Bioactives and extracts affect the physico-chemical properties of concentrated whey protein isolate dispersions

Non-covalent complexation interactions are known to occur between bioactive compounds and proteins. While formulating with these components can have positive outcomes such as stabilization of colors and actives, it can also result in changes to the structures and physical properties of proteins, affecting product functionality and sensory attributes. Previous experiments reported measurable changes in the physico-chemical properties of whey protein isolate (WPI) dispersions upon formulation with Aronia berry extract, ascribing changes to protein-polyphenol (PP) interactions in the systems. Pure gallotannin, beet extract, and cranberry extract, providing a diverse variety structures and sizes, were selected for further experimentation and comparison with the effects of Aronia extract. Concentrated dispersions with varying WPI:sucrose ratios, formulated with several bioactives contents from multiple different sources were analyzed to identify the effects of different bioactives on physico-chemical properties of dispersions. Dispersions formulated with cranberry extract demonstrated the largest increases in surface tensions, viscosities, and particle sizes, while those formulated with beet extract were the least affected by the presence of bioactives, suggesting that different bioactives and extracts had varying propensities for complexation interactions with WPI, despite their relatively low levels of addition (0, 0.5, and 1%).

Bioactive compounds including polyphenols(PP)have been observed to naturally form non-covalent complexation interactions with proteins under mild ph and temperature conditions,affecting protein structures and ***,addition of Aronia berry PP to liquid dispersions containing whey protein isolate(WPI)and sucrose was found to alter characteristics including viscosity,surface tension,and particle sizes,with changes being attributed to protein-PP *** this study we aimed to investigate whether Aronia PP would interact with soy and pea protein isolates(SPI and PPI,respectively)to a similar extent as with WPI in liquid protein-sucrose-PP *** hypothesized that formulations containing PPI(comprised of larger proteins)and hydrolyzed SPI(containing more carboxyl groups)may exhibit increased viscosities and decreased aggregate sizes due to enhanced protein-PP *** liquid dispersions of varied ratios of protein to sucrose contents,containing different protein isolates(WPI,SPI,and PPI),and varied Aronia PP concentrations were formulated,and physical properties were evaluated to elucidate the effects of PP *** addition altered physical characteristics differently depending on the protein isolate used,with changes attributed to protein-PP *** and PPI appeared to have higher propensities for PP interactions and exhibited more extensive shifts in physical properties than WPI *** findings may be useful for practical applications such as formulating products containing fruit and proteins to obtain desirable sensory attributes.

Since 2014,"Bring Your Own Data"workshops(BYODs)have been organised to inform people about the process and benefits of making resources Findable,Accessible,Interoperable,and Reusable(FAIR,and the FAIRification process).The BYOD workshops'content and format differ depending on their goal,context,and the background and needs of ***-focused BYODs educate domain experts on how to make their data FAIR to find new answers to research ***-focused BYODs promote the benefits of making data FAIR and instruct project managers and policy-makers on the characteristics of FAIRification ***-focused BYODs gather software developers and experts on FAIR to implement or improve software resources that are used to support ***,these BYODs intend to foster collaboration between different types of stakeholders involved in data management,curation,and reuse(*** experts,trainers,developers,data owners,data analysts,FAIR experts).The BYODs also serve as an opportunity to learn what kind of support for FAIRification is needed from different communities and to develop teaching materials based on practical examples and *** this paper,we detail the three different structures of the BYODs and describe examples of early BYODs related to plant breeding data,and rare disease registries and biobanks,which have shaped the structure of the *** discuss the latest insights into making BYODs more productive by leveraging our almost ten years of training experience in these workshops,including successes and encountered ***,we examine how the participants'feedback has motivated the research on FAIR,including the development of workflows and software.

Anoctamin-5(ANO5)is a multi-pass membrane protein localized to the sarcolemma and the sarcoplasmic *** were linked to rare autosomal recessive muscle ***,we summarize the clinical spectrum,imaging data and molecular research findings as well as results of animal modeling,which significantly moved forward the understanding of mechanisms underlying ANO5-related muscle *** that precise histological information on inflammatory processes taking place in patient-derived muscle are still lacking,an(immuno)histological study on biopsies derived from six ANO5-patients was performed showing focal accumulation of necrotic fibers,mild fiber-size variances and *** addition,MRI data of four ANO5-patients(including a 10-year follow-up in one patient)are presented and discussed in the context of previously published ***,data presented in this article combining a review of the literature with own myopathological findings address scientific trends and open questions on ANO5-related muscle diseases,which would be of significant interest for a wide neuromuscular diseases *** conclude,a clear genotypeephenotype correlation does not exist,and ANO5-related muscle disorders might represent the next entity of a clinical continuum with varying degree of muscle cell *** addition,results of pre-clinical studies allowed the definition of suitable cell and animal models characterized by certain histological and functional pathologies resembling the human *** models might serve as suitable systems for testing of interventional concepts in future.

UNIFORM SUPERCONVERGENCE OF A FINITE ELEMENT METhOD WITh EDGE STABILIZATION FOR CONVECTION-DIFFUSION PROBLEMS

In the present paper the edge stabilization technique is applied to a convection-diffusion problem with exponential boundary layers on the unit square, using a Shishkin mesh with bilinear finite elements in the layer regions and linear elements on the coarse part of the mesh. An error bound is proved for ‖πu-u^h‖Е, where πu is some interpolant of the solution u and uh the discrete solution. This supercloseness result implies an optimal error estimate with respect to the L2 norm and opens the door to the application of postprocessing for improving the discrete solution.

Objectives:To test for differences in overall and recurrence-free survival between laparoscopic and open surgical approaches in patients undergoing radical nephroureterectomy(RNU)for upper tract urothelial carcinoma(UTUC).Materials and methods:We retrospectively identified patients treated for UTUC from 2010 to 2020 from our institutional *** undergoing laparoscopic or open RNU with no suspicion of metastasis(cM0)were for the current study *** with suspected metastases at diagnosis(cM1)or those undergoing other surgical treatments were *** was performed according to the laparoscopic versus open surgical ***-Meier plots were used to test for differences in overall and recurrence-free survival with regard to the surgical ***,separate Kaplan-Meier plots were used to test the effect of preoperative ureterorenoscopy on overall and recurrence-free survival within the overall study ***:Of the 59 patients who underwent nephroureterectomy,29%(n=17)underwent laparoscopic nephroureterectomy,whereas 71%(n=42)underwent open *** and tumor characteristics were comparable between groups(p≥0.2).The median overall survival was 93 and 73 months in the laparoscopic nephroureterectomy group compared to the open nephroureterectomy group(p=0.5),*** median recurrence-free survival did not differ between open and laparoscopic nephroureterectomies(73 months for both groups;p=0.9).Furthermore,the median overall and recurrence-free survival rates did not differ between patients treated with and without preoperative ***:The results of this retrospective,single-center institution showed that overall and recurrence-free survival rates did not differ between patients with UTUC treated with laparoscopic and open ***,preoperative ureterorenoscopy before RNU was not associated with higher overall or recurrence-free survival rates.

Making FAIR Easy with FAIR Tools:From Creolization to Convergence

Since their publication in 2016 we have seen a rapid adoption of the FAIR principles in many scientific disciplines where the inherent value of research data and,therefore,the importance of good data management and data stewardship,is *** has led to many communities asking“What is FAIR?”and“how FAIR are we currently?”,questions which were addressed respectively by a publication revisiting the principles and the emergence of FAIR ***,early adopters of the FAIR principles have already run into the next question:“how can we become(more)FAIR?”This question is more difficult to answer,as the principles do not prescribe any specific standard or ***,there does not yet exist a mature ecosystem of tools,platforms and standards to support human and machine agents to manage,produce,publish and consume FAIR data in a user-friendly and efficient(i.e.,“easy”)*** this paper we will show,however,that there are already many emerging examples of FAIR tools under *** paper puts forward the position that we are likely already in a creolization phase where FAIR tools and technologies are merging and combining,before converging in a subsequent phase to solutions that make FAIR feasible in daily practice.

Effect of using ascorbic acid and cysteamine supplementation onin-vitrodevelopment of buffalo embryos

Objective:To improvein vitro embryo production in buffalo by supplementation of L-ascorbic acid during maturation and development (experiment 1) and combination with another antioxidant as cysteamine (experiment 2).Methods:Two experiments were performed, the first one aimed to evaluate the different concentrations (0, 25, 50, 100 μM) of L-ascorbic acid on embryo developmental rate of buffalo oocytes. The L-ascorbic acid was added to the maturation and culture media. In the second experiment, oocytes were cultured in media with two type of antioxidant (ascorbic acid + cysteamine) or ascorbic acid ***:There was a significant increase in cleavage rate at 25, 50 μM than 100 μM and control group. But, the blastocyst rate was higher at 50 μM ascorbic acid than other concentrations (0, 25, 100 μM). Supplementation of ascorbic acid and cysteamine to maturation and cultured media improved embryo development than ascorbic acid ***: Using of 50 μM L-ascorbic acid duringin vitro maturation and development improve the developmental competence of buffalo oocytes, this effect was increase with the presence of cysteamine.
