The chromosomal position effect can significantly affect the transgene expression,which may provide an efficient strategy for the inauguration of alien genes in new hosts,but has been less explored *** bacterium Myxococcus xanthus harbors a large circular high-GC genome,and the position effect in this chassis may result in a thousand-fold expression variation of alien natural *** this study,we conducted transposon insertion at TA sites on the *** genome,and used enrichment and dilution indexes to respectively appraise high and low expression potentials of alien genes at insertion *** enrichment sites are characteristically distributed along the genome,and the dilution sites are overlapped well with the horizontal transfer *** experimentally demonstrated the enrichment sites as high expression integration sites(HEISs),and the dilution sites unsuitable for gene integration *** work highlights that HEISs are the plug-and-play sites for efficient expression of integrated genes.

The flocculation and the settling of the cohesive sediment play an important role in the sediment transport process,and they are affected by many *** the coasts and in the estuaries,the organic matter and the salinity are two significant *** this study,experiments are carried out to evaluate the flocculation and the settling of the kaolinite in cases of various organic matter and salinity solutions in quiescent *** optical multi-channel suspended concentration monitoring system is used to measure the sediment concentration and the floc *** results show that the settling velocity decreases with the increase of the organic matter content,and the floc size increases with the increase of the organic matter *** addition,an empirical formula for the settling velocity under the influence of the organic matter and the salinity based on the experimental results is proposed.

Tetrathiafulvalene（TTF） was doped in an SiO2 network and the resulting nanocompesite was used as a mediator for the selective detection of glucose. The uniform TTF-doped silica（TIT@SiO2 ） nanoparticles were prepared by the water-in-oil（W/O） microemulsion method, and were characterized by transmission electron microscopy（TEM）. The core-shell structure TTF@ SiO2 could prevent TIT from leaching out into an aqueous solution. Combined with chitosan （CHIT）, which serves as a scaffold for glucose oxidase and nanocomposite immobilization, the GCE/TTF@ SiO2- CHIT-GOx biosensor was developed. Under optimal conditions, the biosensors exhibit a linear range of 1.0 × 10^-5 5 × 10^-3 mol/L with a detection limit down to 5.0 μmol/L（S/N = 3 ）. The excellent selectivity, sensitivity, and stability of the glucose biosensor show its potential for practical applications.

Based on an unusual reversible sol-gel transition phenomenon. a novel method for the fabrication of a lipase electrode was developed. The response characteristics of the biosensor was studied by potentiometric technique using olive oil as substrate. After optimization, the lipase electrode demonstrated high activity and good stability.

AIM: To establish a high-throughput model for screening anti-tumor agents capable of promoting the polymeriza- tion of tubulin in vitro. METHODS: Tubulin was prepared in different purity for two screening steps. The first step was a high-throughput screening (HTS) for a set of 1500 samples using the GTP-containing tubulin and the end-reading method. The second step was performed on 119 hits from the first screening by a kinetic assay with GTP-lacking tubulin. RESULTS: The HTS for 1500 samples was accomplished in less than 3 h. From the screening, 108 samples were identified with >20 % promotion activity at 10 mg/L. Five of 108 were further confirmed by the kinetic assay using the purified tubulin subsequently. Three of the hit compounds were Epothilone A or its analogs, the other two compounds had new structures with a common pharmacophore for cytotoxic natural products that stabilize microtubules. In an MTT test, the five selected samples from the screening showed a minimal IC50 at0.28±0.06 nmol/Lto Helacells. CONCLUTION: The two-stepscreening methodis a high-throughtput, cost-effective, and efficient approach to identify microtubule-stabilizing agents.

Natural swarms arranged from cells to herds are usually decentralized but display intriguing collective intelligence in coordinating individuals across large scales to efficiently achieve their common goals. Learning from nature may provide new strategies for controlling collective dynamics of synthetic swarms to accomplish specific functions. Here, we present a bioinspired computational framework that steers distributed active swarms to collectively capture and merge targets via reinforcement learning. We exploit collective milling structures of natural herds to cage the targets, and adopt a switching control policy inspired by sperms' chiral dynamics to optimize the trajectories of individuals, through which the active swarms can selforganize to enclose single or multiple distant targets in a dynamical, adaptive and scalable manner. There exists a critical swarm size, beyond which the excessive competition between agents would generate large mechanical forces, leading to capture instability but enabling the transition from short-distance to long-distance merging capture of multiple targets. This work provides physical insights into distributed active swarms and could offer a multilevel, decentralized strategy toward controlling swarm robotics in wide applications such as bio-medical devices, machine immunity, and target clearance.

针对历史街区活力衰退的现象,基于兴趣点(point of interest,POI)数据与公共空间-公共生活(public space-public life,PSPL)传统调研法,运用大数据的点密度分析与传统调研数据的定性、定量分析方法,从宏观与微观两个层面深入地研究青岛市大鲍岛历史街区公共空间的活力特征。研究发现公共空间的衰败、现代生活元素的缺失、与周边活力空间不协调是大鲍岛历史街区活力衰退的主要原因。结合案例分析与自身历史文化属性,从大鲍岛历史街区的物质文化空间、文化功能空间和文化服务空间三大方面提出具体的活力提升策略。研究表明,宏观+微观层面的研究视角、新技术+传统调研的研究方法、定性+定量的数据分析方法相结合,能够全面而具体地研究历史街区公共空间的活力,也可为其他历史街区相关研究提供思路。

The durability of wear-resistant tetrahedral amorphous carbon（ta-C） coatings is intimately linked to their thickness and the concentration of *** enhance the lifespan of the coatings,we have prepared a multilayer coating consisting of alternating layers of Ti and ta-C,denoted as（Ti/ta-C）_n,using arc ion plating.A multi-layer structure in conjunction with stage cooling is employed to enhance the thickness and promote *** results indicate that the ID/IGvalue,representing the ratio of the intensity of the D peak to that of the G peak in Raman spectroscopy,decreases from 0.85 when no stage cooling is applied to 0.35 when stage cooling is employed,suggesting an enhancement in the fraction of sp3bonds within the *** obtained（Ti/ta-C）_n multilayer coating without peeling off has a maximum thickness of 1.14 μm,which is almost double that of the ta-C coating single layer *** the same time,the coating has a low coefficient of friction,down to 0.069,and maintains excellent friction properties with a wear rate of 3.4×10-6mm3·N-1·*** results conclusively show that the preparation technology can enhance both thickness and sp3bonding in ta-C coatings,significantly expanding their utility across tools,molds,and precision engineering components.
