A polyphenol-assisted IL-10 mRNA delivery system for ulcerative colitis

With the development of synthesis technology, modified messenger RNA(mRNA) has emerged as a novel category of therapeutic agents for a broad of diseases. However, effective intracellular delivery of mRNA remains challenging, especially for its sensitivity to enzymatic degradation. Here, we propose a polyphenol-assisted handy delivery strategy for efficient in vivo delivery of IL-10 mRNA.IL-10 mRNA binds to polyphenol ellagic acid through supramolecular binding to yield a negatively charged core, followed by complexing with linear polyetherimide and coating with bilirubin-modified hyaluronic acid to obtain a layer-by-layer nanostructure. The nanostructure specifically up-regulated the level of IL-10, effectively inhibited the expression of inflammatory factors, promoted mucosal repair, protected colonic epithelial cells against apoptosis, and exerted potent therapeutic efficacy in dextran sulfate sodium salt-induced acute and chronic murine models of colitis. The designed delivery system without systemic toxicity has the potential to facilitate the development of a promising platform for mRNA delivery in ulcerative colitis treatment.

Nasopharyngeal carcinoma(NPC)is a common malignant carcinoma of the head and neck,and the biological mechanisms underlying the pathogenesis of NPC remain not fully understood.In the present study,we systematically analyzed four independent NPC transcriptomic datasets and focused on identifying the critical molecular networks and novel key hub genes implicated in NPC.We found totally 170 common overlapping differentially expressed genes(DEGs)in the four NPC datasets.GO and KEGG pathway analysis revealed that cell cycle dysregulation is a critical event in NPC.Protein-protein interaction(PPl)network analysis identified a 15 hub-gene corenetwork with overexpressedkinesin family member2C(KIF2C)as a central regulator.Loss-of-function study demonstrated that knockdown of KiF2C significantly inhibited cell growth and cell motility,and delayed cell cycle progression,accompanied with dramatic mitotic defects in spindle formation in NPC cells.RNA-seq analysis revealed that KIF2c knockdown led to deregulation of various downstream genes.KiF2C could also regulate the AKT/mTOR pathways,and enhance paclitaxel sensitivity in NPC cells.Taken together,our results suggest that cell cycle dysregulation is a critical event during NPC pathogenesis and KIF2C is a novel key mitotic hub gene with therapeutic potential in NPC.

Psychrophilic microalgae successfully survive in the extreme and highly variable polar ecosystems,which represent the energy base of most food webs and play a fundamental role in nutrient cycling.The success of microalgae is rooted in their adaptive evolution.Revealing how they have evolved to thrive in extreme polar environments will help us better understand the origin of life in polar ecosystems.We isolated a psychrophilic unicellular green alga,Microglena sp.YARC,from Antarctic sea ice which has a huge genome.Therefore,we predicted that gene replication may play an important role in its polar adaptive evolution.We found that its protein-coding gene number significantly increased and the duplication time was dated between 37 and 48 million years ago,which is consistent with the formation of the circumpolar Southern Ocean.Most duplicated paralogous genes were enriched in pathways related to photosynthesis,DNA repair,and fatty acid metabolism.Moreover,there were a total of 657 Microglena-specific families,including collagen-like proteins.The divergence in the expression patterns of the duplicated and species-specific genes reflects sub-and neo-functionalization during stress acclimation.Overall,key findings from this study provide new information on how gene duplication and their functional novelty contributed to polar algae adaptation to the highly variable polar environmental conditions.

Inflammatory bowel disease(IBD)is a chronic intestinal disease with painful clinical manifestations and high risks of cancerization.With no curative therapy for IBD at present,the development of effective therapeutics is highly advocated.Drug delivery systems have been extensively studied to transmit therapeutics to inflamed colon sites through the enhanced permeability and retention(EPR)effect caused by the inflammation.However,the drug still could not achieve effective concentration value that merely utilized on EPR effect and display better therapeutic efficacy in the inflamed region because of nontargeted drug release.Substantial researches have shown that some specific receptors and cell adhesion molecules highly expresses on the surface of colonic endothelial and/or immune cells when IBD occurs,ligandmodified drug delivery systems targeting such receptors and cell adhesion molecules can specifically deliver drug into inflamed sites and obtain great curative effects.This review introduces the overexpressed receptors and cell adhesion molecules in inflamed colon sites and retrospects the drug delivery systems functionalized by related ligands.Finally,challenges and future directions in this field are presented to advance the development of the receptor-mediated targeted drug delivery systems for the therapy of IBD.

The paucity of naturally occurring CD4^＋FoxP3^＋ regulatory T cells （Tregs） and difficulties in the identification and isolation of such cells are major obstacles in the study of human Tregs. The availability of human Treg-like cell lines is likely to facilitate a better understanding of the molecular basis of Treg function and aid the discovery of pharmacological reagents to regulate Treg activity in a disease state.

Cognitive J^(2)SAC:Joint Jamming,Sensing,and Communication Under Antagonistic Environment

Prognostic factors of Guillain-Barré syndrome: a 111-case retrospective review

Background: To identify the predictive factors associated with worse prognosis in the Guillain-Barré syndrome (GBS), which can be helpful to fully evaluate the disease progression and provide proper treatments. Methods: Clinical data of 111 GBS patients who were diagnosed from 2010 to 2015 were collected and retrospectively analyzed. Results: Patients with diabetes (P=0.031), high blood pressure at admission (P=0.034), uroschesis (P=0.028), fever (P<0.001), ventilator support (P<0.001) during hospitalization, disorder of consciousness (p=0.007) and absence of preceding respiratory infection(P=0.016) were associated with worse outcome at discharge, while abnormal sensation, ataxia, weakness and decrease of tendon reflex seemed not correlated with the Medical Research Council(MRC) score at discharge. Compared with the subtype of acute inflammatory demyelinating polyneuropathy, prognosis of Miller-Fisher syndrome (p<0.001) and cranial nerve variant (p<0.038) were better, but prognosis of acute motor axonal neuropathy(AMAN) was worse (p<0.032). Laboratory examinations at admission showed that hyperglycemia (P=0.002), high leukocyte count (P=0.010), hyperfibrinogenemia (P=0.001), hyponatremia (P=0.020), hypoalbuminemia (P=0.005), abnormal hepatic (P=0.048) and renal (P=0.009) functions were associated with poorer prognosis at discharge, while albuminocytologic dissociation in cerebrospinal fluid, GM1 and GQ1b antibody showed no correlation with the MRC score at discharge. γ-Globulin and glucocorticoid therapies showed no difference in the MRC score at the discharge. Conclusions: AMAN, diabetes, high blood pressure, uroschesis, high body temperature, ventilator support, consciousness disorder, absence of upper respiratory tract preceding infection, hyperglycemia, hyponatremia, hypoalbuminemia, high leukocyte count, hyperfibrinogenemia, abnormal hepatic and renal function were demonstrated as poor prognostic factors.

民办教育分类管理是我国民办教育改革的重大战略决策,也是民办教育事业持续健康发展的关键所在。当前我国民办高校分类管理整体进展较慢,其原因既包括分类登记程序不清晰、配套政策不健全等客观原因,也包括民办高校举办者分类选择的动力不强等主观原因:若选择非营利性,举办者既担心自身权益受损,又担心学校无法获得有效扶持;若选择营利性,举办者既面临道德压力,又担心学校的发展环境不佳。要在实事求是分析民办高校举办者实际利益诉求的基础上进行针对性的政策突破,尊重举办者切身利益诉求,稳定其办学预期、激发其办学积极性;加大扶持,引导非营利性和营利性民办高校共同发展、良性竞争;完善监管,避免将非营利性民办高校管僵、将营利性民办高校管死。

Design and test of an inside-filling pneumatic precision centralized seed-metering device for rapeseed

The pneumatic precision seed-metering devices used in combination with negative air pressure to suck seed and positive air flow to drop seed could overcome the problem of rapeseed precision planting.However,these devices can only plant a single row at a time or need high airflow pressure,which complicates the seed-metering system and the seeder structure,deteriorate the working reliability and have high power consumption.To overcome above shortcomings,a novel prototype of pneumatic precision centralized seed-metering device with seed inside-filling,sowing six rows at a time,was developed.The main structure and working principle of the prototype were described and the technical parameters were given.Experiments were conducted to investigate the influences of positive pressure,negative pressure,and rotating speed on the seeding performance.The stability of each row and the consistency between all rows associated with the seeding precision were analyzed to evaluate the performance of the prototype.The results indicated that the prototype can meet the requirements of precision seeding and the seed keep intact without any obvious damage.The performance of the prototype was no significant difference when the positive pressure was fluctuated from 200 Pa to 1200 Pa.The best seeding precision parameters in one row of the prototype was 92.48%of the qualified seeding index(Iqs)and 2.55%of the miss-seeding index(Imiss),when the rotating speed was 16 r/min,the positive pressure was 200 Pa and the negative pressure was–1600 Pa.When the rotating speed was 16 r/min,the positive pressure was 200 Pa,and the negative pressure was–1400 Pa or–1600 Pa,the Iqs of six rows were above 86.5%,the stability coefficient of variation(CV)of each row were below 4.6%,and the consistency CV were below 0.9%.Further field experiment showed that the rapeseed sowed by the prototype had acceptable seeding precision.The CV of the numbers of rapeseed seedlings in the field of the total and each row were 6.17%and 6.32%respectively.The research confirmed
