The apoptosis that occurs in the immature testis under physiological conditions is necessary for male germ cell development,whereas improper activation of apoptosis can impair spermatogenesis and cause defects in *** previously demonstrated that in mice,the makorin-2(Mkrn 2)gene is expressed exclusively in the testis and its deletion leads to male *** understand the potential molecular mechanism,in this study,we found that levels of apoptosis in the testis were abnormally high in the absence of Mkrn *** identify specific gene(s)involved,we performed digital gene expression profiling(DGE)and pathway analysis via gene set enrichment analysis(GSEA)and the Kyoto Encyclopedia of Genes and Genomes(KEGG)database,and we found that MKRN2 inhibits p53 apoptosis effector related to PMP22(PERP)expression and that levels of the protein in sperm samples have an inverse correlation with infertility *** additionally indicated that PERP is a negative regulator of spermatogenesis and that its ectopic expression induces male ***,Gene Expression Omnibus(GEO)dataset analysis showed that p53,upstream of PERP,was upregulated in oligoasthenoteratozoospermia(OAT).These observations suggest that Mkrn 2 is crucial for protecting germ cells from excessive apoptosis and implicate Mkrn 2-based suppression of the p53/PERP signaling pathway in spermatogenesis and male fertility.

AIM： To summarize preoperative evaluation and outcome of corneal transplantation for limbal dermoids for ten ***： Eighty-five patients diagnosed with limbal dermoids and treated with corneal transplantation were analyzed retrospectively. All patients were further divided into two groups according to absence or presence of neovascularization surrounding the dermoids in the corneal stroma. Eighty-two eyes were treated with tumor excision combined with partial lamellar sclerokeratoplasty, and the other three eyes were performed by penetrating keratoplasty. The size and location of the tumor, the associated ocular and systemic anomalies, the depth of the corneal penetration of tumor tissues, the preoperative and postoperative best-corrected visual acuity （BCVA）, graft survival and cosmetic outcome, and surgical complications were recorded ***： The average age at surgery was 5.3y （range, 3mo-36y）. The mean size of dermoids was 6.1±1.6 mm. The 43.5% of eyes （37/85） were present with hair at the surface of the dermoid and 72.9% of dermoids were located inferotemporal of the eye. Amplyopia was present in 34.1% of patients （29/85） and 9.4% of patients （8/85） had lipodermoids. Eighteen patients suffered from Goldenhar’s syndrome with an accessory ear. The 75% of patients in group 1 had involvement of the corneal deep stroma down to Descemet’s membrane without involving it, but 71.4% of patients had Descemet’s membrane involvement in group 2. Preoperative BCVA ranged from counting fingers to 20/20. Postoperatively 81.1% had a BCVA of 20/800 or better. There was no significant difference between the post-surgical BCVA of the two groups （t=1.584, P〉0.05）. The grafts of 70.5% patients were present as 1＋ opacity, 21.1% as 2＋ opacity, 8.2% as 3＋ opacity and none as 4＋ opacity. Optical temperature sensing based on the fluorescence intensity ratio(FIR)of red emission for lanthanide ions holds significant relevance in non-contact temperature measurement for biological *** this study,the perovskite-structured KZnF_(3)is utilized as a host material for Er^(3+)to achieve a high-purity upconversion(UC)red *** observed Stark splitting of the red emission peak provides evidence of the energy level splitting of Er^(3+).Group theory is employed to decompose the spectral branching of Er^(3+)under the point group symmetry of KZnF_(3),allowing for the derivation of Stark splitting energy levels induced by the crystal field *** optical temperature-sensing behavior of the red UC luminescence was investigated,specifically examining the FIR of the splitting sub-peaks,which exhibited an exponential relationship with *** KZnF_(3):Yb^(3+),Er^(3+)demonstrated a relative sensitivity(S_(r))of 0.00182%·K^(-1)at 298 K,highlighting its excellent response to *** vivo bio-thermometry experiments conducted on chicken breast validated the material's ability to penetrate biological tissues and showed its significant sensitivity of the FIR to *** results establish KZnF_(3):Yb^(3+),Er^(3+)as a promising material for optical thermometry in various biological applications.

Background:Endothelial cells play a key role in the cytokine storm caused by influenza A virus. MicroRNA-155 (miR-155) is an important regulator in inflammation. Its role in the inflammatory response to influenza A infection, however, has yet to be elucidated. In this study, we explored the role as well as the underlying mechanism of miR-155 in the cytokine production in influenza A-infected endothelial ***:Human pulmonary microvascular endothelial cells (HPMECs) were infected with the influenza A virus strain H1N1. The efficiency of H1N1 infection was confirmed by immunofluorescence. The expression levels of proinflammatory cytokines and miR-155 were determined using real-time polymerase chain reaction. A dual-luciferase reporter assay characterized the interaction between miR-155 and sphingosine-1-phosphate receptor 1 (S1PR1). Changes in the target protein levels were determined using Western blot ***:MiR-155 was elevated in response to the H1N1 infection in HPMECs (24 h post-infection vs. 0 h post-infection, 3.875 ± 0.062 vs. 1.043 ± 0.013, P = 0.001). Over-expression of miR-155 enhanced inflammatory cytokine production (miR-155 mimic vs. negative control, all P < 0.05 in regard of cytokine levels) and activation of nuclear factor kappa B in infected HPMECs (miR-155 mimic vs. negative control, P = 0.004), and down-regulation of miR-155 had the opposite effect. In addition, S1PR1 was a direct target of miR-155 in the HPMECs. Inhibition of miR-155 enhanced the expression of the S1PR1 protein. Down-regulation of S1PR1 decreased the inhibitory effect of the miR-155 blockade on H1N1-induced cytokine production and nuclear factor kappa B activation in HPMECs. Conclusion:MiR-155 maybe modulate influenza A-induced inflammatory response by targeting S1PR1.

Objective: To assess the effects of axial vibrations on gene expression and lumbar intervertebral disc degeneration in vivo. Methods: A modified bipedal rat model was established using a brachial plexus rhizotomy approach to imitate human upright posture. The experimental animals were randomly divided into three groups: control, vertical vibration, and whole-body vibration. Gene expression in degeneration of the intervertebral discs was assessed by reverse transcription-quantitative polymerase chain reaction. Results: The expression of aggrecan, Col1α1, Col2α1, and decorin were shown to be up-regulated in 14-week-old rats in the vertical vibration and whole-body vibration groups, whereas biglycan and versican expression was down-regulated in 14-week-old rats of the two experimental groups. Furthermore, biglycan and versican expression levels were shown to be lower in the whole-body vibration group than in the vertical vibration group(P<0.05). Conclusions: This in-vivo study demonstrated that vibrations can influence the expression of anabolic genes. Furthermore, whole-body vibrations seem to have a greater effect in this regard than vertical vibrations. A new method is expected to relieve the low back pain of the patients through our research.
