Dry environment on the expression of lacrimal gland S100A9,Anxa1,and Clu in rats via proteomics

●AIM:To investigate the underlying mechanism of dry environment(autumn dryness)affecting the lacrimal glands in rats.●METHODS:Twenty Sprague-Dawley rats were randomly divided into two *** rats were fed in specific pathogen free environment as the control group(n=10),and the rats fed in dry environment as the dryness group(n=10).After 24d,lacrimal glands were collected from the *** tissues morphology was observed by hematoxylineosin(HE)*** mass tags(TMT)quantitative proteomics analysis technology was used to screen the differential expressed proteins of lacrimal glands between the two groups,then bioinformatics analysis was ***,the immunohistochemical(IHC)method was used to verify the target proteins.●RESULTS:In dryness group,the lacrimal glands lobule atrophied,the glandular cavities enlarged,the sparse nuclear distribution and scattered inflammatory infiltration between the acinus were *** proteomics exhibited that a total of 195 up-regulated and 236 downregulated differential expressed proteins screened from the lacrimal glands of *** was indicated that the biological processes(BP)of differential expressed proteins mainly included cell processes and single *** cellular compositions of differential expressed proteins mainly located in cells,*** molecular functions of differential expressed proteins mainly included binding,catalytic ***,the Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis showed that the differential expressed proteins mainly involved lysosome,complement and coagulation cascade,and ribosome *** IHC result verified that the up-regulated expression proteins of Protein S100A9(S100A9),Annexin A1(Anxa1),and Clusterin(Clu)in lacrimal glands of rats in dryness group were higher than control group.●CONCLUSION:The up-regulated expression proteins of S100A9,Anxa1,and Clu may be the potential mechanisms of dry eye symptoms caused by dry *** study pr

Neddylation plays a vital role in post-translational modification, intricately shaping the regulation of diverse biological processes, including those related to cellular immune responses. In fact, neddylation exerts control over both innate and adaptive immune systems via various mechanisms. Specifically, neddylation influences the function and survival of innate immune cells, activation of pattern recognition receptors and GMP-AMP synthase–stimulator of interferon genes pathways, as well as the release of various cytokines in innate immune reactions. Moreover, neddylation also governs the function and survival of antigen-presenting cells, which are crucial for initiating adaptive immune reactions. In addition, neddylation regulates T cell activation, proliferation, differentiation, survival, and their effector functions, thereby ensuring an appropriate adaptive immune response. In this review, we summarize the most recent findings in these aspects and delve into the connection between dysregulated neddylation events and immunological disorders, especially inflammatory diseases. Lastly, we propose future directions and potential treatments for these diseases by targeting neddylation.

Non-synchronous vibration of rotor blade in a six-stage transonic compressor

This paper presents an experimental study on the Non-Synchronous Vibration(NSV)in a six-stage transonic *** first part of the paper describes the NSV phenomenon of Rotor 1,which occurs when both Stator 1(S1)and Stator 2(S2)or S1 only are *** measurements and analysis are carried out for the former case through the unsteady wall pressure and the Blade Strain(BS).The spinning mode theory used in the rotor/stator interaction noise is employed to explain the relation between the circumferential wave number of the aerodynamic disturbance and the Nodal Diameter(ND)of the blade *** variations of the vibration amplitudes of different blades and the Inter-Blade Phase Angles(IBPAs)at different moments suggest that the evolution of NSV is a highly nonuniform phenomenon along the circumferential *** addition,the difference between the wall-pressure spectra generated by the NSV and the classic flutter has been *** the second part,the variations of aerodynamic loading due to the adjustment of the staggers of the Inlet Guide Vane(IGV),S1 and S2 have been *** is found that closing S1 only can result in a great fluctuation to the performance of the front stages,which might be detrimental to the flow organization and increase the risk of *** contrast,the effect of closing S2 only on the performance of the first two stages appears to be slighter relatively.

The Functional Redundancy of Neddylation E2s and E3s in Modulating the Fitness of Regulatory T Cells

Neddylation is necessary for activation of Cullin-RING ligases(CRLs),which degrade various immune regulatory *** recent study showed that while depletion of neddylation E2-E3 pair Ube2f-Sag in regulatory T(T_(reg))cells had no obvious phenotype,the same depletion of either Ube2m or Rbx1 caused inflammation disorders with different *** these E2s or E3s compensate each other in functional regulations of T_(reg)cells is,however,previously *** this report,we generated Foxp3^(Cre);Ube2m^(fl/fl);Ube2f^(fl/fl)or Foxp3^(Cre);Rbx1^(fl/fl);Sag^(fl/fl)double-null mice by simultaneous deletion of both neddylation E2s or E3s in T_(reg)cells,***,Ube2m&Ube2f double-null mice developed much severe autoimmune phenotypes than did Ube2m-null mice,indicating that Ube2m markedly compensates Ube2f in T_(reg)*** minor worsened autoimmune phenotypes seen at the very early stage in Rbx1&Sag double-null than Rbx1-null mice is likely due to already severe phenotypes of the later,indicating a minor compensation of Rbx1 for *** RNA profiling-based analyses revealed that up-and down-regulations of few signaling pathways in T_(reg)cells are associated with the severity of autoimmune ***,severer inflammation phenotypes seen in mice with double E3-null than with double E2-null T_(reg)cells indicate a neddylation-independent mechanism of 2 E3s,also known to serve as the RING component of CRLs in regulation of T_(reg)cell fitness.

目的:探究个人的购物出行模式随时间的变化差异性。探讨个人在星期中的天变化,量化多样化的特征和识别个体间的异质性。创新点:1.使用连续6周的数据进行动态交通行为分析;2.建立二元逻辑回归模型,识别个体间购物出行模式的异质性。方法:1.通过描述性统计分析,解析不同群体之间的购物行为的差异、每周购物天数的分布以及每周每人购物天数的标准偏差(表1、图1和2);2.通过回归模型分析,得出解释变量对购物地点和频率的影响结论(表2和3);3.构建二元逻辑回归模型,识别个体间购物出行模式的异质性,找出具有常规购物行为的群体(表4)。结论:1.低收入和卡尔斯鲁厄居住地对日常购物目的地的多样性呈正影响关系,而全职工作和男性对日常购物目的地的多样性呈负影响关系;2.连续6周的日常购物出行次数受到35~44岁的年龄范围、2500~2999德国马克的家庭月收入以及市中心居住地区的积极影响;3.购物地点和频率的多样性是随着个人属性和居住地点以系统的方式变化。

Feasibility Study on Recovering Human-Occupied Vehicle with An Offshore Crane

Launching and recovering human-occupied vehicles(HOVs)has always been a challenging *** current recovery process requires staff to manually complete the tethering task,which is inefficient and endangers the lives of *** paper suggests moving the recovery position from the surface to underwater at approximately half the wavelength of the water depth(30−50 m underwater),where the HOV experiences less environmental *** ROV equipped with a ultra-short baseline beacon(USBL)and a manipulator was used to complete the tethering ***,a shackle customized to the shape of the manipulator’s gripper is fitted to the end of the cable to simplify the tethering *** investigate the dynamic response of recovering the HOV using this suggested method,a comprehensive numerical model is developed in this *** effects of wind,surface waves,ocean currents,and nonlinear interaction between the installation vessel and the HOV are quantitatively *** results show that the proposed recovery method can reduce the motion amplitude of the HOV and that the wave has the greatest influence on the dynamic response of the HOV during the recovery *** model provides better insight into the proposed HOV recovery method and confirms the effectiveness of the heave compensation *** proposed approach aims to enhance safety and operational efficiency by reducing direct human involvement in the recovery process and mitigating potential *** finding holds particular significance,especially in environmentally sensitive areas,where reducing the impact on the surrounding ecosystem is crucial.

This paper presents a superhydrophobic melamine(ME)sponge(ME-g-PLMA)prepared via high-energy radiation-induced in situ covalent grafting of long-alkyl-chain dodecyl methacrylate(LMA)onto an ME sponge for efficient oil–water *** obtained ME-g-PLMA sponge had an excellent pore structure with superhydrophobic(water contact angle of 154°)and superoleophilic *** can absorb various types of oils up to 66–168 times its *** ME-g-PLMA sponge can continuously separate oil slicks in water by connecting a pump or separating oil underwater with a gravity-driven *** addition,it maintained its highly hydrophobic properties even after long-term immersion in different corrosive solutions and repeated oil *** modified ME-g-PLMA sponge exhibited excellent separation properties and potential for oil spill cleanup.
