Design and test of the plant-correcting reel for harvesting lodging garlic plants

Crops are prone to lodging with the decline of stem moisture and the intervention of other factors in the mature harvest period,such as garlic,which is difficult to harvest mechanically.To solve this problem,the plant-correcting reel for harvesting lodging garlic plants,bumped and deformed with plants many times to pull and lift them into a conveyor,is proposed in this study.We analyzed the motion trajectory equation and key influencing factors of the reel and defined the lifting and plant-correcting stages as three processes of contact,stirring and release.For example,the contact deformation model and system energy equations were established in the contact process.Besides,in the stirring process,the garlic plant-correcting conditions were established through the dynamic simulation test analysis of garlic seedling trajectories and the deflection model of garlic stem was constructed.Furthermore,in the release process,the expressions of rubber bars rotation and garlic plant offset bending curvature were constructed and the optimal number and distribution form of bars were determined.Meanwhile,the mechanism and key operating parameters of the auxiliary lifting mechanism of the divider were established.Through the single-factor test,the influence of reel speed,forward speed and reel height on the success feeding rate was analyzed under different bars distribution forms;Through multi-factor experiments,the interaction contour map of various factors was constructed.When reel speed,forward speed and reel height were 3 rad/s,3.5 m/s,and 540 mm,the feeding success rate was 98.73%.The optimization factors were tested and verified,which met the operational requirements of a high feeding success rate and low loss rate of garlic harvest.This study combines laboratory virtual as well as field experiments and analyzes of trajectory of bars,contact deformation and deflection model of garlic plant,and reel rotation and garlic plant offset bending curvature to solve the problem of garlic lodging mechanized harvest and yield reduct

This paper offers an extensive overview of the utilization of sequential approximate optimization approaches in the context of numerically simulated large-scale continuum structures.These structures,commonly encountered in engineering applications,often involve complex objective and constraint functions that cannot be readily expressed as explicit functions of the design variables.As a result,sequential approximation techniques have emerged as the preferred strategy for addressing a wide array of topology optimization challenges.Over the past several decades,topology optimization methods have been advanced remarkably and successfully applied to solve engineering problems incorporating diverse physical backgrounds.In comparison to the large-scale equation solution,sensitivity analysis,graphics post-processing,etc.,the progress of the sequential approximation functions and their corresponding optimizersmake sluggish progress.Researchers,particularly novices,pay special attention to their difficulties with a particular problem.Thus,this paper provides an overview of sequential approximation functions,related literature on topology optimization methods,and their applications.Starting from optimality criteria and sequential linear programming,the other sequential approximate optimizations are introduced by employing Taylor expansion and intervening variables.In addition,recent advancements have led to the emergence of approaches such as Augmented Lagrange,sequential approximate integer,and non-gradient approximation are also introduced.By highlighting real-world applications and case studies,the paper not only demonstrates the practical relevance of these methods but also underscores the need for continued exploration in this area.Furthermore,to provide a comprehensive overview,this paper offers several novel developments that aim to illuminate potential directions for future research.

Phosphatase and tensin homolog(PTEN)is a multifunctional gene involved in a variety of physiological and pathological processes.Circular RNAs(circRNAs)are generated from back-splicing events during mRNA processing and participate in cell biological processes through binding to RNAs or proteins.However,PTEN-related circRNAs are largely unknown.Here,we report that circPTEN-mitochondria(MT)(hsa_circ_0002934)is a circular RNA encoded by exons 3,4,and 5 of PTEN and is a critical regulator of mitochondrial energy metabolism.CircPTEN-MT is localized to mitochondria and physically associated with leucine-rich pentatricopeptide repeat-containing protein(LRPPRC),which regulates posttranscriptional gene expression in mitochondria.Knocking down circPTEN-MT reduces the interaction of LRPPRC and steroid receptor RNA activator(SRA)stem-loop interacting RNA binding protein(SliRP)and inhibits the polyadenylation of mitochondrial mRNA,which decreases the mRNA level of the mitochondrial complex I subunit and reduces mitochondrial membrane potential and adenosine triphosphate production.Our data demonstrate that circPTEN-MT is an important regulator of cellular energy metabolism.This study expands our understanding of the role of PTEN,which produces both linear and circular RNAs with different and independent functions.

Background:Sepsis is a systemic inflammatory syndrome induced by several infectious agents.Multiple organs are affected by sepsis,including the liver,which plays an important role in metabolism and immune homeostasis.Fibroblast growth factors(FGFs)participate in several biological processes,although the role of FGF5 in sepsis is unclear.Methods:In this study,lipopolysaccharide(LPS)was administrated to mice to establish a sepsis-induced liver injury.A similar in vitro study was conducted using L-02 hepatocytes.Western blot and immunohistochemistry staining were performed to evaluate the FGF5 expression level in liver tissues and cells.Inflammatory cell infiltrations,cleaved-caspase-3 expressions,reactive oxygen species and levels of inflammatory cytokines were detected by immunofluorescence,dihydroethidium staining,and reverse transcription quantitative polymerase chain reaction analysis,respectively.Flow cytometry was used to detect the apoptosis level of cells.In addition,ribonucleic acid(RNA)-sequencing was applied to explore the possible mechanism by which FGF5 exerted effects.Results:LPS administration caused FGF5 down-regulation in the mouse liver as well as in L-02 hepatocytes.Additionally,with FGF5 overexpression,liver injury and the level of hepatocyte apoptosis were ameliorated.Further,RNA sequencing performed in hepatocytes revealed the phosphoinositide-3-kinase/protein kinase B(PI3K/AKT)pathway as a possible pathway regulated by FGF5.This was supported using an inhibitor of the PI3K/AKT pathway,which abrogated the protective effect of FGF5 in LPSinduced hepatocyte injury.Conclusion:The anti-apoptotic effect of FGF5 on hepatocytes suffering from LPS has been demonstrated and was dependent on the activation of the PI3K/AKT signaling pathway.

Dear Editor An optimal vaccination program that can elicit Iong-lasting humoral and cellular immune response is key to Iong-term prevention of coronavirus disease 2019(COVID-19).Multiple variants of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)continue to emerge.The neutralizing capacities of available COVID-19 vaccines against these variants have declined.^(1) In such a scenario,heterologous prime-boost programs for COVID-19 vaccines have attracted considerable interest.

To date,the coronavirus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has determined 399,600,607 cases and 5,757,562 deaths worldwide.COVID-19 is a serious threat to human health globally.The World Health Organization(WHO)has declared COVID-19 pandemic a major public health emergency.Vaccination is the most effective and economical intervention for controlling the spread of epidemics,and consequently saving lives and protecting the health of the population.Various techniques have been employed in the development of COVID-19 vaccines.Among these,the COVID-19 messenger RNA(mRNA)vaccine has been drawing increasing attention owing to its great application prospects and advantages,which include short development cycle,easy industrialization,simple production process,flexibility to respond to new variants,and the capacity to induce better immune response.This review summarizes current knowledge on the structural characteristics,antigen design strategies,delivery systems,industrialization potential,quality control,latest clinical trials and real-world data of COVID-19 mRNA vaccines as well as mRNA technology.Current challenges and future directions in the development of preventive mRNA vaccines for major infectious diseases are also discussed.

Design and validation of a navigation system of multimodal medical images for neurosurgery based on mixed reality

Purpose: This paper aims to develop a navigation system based on mixed reality, which can displaymultimodal medical images in an immersive environment and help surgeons locate the target areaand surrounding important tissues precisely.Methods: To be displayed properly in mixed reality, medical images are processed in this system.High-quality cerebral vessels and nerve fibers with proper colors are reconstructed and exported tomixed reality environment. Multimodal images and models are registered and fused, extracting theirkey information. The multiple processed images are fused with the real patient in the same coordinatesystem to guide the surgery.Results: The multimodal image system is designed and validated properly. In phantom experiments,the average error of preoperative registration is 1.003 mm and the standard deviation is 0.096 mm.The average proportion of well-registered areas is 94.9%. In patient experiments, the surgeons whoparticipated in the experiments generally indicated that the system had excellent performance andgreat application prospect for neurosurgery.Conclusion: This article proposes a navigation system of multimodal images for neurosurgery basedon mixed reality. Compared with other navigation methods, this system can help surgeons locate thetarget area and surrounding important tissues more precisely and rapidly.

A Novel Feature Aggregation Approach for Image Retrieval Using Local and Global Features

The current deep convolution features based on retrievalmethods cannot fully use the characteristics of the salient image regions.Also,they cannot effectively suppress the background noises,so it is a challenging task to retrieve objects in cluttered scenarios.To solve the problem,we propose a new image retrieval method that employs a novel feature aggregation approach with an attention mechanism and utilizes a combination of local and global features.The method first extracts global and local features of the input image and then selects keypoints from local features by using the attention mechanism.After that,the feature aggregation mechanism aggregates the keypoints to a compact vector representation according to the scores evaluated by the attention mechanism.The core of the aggregation mechanism is to allow features with high scores to participate in residual operations of all cluster centers.Finally,we get the improved image representation by fusing aggregated feature descriptor and global feature of the input image.To effectively evaluate the proposedmethod,we have carried out a series of experiments on large-scale image datasets and compared them with other state-of-the-art methods.Experiments show that this method greatly improves the precision of image retrieval and computational efficiency.

PuHox52 promotes coordinated uptake of nitrate,phosphate, and iron under nitrogen deficiency in Populus ussuriensis

It is of great importance to better understand how trees regulate nitrogen(N) uptake under N deficiency conditions which severely challenge afforestation practices, yet the underlying molecular mechanisms have not been well elucidated. Here,we functionally characterized PuHox52, a Populus ussuriensis HD-ZIP transcription factor, whose overexpression greatly enhanced nutrient uptake and plant growth under N deficiency. We first conducted an RNA sequencing experiment to obtain root transcriptome using PuHox52-overexpression lines of P. ussuriensis under low N treatment. We then performed multiple genetic and phenotypic analyses to identify key target genes of PuHox52 and validated how they acted against N deficiency under PuHox52 regulation.PuHox52 was specifically induced in roots by N deficiency, and overexpression of PuHox52promoted N uptake, plant growth, and root development. We demonstrated that several nitrate-responsive genes(PuNRT1.1, PuNRT2.4,PuCLC-b, PuNIA2, PuNIR1, and PuNLP1),phosphate-responsive genes(PuPHL1A and PuPHL1B), and an iron transporter gene(PuIRT1) were substantiated to be direct targets of PuHox52. Among them, PuNRT1.1, PuPHL1A/B, and PuIRT1 were upregulated to relatively higher levels during PuHox52-mediated responses against N deficiency in PuHox52-overexpression lines compared to WT. Our study revealed a novel regulatory mechanism underlying root adaption to N deficiency where PuHox52 modulated a coordinated uptake of nitrate, phosphate, and iron through 'PuHox52-PuNRT1.1', 'PuHox52-PuPHL1A/PuPHL1B', and'PuHox52-PuIRT1' regulatory relationships in poplar roots.

Sentiment analysis using deep learning approaches:an overview

Nowadays, with the increasing number of Web 2.0 tools, users generate huge amounts of data in an enormous and dynamic way. In this regard, the sentiment analysis appeared to be an important tool that allows the automation of getting insight from the user-generated data. Recently, deep learning approaches have been proposed for different sentiment analysis tasks and have achieved state-of-the-art results. Therefore, in order to help researchers to depict quickly the current progress as well as current issues to be addressed, in this paper, we review deep learning approaches that have been applied to various sentiment analysis tasks and their trends of development. This study also provides the performance analysis of different deep learning models on a particular dataset at the end of each sentiment analysis task. Toward the end, the review highlights current issues and hypothesized solutions to be taken into account in future work. Moreover, based on knowledge learned from previous studies, the future work subsection shows the suggestions that can be incorporated into new deep learning models to yield better performance. Suggestions include the use of bidirectional encoder representations from transformers(BERT), sentiment-specific word embedding models, cognition-based attention models, common sense knowledge, reinforcement learning, and generative adversarial networks.
