Transcriptomic analysis of epigenetic modification genes in the termite Reticulitermes speratus

Eusocial insects display a caste system in which different castes are morpho-logically and physiologically specialized for different *** studies have revealed that epigenetic modifications,including DNA methylation and histone modification,me-diate caste determination and differentiation,longevity,and polyethism in eusocial *** there has been a growing interest in the relationship between epigenetic mech-anisms and phenotypic plasticity in termites,there is ltte information about differential expression levels among castes and expression sites for these genes in *** we show royaltissuc-specific expression of epigenetic modification genes in the termite Reticulitermes *** RNA-seq,we identified 74 genes,including three DNA methyltransferases,seven sirtuins,48 Trithorax group proteins,and 16 Polycomb group *** these genes,15 showed king-specific expression,and 52 showed age-dependent differential expression in kings and *** real-time PCR revealed that DNA methyltransferase 3 is expressed specifically in the king's testis and fat body,whereas some histone modification genes are remarkably expressed in the king's testis and queen's *** findings imply that epigenetic modification plays important roles in the gamete production process in termite kings and queens.

To the Editor:Obstructive jaundice due to unresectable malignant hilar obstruction can be treated by deployment of bilateral uncovered self-expandable metal stents(UCSEMSs),which enable longer stent patency to be obtained compared with multiple plastic stents[1-3].However,re-intervention is needed if obstructive jaundice occurs after bilateral UCSEMS *** re-intervention procedures,insertion of the stent delivery system across the mesh of a UCSEMS and into the bile duct can be challenging.A novel drill dilator has recently become available in Japan(Tornus ES;Asahi Intecc,Nagoya,Japan)(Fig.1)[4].The tract is easily dilated using a clockwise *** herein describe a successful re-intervention using this device for a patient with occluded bilateral UCSEMSs.

Disruption of the TWEAK/Fn14 pathway prevents 5-fluorouracil-induced diarrhea in mice

AIM To clarify the roles of TWEAK and its receptor Fn14 in 5-fluorouracil(5-FU)-induced *** Diarrhea was induced in wild-type(WT), Fn14 knockout(KO), and IL-13 receptor(IL-13R)α1 KO BALB/c mice using a single injection of 5-FU. Histological analysis, cytokine analysis, and flow cytometry was performed on ileal tissues and cells. Murine colon carcinomabearing mice were co-treated with an anti-TWEAK antibody and 5-FU. Embryonic fibroblast response to cytokines was also ***5-FU induced high Fn14 expression in epithelial cells. The severity of 5-FU-induced diarrhea was lower in Fn14 KO mice compared with WT mice. Administration of anti-TWEAK antibody reduced 5-FU-induced diarrhea without affecting the antitumor effects of 5-FU in vivo. 5-FU-induced expression of IL-13, IL-17 A, TNF-α, and IFN-γ in the ileum was Fn14 dependent. The severity of 5-FU-induced diarrhea was lower in IL-13Rα1 KO mice, indicating major role for IL-13 signaling via IL-13Rα1 in pathogenesis. We found that IL-13Rα2, an IL-13 neutralizing/cell protective receptor, was strongly induced by IL-33 in vitro and in vivo. IL-13Rα2 was upregulated in the ileum of 5-FU-treated Fn14 KO mice. Thus, the deletion of Fn14 upregulated IL-13Rα2 expression, which reduced IL-13 expression and activity. CONCLUSION Disruption of the TWEAK/Fn14 pathway affects several interconnected pathways, including those associated with IL-13, IL-33, and IL-13Rα2, to attenuate 5-FUinduced intestinal side effects.

Infrasound observations at Syowa Station,East Antarctica:Implications for detecting the surface environmental variations in the polar regions

Characteristic infrasound waves observed at Antarctic stations demonstrate physical interaction involving environmental changes in the Antarctic continent and the surrounding oceans. A Chaparraltype infrasound sensor was installed at Syowa Station （SYO; 39°E, 69°S）, East Antarctica, as one of the projects of the International Polar Year （IPY2007-2008）. Data continuously recorded during the three seasons in 2008-2010 clearly indicate a contamination of the background oceanic signals （microbaroms） with peaks between 4 and 10 s observed during a whole season. The peak amplitudes of the microbaroms have relatively lower values during austral winters, caused by a larger amount of sea-ice extending around the Lutzow-Holm Bay near SYO, with decreasing ocean wave loading effects. Micro- baroms measurements are useful tool for characterizing ocean wave climate, complementing other oceanographic and geophysical data. A continuous monitoring by infrasound sensors in the Antarctic firmly contributes to the Comprehensive Nuclear-Test-Ban Treaty （CTBT） in the southern high latitude, together with the Pan-Antarctic Observations System （PAntOS） under the Scientific Committee on Antarctic Research （SCAR）. Detailed measurements of the infrasound waves in Antarctica, consequently, could be a new proxy for monitoring regional environmental change as well as the temporal climate variations in the polar regions.

Synthesis of CuInS_2 nanocrystals and their structural transformation triggered by ligand exchange

Chalcopyrite-type CuInS2 NCs was synthesized by the hot-soap method. Mixed solutions, CuI and InCl3 dissolved in the mixture of the tri-octylphosphite and 1-octadecene and the sulfur dissolved in tri-phenylphosphite, were used as source solutions; the hexadecylamine was additionally mixed as a surfactant before the reaction. It was observed that the product CuInS2 NCs structurally transformed from the chalcopyrite(CP)-or zincblende(ZB)-to the wurtzite(WZ)-type depending on the amount of the surfactant and a storage time after the surfactant addition. Very weak photoluminescence in the near-infrared region was observed for the CP-or ZB-type NCs. This band was attributable to the electronhole recombination via defect levels. No photoluminescence was detected for the wurtzite-type NCs.

Knockdown of Dock7 <i>in vivo</i>specifically affects myelination by Schwann cells and increases myelin thickness in sciatic nerves without affecting axon thickness

During development of the peripheral nervous system (PNS), Schwann cells (SCs) wrap individual axons to form myelin sheaths, which act as surrounding insulators and markedly enhance the propagation of the action potential. In peripheral neuropathies such as Guillain-Barré syndrome (GBS) and inherited demyelinating Charcot-Marie-Tooth (CMT) disease and diabetic neuropathies, chronic demyelination and defective remyelination are repeated, causing more severe neuropathies. It is thus thought that development of a drug that promotes proper myelination with minimal side effects could provide an effective therapy for these diseases. As yet, however, little is known about therapeutic target molecules and genetically-modified mice for testing such approaches. We previously cloned the dock7 gene and characterized Dock7 as the regulator controlling SC myelination;however, an important issue, whether knockdown of Dock7 specifically affects myelination by SCs but not leaves neurons unaffected, has remained unclear. Here, we generate newly-produced transgenic mice harboring short-hairpin RNA (shRNA) targeting Dock7. We also describe that Dock7 shRNA transgenic mice exhibit enhanced myelin thickness without affecting axon thickness in sciatic nerves of the PNS, as reduced thickness of the axon diameter is the primary indicator of denatured neurons. Similarly, purified in vitro SC-neuronal cocultures established from transgenic mice exhibit enhanced formation of myelin segments, suggesting that knockdown of Dock7 promotes myelination by SCs. Collectively, Dock7 knockdown specifically affects SC myelination in sciatic nerves, providing evidence that Dock7 may be a promising drug-target-specific molecules for developing a therapy for peripheral neuropathies that aims to enhance myeliantion.

Length-Sorted Semiconducting Carbon Nanotubes for High-Mobility Thin Film Transistors

We have developed a process for chemical purification of carbon nanotubes for solution-processable thin-film transistors （TFTs） having high mobility. Films of the purified carbon nanotubes fabricated by simple drop coating showed carrier mobilities as high as 164 cm2V-1s -1 normalized transconductances of 0.78 Sm-1 and on/off current ratios of 10^6. Such high performance requires the preparation of a suspension of micrometer-long and highly purified semiconducting single-walled carbon nanotubes （SWCNTs）. Our purification process includes length and electronic-type selective trapping of SWCNTs using recycling gel filtration with a mixture of surfactants. The results provide an important milestone toward printed high-speed and large-area electronics with roll-to-roll and ink-jet device fabrication.

The Keras deep learning framework is employed to study MRI brain data in a preliminary analysis of brain structure using a convolutional neural *** results obtained are matched with the content of personality *** Big Five personality traits provide easy differentiation for dividing personalities into different *** now,the highest accuracy obtained from the results of personality prediction from the analysis of brain structure is about 70%.Although there is still no effective evidence to prove a clear relationship between brain structure and personality,the obtained results could prove helpful in understanding the basic relationship between brain structure and personality characteristics.
