Rab5对禽偏肺病毒C型复制影响的研究

为探究禽偏肺病毒C型(aMPV/C)感染A549细胞后,脑内Ras类似物5(Rab5)对病毒复制的影响,本研究将重组质粒p GFP-Rab5和负显性突变的Rab5(pGFP-Rab5-DN)分别转染A549细胞,24 h后分别以MOI 1的a MPV/C感染该细胞,48 h后进行激光共聚焦显微镜观察,结果显示Rab5、Rab5-DN和aMPV/C N蛋白均分布于细胞质中,且Rab5与aMPV/C N蛋白存在明显的共定位。进一步采用Image J软件对细胞内的荧光强度分析后发现,Rab5与aMPV/C N蛋白存在多处共定位,而Rab5-DN与aMPV/C N蛋白无明显共定位,表明Rab5参与aMPV/C的复制。将p CMV-Flag-Rab5和p CMV-Flag分别转染A549细胞,24 h后分别以MOI 1的aMPV/C感染该细胞,48 h后分别收集上清和细胞样品。分别采用western blot检测Rab5及aMPV/C N蛋白的表达水平、荧光定量PCR(qPCR)检测a MPV/C N基因转录水平、TCID_(50)检测细胞上清的病毒效价。结果显示,与转染p CMV-Flag的细胞相比,转染p CMV-Flag-Rab5的细胞中Rab5的表达水平明显升高;此外,p CMV-Flag-Rab5转染组中的aMPV/C N蛋白表达水平和N基因的转录水平均极显著升高(P<0.01),病毒效价显著升高(P<0.05)。表明,过表达Rab5促进aMPV/C的复制。将siRab5和siNC分别转染A549细胞,24 h后分别以MOI 1的aMPV/C感染该细胞,48 h后分别收集上清和细胞样品。分别采用western blot检测Rab5及aMPV/C N蛋白的表达水平、q PCR检测aMPV/C N基因的转录水平、TCID_(50)检测细胞上清的病毒效价。结果显示,与转染si NC的细胞相比,转染siRab5的细胞中Rab5的表达水平明显降低;同时,siRab5转染组中aMPV/C N蛋白的表达水平和N基因的转录水平均极显著降低(P<0.01),病毒效价显著降低(P<0.05)。表明,干扰Rab5表达可抑制aMPV/C的复制。将p CMV-mcherry-Rab5与表达融合GFP的aMPV/C结构蛋白的重组质粒分别转染A549细胞,24 h后采用激光共聚焦显微镜观察发现Rab5蛋白与aMPV/C N、F、M、G、SH、P、M2-1、M2-2、L1、L2、L3蛋白均在细胞质中存在荧光信号重叠,但该蛋白与L4蛋白无该现象,表明Rab5蛋白可与多个aMPV/C蛋白共定位。将p CMV-Flag-Rab5分别与表达融合GFP的aMPV/C结构蛋白的重组质粒共转染HEK293T细胞,48 h后采用Co-IP试验检测Rab5与不同aMPV/C结构蛋白的相互作用。结果显示,IP样品中表达G和L2蛋白的位置出现了特异性条带,而其他蛋白无条带,表明Rab5与aMPV/C G和L2蛋白存在互作。上述结果首次证实,Rab5通过与aMPV/C多个蛋白(G和L2)的互作参与aMPV/C的复制机制。本研究为阐明Rab5调控aMPV/C复制的分子机理提供研究借鉴。

Machine learning, particularly deep learning, has achieved remarkable success across a wide range of tasks. However,most of these tasks demand a substantial amount of labeled training data, which can be challenging to obtain in many real-world applications due to difficulties, costs, or time constraints associated with labeling. In this context,semi-supervised learning (SSL) has emerged as a promising paradigm to alleviate the dependency on large labeled datasets by harnessing the power of unlabeled data [1–3].

Low desulfurization efficiency impedes the wide application of dry desulfurization technology,which is a low-cost and simple process,and one significant solution is the development and manufacture of high-performance *** this study,firstly,a steam jet mill was used to digest quicklime;then,we utilized numerical simulation to study the flow field distribution and analyze the driving factors of quicklime digestion;and lastly,the desulfurization performance of the desulfurizer was evaluated under different relative *** results show that the desulfurizer prepared via the steam jet mill had better apparent activity than traditional ***,the entire jet flow field of the steam jet mill is in a supersonic and highly turbulent flow state,with high crushing intensity and good particle acceleration *** contact with the nascent surface maximizes the formation of slaked *** experiments demonstrated that the operating time with 100%desulfurization efficiency and the“break-through”time for the desulfurizer prepared via the steam jet mill is longer than that of traditional desulfurizers,and has significant advantages,especially at low flue gas relative *** with traditional desulfurizers,the desulfurizer prepared via steam jet mill expands the range of acceptable flue gas temperature,and the failure temperature is 1.625 times that of traditional *** work breaks through the technical bottleneck of low dry desulfurization efficiency,which is an important step in pushing forward the application of dry desulfurization.

The continued submission of high-quality contributions from these community developers has led to numerous corrections, improvements, and innovations in many successful OSS projects [1, 2].

This study aims to revise the Belief in a Just World Scale(BJWS)for Chinese college students and test its reliability and validity(construct validity,convergent and divergent validity).Two samples of 546 and 595 college students were selected,respectively,using stratified cluster random *** analysis,exploratory factor analysis(EFA),confirmatory factor analysis(CFA),reliability analysis and convergent and divergent validity tests were carried *** results showed that the 13 items of the BJWS have good item *** corrected item–total correlation in the general belief in a just world subscale was found to range from 0.464 to 0.655,and that in the personal belief in a just world subscale was 0.553 to *** internal consistency coefficients of the revised version of the BJWS and its subscales are *** EFA and CFA results show that the structure and items of the revised scale are the same as those of the original *** in a just world was found to have significant positive correlations with gratitude and empathy,and has a significant negative correlation with anxiety,thereby exhibiting good convergent and divergent ***,the Chinese revised version of the BJWS has good reliability and validity.

Multichromophoric architectures with wellorganized chromophoric arrangements are of vital importance to gain insights into interchromophoric ***,we report a novel heteroleptic self-assembly strategy and construct three achiral and one chiral sandwich-like heterochromophore complexes(Zn_(4)L^(G)L^(A-C)_(2)and Zn_(4)L_(G)L^(D)_(2)).Possible dynamic products,including homoleptic oligomers and isomers,are eliminated,producing achiral complexes with exceptional *** are observed in chiral complexes due to the flexible geometry of *** aligned chromophore arrangements with precise spacing and orientations are evidenced by single-crystal X-ray *** distinct segments in the ligands lead to different interchromophoric ***,energy transfer and tunable emission,including white-light emission,are achieved for Zn_(4)L^(G)L^(A)_(2)and Zn_(4)L^(G)L^(B)_(2).In addition,chirality transfer is observed for Zn_(4)L_(G)L^(D)_(2),due to conformational restriction from chiral LD,leading to chiral rotational conformations of tetraphenylethylene moiety in LG.
