The clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9(Cas9)is widely used for targeted genomic and epigenomic modifications and imaging in cells and organisms,and holds tremendous promise in clinical *** efficiency and accuracy of the technology are partly determined by the target binding affinity and residence time of Cas9-single-guide RNA(sgRNA)at a given ***,little attention has been paid to the effect of target binding affinity and residence duration on the repair of Cas9-induced DNA double-strand breaks(DSBs).We propose that the choice of DSB repair pathway may be altered by variation in the binding affinity and residence duration of Cas9-sgRNA at the cleaved target,contributing to significantly heterogeneous mutations in CRISPR/Cas9 genome ***,we discuss the effect of Cas9-sgRNA target binding and residence on the choice of DSB repair pathway in CRISPR/Cas9 genome editing,and the opportunity this presents to optimize Cas9-based technology.

Performance and bioparticle growth of anaerobic baffled reactor （ABR） fed with low-strength domestic sewage

We investigated the performance of a 15.3 L capacity anaerobic baffled reactor （ABR） toward the treatment of low-strength domestic wastewater. The start- up period of the ABR was finished within approximately 130 days at a temperature below 25~C. The average CODcr in the effluent was 165 mg.L 1 and the corresponding CODcr removal efficiency of the ABR was 52.3%. During the third stage （from day 130 to day 233） of ABR operation, the average CODcr in the effluent reached 71 mg· L^-1, which meets the secondary discharge requirement of the Integrated Wastewater Discharge Standard （GB 18918-2002, China）. Moreover, partial microbial separa- tion was observed along the five ABR compartments through scanning electron microscopic images. The geometric mean diameter of bioparticles in the five compartments increased from 0.050 mm to 0.111, 0.107, 0.104, 0.110, and 0.103 mm during the start-up stage. After operation for 179days, the further increased to 0.376, corresponding diameters 0.225, 0.253, 0.239, and 0.288mm, respectively. The fractal dimensions of the bioparticles indicated that these particles have smoother surfaces and more compact structures during ABR operation. Morphological analysis of the bioparticle sections demonstrated that the bioparticles have a pore volume of 30%-55%. The highest porosity was observed for the bioparticles in the second ABR compartment, whereas the lowest fractal dimension ofbioparticle section was observed in the fifth compartment.

Optimization design of hydraulic manifold blocks (HMB) is studied as acomplex solid spatial layout problem. Based on comprehensive research into structure features anddesign rules of HMB, an optimal mathematical model for this problem is presented. Usinghuman-computer cooperative genetic algorithm (GA) and its hybrid optitation strategies, integratedlayout and connection design schemes of HMB can be automatically optimized. An example is given totestify it.

Distraction osteogenesis(DO)is a mechanobiological process of regenerating bone tissue by tension *** is used clinically to lengthen bones or to treat critical size bone *** DO provides satisfactory results in many cases,the prolonged period of treatment remains a major challenge that needs to be *** attempts have been devoted to accelerating bone regeneration during *** common approach is manipulation of the applied mechanical loading by altering distraction *** this article,we reviewed relevant in vivo animal studies exploring the effects of changing mechanical environments,by varying distraction parameters(e.g.,rate and frequency)or adding compression loading,on bone regeneration in *** further presented how the mechanically-regulated bone regeneration process during DO could be simulated by in silico models incorporating mechano-regulatory tissue differentiation rules.A comprehensive review of those in vivo and in silico studies may not solely provide important references for development of improved DO protocols in clinic,but also promote a deeper understanding of the mechanobiological mechanism of bone regeneration.

为研究燃煤电厂重金属排放对周边土壤重金属污染的影响,测定了某燃煤电厂烟囱入口烟气中Cd、Cr、Hg、Ni、Pb、As含量,同时根据当地气象及地形条件,在燃煤电厂周边采集19个土壤样品,分析了土壤中重金属含量及分布情况,并对其污染程度进行评价。结果表明,燃煤电厂烟气中6种重金属元素的质量浓度为0.02~2.00μg/m^(3),向大气中排放各种重金属元素为0.18~19.08 kg/a,单位发电量的重金属排放量满足美国环境保护署对火电厂重金属排放的要求。Cr、Hg在燃煤电厂常年主导风向下风向含量较高,与周边土壤中含量呈正相关关系,是燃煤电厂气态污染排放的特征元素。燃煤电厂周边各个方向土壤中重金属元素含量随烟气排放距离增加呈先增大后减小的趋势。燃煤电厂周边土壤中Cd、Hg、Ni、As处于中度污染水平,Cr、Pb处于轻微污染水平,需进一步加强烟气中重金属脱除的技术手段。
