As an important spice species in Rutaceae, the Sichuan pepper (zanthoxylum armatum) can provide pungent and numbing taste, as well as aroma in its mature fruit. Here we assembled a chromosome-level genome of green prickly ash which was widely cultivated in a major production area including Chongqing and Sichuan province, China. We generated 712 Gb (~112×) Pacbio long reads and 511 Gb (~82×) Hi-C data, and yielded an assembly of 99 pseudochromosomes with total size of 5.32 Gb and contig N50 of 796 kb. The genomic analyses and cytogenetic experiments both indicated that the cultivarzhuye Huajiao’ was a triploid. We identified a zanthoxylum-specific whole genome duplication event emerging about 24.8 million years ago (Mya). We also detected a transposition burst event (0.3-0.4 Mya) responsible for the large genome size of z. armatum. Metabolomic analysis of the zanthoxylum fruits during development stages revealed profiles of39 volatile aroma compounds and 528 secondary metabolites, from which six types of sanshools were identified. based on metabolomic and transcriptomic network analysis, we screened candidate genes encoding long chain acyl-CoA synthetase, fatty acid desaturase,branched-chain amino acid aminotransferase involved in sanshool biosynthesis and three genes encoding terpene synthase during fruit development. The multi-omics data provide insights into the evolution of zanthoxylum and molecular basis of numbing and aroma flavor of Sichuan pepper.

Ten novel isocoumarins,including four pairs of enantiomers,were isolated from Artemisia dubia ***(Asteraceae).Compounds 1,2 and 3a/3b possessed a unique 6/6/6-tricyclic system comprising an unusual 1-(2-methylcyclohexyl)propan-1-one moiety fused with isocoumarin core *** 4a/4b were characterized as an unexpected 2,5-dimethylcyclohexan-1-one scaffold,and compounds 5a/5b and 6a/6b were rare 1,*** structures and absolute configurations were elucidated through spectroscopic data,X-ray crystallography,ECD and NMR calculations with DP4+*** biosynthetic pathways were proposed from the naturally occurring *** pharmacological analysis suggested that the targets of compound 1 were significantly enriched in the cell cycle and Pl3K-Akt signaling *** molecular docking revealed that compound 1 had high binding affinity with CDK2(total score:6.8717).Furthermore,compounds 1 and 2 exhibited inhibitory activity on three human hepatoma cell lines,with inhibitory ratios of 85.1% and 84.5%(HepG2),88.2% and 87.3%(Huh7),and 69.2% and 69.1%(SK-Hep-1)at 200μmol·L^(-1),respectively.

We propose an efficient scheme to produce ultrahigh-brightness tens of MeV electron beams by designing a density-tailored plasma to induce a wakefield in the weakly nonlinear regime with a moderate laser energy of 120 *** this scheme,the second bucket of the wakefield can have a much lower phase velocity at the steep plasma density down-ramp than the first bucket and can be exploited to implement longitudinal electron injection at a lower laser intensity,leading to the generation of bright electron beams with ultralow emittance together with low energy ***-dimensional particle-in-cell simulations are carried out and demonstrate that high-quality electron beams with a peak energy of 50 MeV,ultralow emittance of28 nm rad,energy spread of 1%,charge of 4.4 pC,and short duration less than 5 fs can be obtained within a 1-mm-long tailored plasma density,resulting in an ultrahigh six-dimensional brightness b6D,n of2×1017 A/m2/0.1%.by changing the density parameters,tunable bright electron beams with peak energies ranging from 5 to 70 MeV,a small emittance of b0.1 mm mrad,and a low energy spread at a few-percent level can be *** bright MeV-class electron beams have a variety of potential applications,for example,as ultrafast electron probes for diffraction and imaging,in laboratory astrophysics,in coherent radiation source generation,and as injectors for GeV particle accelerators.

Combining theoretical prediction and experimental exploration, the first sandwich-type polyoxovanadoborate(POVb) with an odd number of nuclei, [V_(11)b_(26)O_(73)(OH)_(10)]^(8-)(1), was discovered under hydrothermal conditions. Density-functional theory(DFT)gives a rationalization for the existence of this structure and the structural feature is consistent with the {V_Nb_(2N+X)}(X = 2, 4, 6, 8)general formula. Excitingly, when Co and 1,10-phenanthroline(phen) are introduced into the synthesis of vanadoborates system,a 100-nuclei [Co_6(phen)_(10)(H_(2)O)_8][V_(14)Co_(12)b_(72)O_(144)(OH)_(20)]·28H_(2)O(2) and a 102-nuclei [Co_6(phen)_(14)(H_(2)O)_(4)][V_(14)Co_(12)b_(72)O_(144)(OH)_(20)]·23H_(2)O(3) with the same core structure of [V_(14)Co_(12)b_(72)]^(12-)are obtained. The vase-shaped [V_(14)Co_(12)b_(72)] polyanion is a multilayered sandwich-type structure containing seven distinct structural layers and is the largest known POVb. Furthermore, 2demonstrates exceptional thermal and chemical stability and serves as an efficient and stable catalyst for the photoreduction of carbon dioxide.
