The Functional Redundancy of Neddylation E2s and E3s in Modulating the Fitness of Regulatory T Cells

Neddylation is necessary for activation of Cullin-RING ligases(CRLs),which degrade various immune regulatory *** recent study showed that while depletion of neddylation E2-E3 pair ube2f-Sag in regulatory T(T_(reg))cells had no obvious phenotype,the same depletion of either ube2m or Rbx1 caused inflammation disorders with different *** these E2s or E3s compensate each other in functional regulations of T_(reg)cells is,however,previously *** this report,we generated Foxp3^(Cre);ube2m^(fl/fl);ube2f^(fl/fl)or Foxp3^(Cre);Rbx1^(fl/fl);Sag^(fl/fl)double-null mice by simultaneous deletion of both neddylation E2s or E3s in T_(reg)cells,***,ube2m&ube2f double-null mice developed much severe autoimmune phenotypes than did ube2m-null mice,indicating that ube2m markedly compensates ube2f in T_(reg)*** minor worsened autoimmune phenotypes seen at the very early stage in Rbx1&Sag double-null than Rbx1-null mice is likely due to already severe phenotypes of the later,indicating a minor compensation of Rbx1 for *** RNA profiling-based analyses revealed that up-and down-regulations of few signaling pathways in T_(reg)cells are associated with the severity of autoimmune ***,severer inflammation phenotypes seen in mice with double E3-null than with double E2-null T_(reg)cells indicate a neddylation-independent mechanism of 2 E3s,also known to serve as the RING component of CRLs in regulation of T_(reg)cell fitness.

Influence of the Acoustic Liner in Large Eddy Simulation of Longitudinal Thermoacoustic Instability in a Model Annular Combustor

Although extensive efforts have been made to dampen the thermoacoustic instability,successfully controlling the pressure oscillations in modern gas turbines or aeroengines remains *** influence of the acoustic liner on the longitudinal thermoacoustic mode in a model annular combustor is investigated by Large Eddy Simulation(LES) in this *** result of the self-excited longitudinal thermoacoustic instability without the liner agrees well with the frequency and acoustic analysis of the pressure mode based on experimental *** different bias flow velocities of the liner located downstream of the combustor are then *** results reveal that the existence of the liner influences not only the acoustic field but also the flow *** the bias velocity is large,it leads to intense turbulence-induced fluctuations,and the pressure oscillation is modulated *** shows that the weak coupling between flow and pressure oscillations plays a significant role in the onset of the intermittency of a thermoacoustic *** on the dynamic analysis of the thermoacoustic system with the acoustic liner,this intermittency is caused by the influence of the flow field on the flame-acoustic ***,a low-order modeling method based on Van der Pol(VdP) oscillator with additive stochastic forcing is conducted to reproduce the evolving dynamics of the thermoacoustic *** the numerical cases demonstrated in this work are relatively simpler than those in a practical combustion system,the results are helpful for us to understand the effect of the acoustic liner and show the attractive potential to apply this device to suppress thermoacoustic instability.

An Encoding-Decoding Framework Based on CNN for circ RNA-RBP Binding Sites Prediction

Predicting RNA binding protein(RBP) binding sites on circular RNAs(circ RNAs) is a fundamental step to understand their interaction mechanism. Numerous computational methods are developed to solve this problem, but they cannot fully learn the features. Therefore, we propose circ-CNNED, a convolutional neural network(CNN)-based encoding and decoding framework. We first adopt two encoding methods to obtain two original matrices. We preprocess them using CNN before fusion. To capture the feature dependencies, we utilize temporal convolutional network(TCN) and CNN to construct encoding and decoding blocks, respectively. Then we introduce global expectation pooling to learn latent information and enhance the robustness of circ-CNNED. We perform circ-CNNED across 37 datasets to evaluate its effect. The comparison and ablation experiments demonstrate that our method is superior. In addition, motif enrichment analysis on four datasets helps us to explore the reason for performance improvement of circ-CNNED.

Due to the rapid evolution of Advanced Persistent Threats(APTs)attacks,the emergence of new and rare attack samples,and even those never seen before,make it challenging for traditional rule-based detection methods to extract universal rules for effective *** the progress in techniques such as transfer learning and meta-learning,few-shot network attack detection has ***,challenges in few-shot network attack detection arise from the inability of time sequence flow features to adapt to the fixed length input requirement of deep learning,difficulties in capturing rich information from original flow in the case of insufficient samples,and the challenge of high-level abstract *** address these challenges,a few-shot network attack detection based on NFHP(Network Flow Holographic Picture)-RN(ResNet)is ***,leveraging inherent properties of images such as translation invariance,rotation invariance,scale invariance,and illumination invariance,network attack traffic features and contextual relationships are intuitively represented in *** addition,an improved RN network model is employed for high-level abstract feature extraction,ensuring that the extracted high-level abstract features maintain the detailed characteristics of the original traffic behavior,regardless of changes in background ***,a meta-learning model based on the self-attention mechanism is constructed,achieving the detection of novel APT few-shot network attacks through the empirical generalization of high-level abstract feature representations of known-class network attack *** results demonstrate that the proposed method can learn high-level abstract features of network attacks across different traffic detail *** state-of-the-artmethods,it achieves favorable accuracy,precision,recall,and F1 scores for the identification of unknown-class network attacks through cross-validation onmultiple datasets.

Relationship between seismic structures and the diverse rupture processes of the 2023 Türkiye earthquake doublet

Recent geodetic and seismological observations of two major earthquakes in southeastern Türkiye in February 2023 have revealed complex rupture initiation,propagation,and segmentation along the East Anatolian Fault Zone(EAFZ)and surrounding ***,the role of upper crust structures along the EAFZ in determining the diverse rupture processes of this earthquake doublet remains *** further investigate this,we employed double-difference location and seismic tomography techniques to determine high-resolution seismic velocities(V_(P),V_(S))and Poisson’s ratio(σ)structures using a multiparameter joint tomographic *** dataset includes 100,833 high-quality source-receiver travel-time pairs of P-and *** find that the unique rupture processes of this earthquake doublet were primarily influenced by contrasting crustal seismic structures and localized geological *** M_(w)7.8 mainshock was initiated within a transitional edge zone characterized by a rigid part(asperity)of the seismogenic zone with sharp contrast variations in rock strength ranging from low to high along the *** comparison,the M_(w)7.6 rupture originated in a ductile belt featuring fluid saturation with low-VP,low-VS,and high-σvalues that extended parallel to the Cardak *** pronounced contrast structures observed along the former rupture can be attributed to the oblique collision system between the weakened section of the east Anatolian plateau and the brittle Arabian platform,while the latter rupture was initiated within the ductile structure associated with fluid intrusion caused by the northward subduction of the Cyprus slab and subsequent ***,the occurrence of the first earthquake(E1)serves to alleviate shear stress on the second earthquake(E2)fault,potentially impeding the initiation of an E2 *** the contrary,this event also significantly reduces the normal stress acting on the E2 fault due to a double left-lateral strike-slip system

The alloy-type material Sb is widely used in the anode materials of lithium-ion batteries(LIBs)due to its high theoretical specific ***,its serious volume expansion problem during alloying/dealloying of Li~+limits its practical *** this work,C-Sb composite was constructed as anode material of LIBs by electrospinning route for the first time,Sb was introduced into the polyacrylonitrile-based hard carbon and coal tar pitch-based soft carbon composite amorphous carbon fiber with a diameter of 300-600 nm,which realized high cycling *** C-Sb-2(the mass ratio of polyacrylonitrile to Sb source is 1:2)electrode displayed charge capacities of 1098.5,930.3,841.7,753.5,643.9 and 545.8 mAh·g^(-1)at 0.1,0.2,0.3,0.5,1 and 2 A·g^(-1),*** when the current density returned to 0.1 A·g^(-1),the charge capacity was 939.3 mAh·g^(-1),revealing good stability and *** introduction of Sb into the amorphous carbon improved its conductivity and addressed the volume expansion issue of high specific capacity Sb during charge/***-situ XRD analysis confirmed the high reversibility of the C-Sb-2 during charging and *** functional theory(DFT)calculations revealed the gradual enhancement of the interface interaction between Sb_(x)Li_(y) and amorphous carbon(AMC)with increasing lithium content,contributing to the anchoring of alloy nanoparticles on the AMC surface and buffering the volume change of the ***,the gradual lithiation of Sb facilitated the electron transfer from Sb_(x)Li_(y) to *** findings hold promise for designing lithium storage materials with exceptional performance,highlighting the potential of C-Sb composites as anode materials for efficient next-generation lithium storage.

Efficient 2-D MuSIC algorithm for super-resolution moving target tracking based on an FMCW radar

Frequency modulated continuous wave(FMCW)radar is an advantageous sensor scheme for target estimation and environmental ***,existing algorithms based on discrete Fourier transform(DFT),multiple signal classification(MuSIC)and compressed sensing,etc.,cannot achieve both low complexity and high resolution *** paper proposes an efficient 2-D MuSIC algorithm for super-resolution target estimation/tracking based on FMCW ***,we enhance the efficiency of 2-D MuSIC azimuth-range spectrum estimation by incorporating 2-D DFT and multi-level resolution searching ***,we apply the gradient descent method to tightly integrate the spatial continuity of object motion into spectrum estimation when processing multi-epoch radar data,which improves the efficiency of continuous target *** two approaches have improved the algorithm efficiency by nearly 2-4 orders of magnitude without losing accuracy and *** experiments are conducted to validate the effectiveness of the algorithm in both single-epoch estimation and multi-epoch tracking scenarios.

Neddylation plays a vital role in post-translational modification, intricately shaping the regulation of diverse biological processes, including those related to cellular immune responses. In fact, neddylation exerts control over both innate and adaptive immune systems via various mechanisms. Specifically, neddylation influences the function and survival of innate immune cells, activation of pattern recognition receptors and GMP-AMP synthase–stimulator of interferon genes pathways, as well as the release of various cytokines in innate immune reactions. Moreover, neddylation also governs the function and survival of antigen-presenting cells, which are crucial for initiating adaptive immune reactions. In addition, neddylation regulates T cell activation, proliferation, differentiation, survival, and their effector functions, thereby ensuring an appropriate adaptive immune response. In this review, we summarize the most recent findings in these aspects and delve into the connection between dysregulated neddylation events and immunological disorders, especially inflammatory diseases. Lastly, we propose future directions and potential treatments for these diseases by targeting neddylation.
