public h.alth.risk posed by th. h.0N3 virus. A spillover event of AIV to h.mans could be a foretaste of a looming pandemic.
