Fructose aggravating colon barrier dysfunction by decreasing gut bacteria metabolites indole-3-carboxaldehyde and inhibiting activation of aryl hydrocarbon receptor in vivo and in vitro

Background: Fructose may induce non-alcoholic fatty acids (NAFLD) due to the gut-liver axis interactions. The mechanism of fructose impairing colon barrier is unrevealed. Methods: Normal and DSS-induced Sprague-Dawley rats fed by 35% fructose diets were used to evaluate colon barrier functions. Microbiome and metabolome were applied to screen potential biomarker bacteria and metabolites induced by fructose. HT-29 cells were applied to validate metabolite biomarker indoleacrylic acid (IAA) and indole-3-carboxaldehyde (I3A) function in colon barrier which impaired by fructose. Results: Fructose induced colon barrier dysfunction， aggravated colon impairment in DSS-induced rats. With fructose intake， the colon length shortened， goblet numbers declined， inflammation infiltration induced， inflammatory cytokines increased， and apoptosis signals upregulated in colon tissue. Moreover， fructose induced dysbiosis of microbiota and their metabolites. Adlercreutzia， Holdemania were screened out as potential bacteria biomarkers， IAA and I3A as tryptophan metabolites were selected as metabolite biomarkers inhibited by fructose. IAA and I3A treatment alleviated the impairment induced by fructose by increasing trans epithelial electric resistance (TEER) value， tight junction proteins (TJPs)， and AhR activity in HT-29 cell. Conclusion: Fructose stimulated inflammation， apoptosis， gut bacteria alteration， and induced the reduction of IAA and I3A. Since fructose inhibited production of IAA and I3A， AhR remained inactivated and consequently induced colon barrier dysfunction.

General anesthetic agents can impact brain function through interactions with neurons and their effects on glial *** perform essential roles in the central nervous system,including myelin sheath formation,axonal metabolism,and neuroplasticity *** are particularly vulnerable to the effects of general anesthetic agents resulting in impaired proliferation,differentiation,and *** are increasingly interested in the effects of general anesthetic agents on *** agents not only act on the surface receptors of oligodendrocytes to elicit neuroinflammation through modulation of signaling pathways,but also disrupt metabolic processes and alter the expression of genes involved in oligodendrocyte development and *** this review,we summarize the effects of general anesthetic agents on *** anticipate that future research will continue to explore these effects and develop strategies to decrease the incidence of adverse reactions associated with the use of general anesthetic agents.

BACKGROUND Myopericytoma is a benign tumor that typically occurs within subcutaneous tissue and most often involves the distal extremities,followed by the proximal extremities,neck,thoracic vertebrae and oral *** resection is often *** myopericytoma is extremely rare and has a poor ***,we report for the first time a case of malignant myopericytoma originating from the *** SUMMARY A 69-year-old male was admitted to our hospital with right upper quadrant pain for five *** suggested a liver mass with hemorrhage.A malignant hepatic tumor was the initial *** resection was performed after a complete preoperative work *** postoperative pathology suggested that the mass was a malignant myoblastoma unrelated to the *** months after the first surgery,an enhanced computed tomography(CT)scan revealed a recurrence of the *** diagnosis of malignant myopericytoma derived from the colon was confirmed on histopathological examination of the specimen from the second *** patient did not return to the hospital regularly for *** first postoperative abdominal CT examination six months after the second surgery demonstrated multiple liver *** time between the diagnosis of the tumor to death was approximately one *** Malignant myopericytoma is a rare *** diagnosis may be *** to a lack of treatment options,prognosis is poor.

The Orthopoxvirus genus,especially variola virus(VARV),monkeypox virus(MPXV),remains a significant public health threat *** development of therapeutic antibodies against orthopoxviruses is largely hampered by the high cost of antibody engineering and manufacturing ***-encoded antibodies have emerged as a powerful and universal platform for rapid antibody ***,by using the established lipid nanoparticle(LNP)-encapsulated mRNA platform,we constructed four mRNA combinations that encode monoclonal antibodies with broad neutralization activities against *** vivo characterization demonstrated that a single intravenous injection of each LNP-encapsulated mRNA antibody in mice resulted in the rapid production of neutralizing *** importantly,mRNA antibody treatments showed significant protection from weight loss and mortality in the vaccinia virus(VACV)lethal challenge mouse model,and a unique mRNA antibody cocktail,Mix2a,exhibited superior in vivo protection by targeting both intracellular mature virus(IMV)-form and extracellular enveloped virus(EEV)-form *** summary,our results demonstrate the proof-of-concept production of orthopoxvirus antibodies via the LNP-mRNA platform,highlighting the great potential of tailored mRNA antibody combinations as a universal strategy to combat orthopoxvirus as well as otheremerging viruses.

We have applied strong coupling unitary transformation method combined with Bose–Einstein statistical law to investigate magnetopolaron energy level temperature effects in halogen ion crystal quantum *** obtained results showed that under magnetic field effect,magnetopolaron quasiparticle was formed through the interaction of electrons and surrounding *** the same time,magnetopolaron was influenced by phonon temperature statistical law and important energy level shifts down and binding energy *** revealed that lattice temperature and magnetic field could easily affect magnetopolaron and the above results could play key roles in exploring thermoelectric conversion and conductivity of crystal materials.

Molecular mechanism of hypoxia and alpha-ketoglutaric acid on collagen expression in scleral fibroblasts

Based on the unified Hauser–Feshbach and exciton model,which can describe the particle emission processes between discrete energy levels with energy,angular momentum,and parity conservations,a statistical theory of light nucleus reaction(STLN)is developed to calculate the double-differential cross-sections of the outgoing neutron and light charged particles for the proton-induced^(6) Li reaction.A significant difference is observed between the p+^(6) Li and p+^(7) Li reactions owing to the discrepancies in the energy-level structures of the *** reaction channels,including sequential and simultaneous emission processes,are analyzed in *** the double-differential cross-sections of the outgoing proton as an example,the influence of contaminations(such as^(1) H,^(7)Li,^(12)C,and^(16)O)on the target is identified in terms of the kinetic energy of the first emitted *** optical potential parameters of the proton are obtained by fitting the elastic scattering differential *** calculated total double-differential cross-sections of the outgoing proton and deuteron at E_(p)=14.MeV agree well with the experimental data for different outgoing ***,the mixed double differential cross-sections of^(3) He andαare in good agreement with the *** agreement between the measured data and calculated results indicates that the two-body and three-body breakup reactions need to be considered,and the pre-equilibrium reaction mechanism dominates the reaction *** on the STLN model,a PLUNF code for the p+^(6) Li reaction is developed to obtain an ENDF-6-formatted file of the double-differential cross-sections of the nucleon and light composite charged particles.
