Development and application of a multi-step porcine in vitro system to evaluate feedstuffs and feed additives for their efficacy in nutrient digestion, digesta characteristics, and intestinal immune responses

In vitro model provides alternatives to the use of live animals in research. In pig nutrition, there has been a tremendous increase in in vivo research over the decades. Proper utilization of in vitro models could provide a screening tool to reduce the needs of in vivo studies, research duration, cost, and the use of animals and feeds. This study aimed to develop a multi-step porcine in vitro system to simulate nutrient digestion and intestinal epithelial immune responses affected by feedstuffs and feed additives. Seven feedstuffs(corn, corn distillers dried grains with solubles [corn DDGS], barley, wheat, soybean meal, soy protein concentrates, and Corynebacterium glutamicum cell mass [CGCM]), feed enzymes(xylanase and phytase), and supplemental amino acids(arginine, methionine, and tryptophan), were used in this in vitro evaluation for their efficacy on digestibility, digesta characteristics, and intestinal health compar.d with the results from previously published in vivo studies. All in vitro evaluations were triplicated. Data were analyzed using Mixed procedure of SAS9.4. Evaluations included(1) nutrient digestibility of feedstuffs,(2) the effects of feed enzymes, xylanase and phytase, on digestibility of feedstuffs and specific substrates, and(3) the effects of amino acids, arginine, tryptophan, and methionine, on anti-inflammatory, anti-oxidative, and anti-heat stress statuses showing their effects(P < 0.05) on the measured items. Differences in dry matter and crude protein digestibility among the feedstuffs as well as effects of xylanase and phytase were detected(P < 0.05), including xylo-oligosaccharide profiles and phosphorus release from phytate. Supplementation of arginine, tryptophan, and methionine modulated(P < 0.05) cellular inflammatory and oxidative stress responses. Seasonal Prediction of Monthly Precipitation in China Using Large-Scale Climate Indices

In this study, seasonal predictions were applied to precipitation in China on a monthly basis based on a multivariate linear regression with an adaptive choice of predictors drawn from regularly updated climate indices with a two to twelve month lead time. A leave-one-out cross validation was applied to obtain hindcast skill at a 1% significance level. The skill of forecast models at a monthly scale and their significance levels were evaluated using Anomaly Correlation Coefficients （ACC） and Coefficients Of Determination （COD）. The monthly ACC skill ranged between 0.43 and 0.50 in Central China, 0.41-0.57 in East China, and 0.41 0.60 in South China. The dynamic link between large-scale climate indices with lead time and the precipitation in China is also discussed based on Singular Value Decomposition Analysis （SVDA） and Correlation Analysis （CA）.

Investigations of an Air Starting Motor of Marine Medium-Speed Diesel Engine with Numerical Analyses

The marine medium-speed diesel engines are started by two methods;one is the electric motors,and the other air starting *** though air starting motor is dependent of the engine types and sizes,it has been widely used in this area due to its simplicity,convenience and *** purpose of this paper is to give the designing par.meters in order to make a proper "Air Starting Motor" using *** aerodynamic approaches were given to understand the internal flow characteristics of the air starting *** addition,we have carried out the investigation of effects of tip *** the calculations the tip clearance of air starting motor has been varied between 0,2.8,4.3 and 5.7% of blade *** results of computation are the tip clearance increased to 2.8%,the torque decreased 24%,and there was no more large changes when the clearances increased to 4.3% and 5.7%.

Cloud Radiative Effects and Changes Simulated by the Coupled Model Intercompar.son Project Phase 5 Models

Using 32 CMIP5（Coupled Model Intercompar.son Project Phase 5） models, this study examines the veracity in the simulation of cloud amount and their radiative effects（CREs） in the historical run driven by observed external radiative forcing for 1850-2005, and their future changes in the RCP（Representative Concentration Pathway） 4.5 scenario runs for2006-2100. Validation metrics for the historical run are designed to examine the accuracy in the representation of spatial patterns for climatological mean, and annual and interannual variations of clouds and CREs. The models show large spread in the simulation of cloud amounts, specifically in the low cloud amount. The observed relationship between cloud amount and the controlling large-scale environment are also reproduced diversely by various models. Based on the validation metrics,four models-ACCESS1.0, ACCESS1.3, Had GEM2-CC, and Had GEM2-ES-are selected as best models, and the average of the four models performs more skillfully than the multimodel ensemble *** models project global-mean SST warming at the increase of the greenhouse gases, but the magnitude varies across the simulations between 1 and 2 K, which is largely attributable to the difference in the change of cloud amount and distribution. The models that simulate more SST warming show a greater increase in the net CRE due to reduced low cloud and increased incoming shortwave radiation, par.icularly over the regions of marine boundary layer in the subtropics. Selected best-performing models project a significant reduction in global-mean cloud amount of about-0.99% K^-1and net radiative warming of 0.46 W m^-2K^-1, suggesting a role of positive feedback to global warming.

Temperature Distribution and Thermal Stresses in Various Conditions of Moving Heating Source during Line Heating Process

Line heating method is widely used to manufacture curved surfaces in ship building. The main factors governing the quality of the manufactured products are the moving velocity of the heating source, heating strength, and heating ways. In this study, the temperature distributions of the heated plate were investigated with the condition that the line heating process was automatic. The temperature variations were also investigated with the changes of those three variables. The numerical results showed that the peak temperature decreased as the moving velocity of the heating source increased. It also revealed that the peak temperatures changed linearly with the changes of the heating source.

Effect of modified fasting therapy on body weight, fat and muscle mass, and blood chemistry in patients with obesity

OBJECTIVE: The aim of this study was to investigate the effects and safety of modified fasting therapy using fermented medicinal herbs and exercise on body weight, fat and muscle mass, and blood chemistry in obese ***: Twenty-six patients par.icipated in a14-day fast, during which they ingested a supplement made from fermented medicinal herbs and carbohydrates(intake: 400-600 kcal/d). The schedule included 7 prefasting relief days and 14 days of stepwise reintroduction of food. The patients also took par. in an exercise program that incorporated Qigong, weight training, and walking exercises. The efficacy of treatments was observed by assessing body fat mass and muscle mass, and alanine aminotransferase(ALT), aspar.ate aminotransferase(AST),cholesterol, and triglycerides in each study *** symptoms or side effects were ***: Body weight and body fat mass both decreased significantly by(5.16 ± 0.95) and(3.89 ±0.79) kg(both P < 0.05), while muscle mass decreased by an average of(0.26 ± 0.22) kg, without statistical significance. ALT levels were significantly decreased(P < 0.05), while AST levels decreased without statistical significance(P = 0.052). The levels of total cholesterol and triglycerides were also significantly decreased(both P < 0.05). There were few adverse events except for mild dizziness, which did not affect everyday ***: These results suggest that modified fasting therapy using fermented medicinal herbs and exercise could be effective and safe on obese patients.

Polyherbal formula SC-E3 inhibits rheumatoid arthritis activity in a mouse model of type-Ⅱcollagen-induced arthritis

Objective:SC-E3 is a polyherbal formula that contains five medicinal herbs used frequently in traditional herbal *** our previous study,we demonstrated the antioxidant and anti-inflammatory effects of *** present study examined the effects of SC-E3 in a mouse model of type-II collagen-induced arthritis(CIA).Methods:In vivo,male DBA/1 J mice were immunized by intradermal injection of bovine type-II collagen and complete or incomplete Freund’s adjuvant,to induce ***-E3 was orally administered daily for 23 *** vitro,bone marrow-derived macrophages(BMMs)were treated with macrophage colony-stimulating factor(M-CSF)and receptor activator of nuclear factor-j B ligand(RANKL)in the absence or presence of ***:Administrations of SC-E3 were found to have anti-arthritic effects in the joints of CIA mice,as evidenced by reduced paw swelling,bone erosion and deformation,inflammatory cell infiltration,and inflammation in synovial ***-E3 also reduced serum levels of tumor necrosis factor-a,interleukin-1 b,aspar.ate aminotransferase and alanine ***,tartrateresistant acid phosphatase-positive osteoclast numbers in the joints were significantly lower in SC-E3-treated CIA mice than in CIA *** addition,the differentiations of BMMs to multinucleated osteoclasts induced by M-CSF and RANKL stimulation were dose-dependently reduced by ***:These results suggest that SC-E3 possesses substantial anti-arthritic activity because it inhibits pro-inflammatory cytokines and osteoclastogenesis,and that SC-E3 has potential therapeutic use for the treatment of rheumatoid arthritis.
