Mechanistic links between gut microbial community dynamics, microbial functions and metabolic health

Gut microbes comprise a high density, biologically active community that lies at the interface of an animal with its nutritional environment. Consequently their activity profoundly influences many aspects of the physiology and metabolism of the host animal. A range of microbial structural components and metabolites directly interact with host intestinal cells and tissues to influence nutrient uptake and epithelial health. Endocrine, neuronal and lymphoid cells in the gut also integrate signals from these microbial factors to influence systemic responses. Dysregulation of these host-microbe interactions is now recognised as a major risk factor in the development of metabolic dysfunction. This is a two-way process and understanding the factors that tip host-microbiome homeostasis over to dysbiosis requires greater appreciation of the host feedbacks that contribute to regulation of microbial community composition. To date, numerous studies have employed taxonomic profiling approaches to explore the links between microbial composition and host outcomes(especially obesity and its comorbidities), but inconsistent host-microbe associations have been reported. Available data indicates multiple factors have contributed to discrepancies between studies. These include the high level of functional redundancy in host-microbiome interactions combined with individual variation in microbiome composition; differences in study design, diet composition and host system between studies; and inherent limitations to the resolution of r RNA-based community profiling. Accounting for these factors allows for recognition of the common microbial and host factors driving community composition and development of dysbiosis on high fat diets. New therapeutic intervention options are now emerging.

A Novel Multiple-Network Analysis Integrating Metabolomics and Pharmacology for Mechanism Study of Medicine:A Case Study of CDD-2101 Against Constipation

Constipation aff ects more than 14% of the global population with complex pathogenesis. Compound Traditional Chinese medicines(TCM), such as CDD-2101, a manufactured version of a time-tested TCM, hold great promise for constipation treatment due to the multiple-component-multiple-target strategy. Here, a novel multiple network analysis based on metabolomics and network pharmacology was developed to decipher the complex interplays between CDD-2101 and constipation. First, CDD-2101 alleviated constipation in a loperamide-induced acute constipation mice model and no adverse eff ect was observed. Then, fecal metabolomics and phytochemistry profi les were conducted by liquid chromatography-mass spectrometry analysis. The untargeted metabolomics built a metabolomic network related to signifi cantly modulated pathways during the constipation alleviation, including bile acid secretion, steroid hormone biosynthesis, glycerophospholipid metabolism, and linoleic acid metabolism. Based on the phytochemistry, network pharmacology predicted similar modulated pathways and built a hub-target network including retinoid X receptor alpha(RXRA), cytochrome P450 1A1(CyP1A1), cytochrome P450 1A2(CyP1A2), and phospholipase A2-IVA(PLA2G4). Third, integrated networks were built to select the key pathways and targets, as well as the key ingredients, including rhein, hesperetin, albifl orin, and magnolol from the TCM, which were corroborated by molecular docking, in vitro enzyme assays, and targeted metabolomics results. Finally, an integrated network was built to elucidate the multi-ingredient-multi-symptom linkage during the treatment. In conclusion, the novel multiple-network analysis revealed the complex “multi-component-multi-target-multi-pathway” mechanisms of CDD-2101 against constipation. Our work provides insight into the understanding of active compounds and pharmacological mechanisms, contributing to the development of natural medicines.

Vaccinations are essential for preventing and treating disease,especially cancer nanovaccines,which have gained considerable interest recently for their strong anti-tumor immune *** can prompt the immune system to generate antibodies and activate various immune cells,leading to a response against tumor tissues and reducing the negative effects and recurrence risks of traditional chemotherapy and *** enhance the flexibility and targeting of vaccines,nanovaccines utilize nanotechnology to encapsulate or carry antigens at the nanoscale level,enabling more controlled and precise drug delivery to enhance immune *** nanovaccines function by encapsulating tumor-specific antigens or tumor-associated antigens within *** small size of these nanomaterials allows for precise targeting of T cells,dendritic cells,or cancer cells,thereby eliciting a more potent anti-tumor *** this paper,we focus on the classification of carriers for cancer nanovaccines,the roles of different target cells,and clinically tested cancer nanovaccines,discussing strategies for effectively inducing cytotoxic T lymphocytes responses and optimizing antigen presentation,while also looking ahead to the translational challenges of moving from animal experiments to clinical trials.

High Fe‑Loading Single‑Atom Catalyst Boosts ROS Production by Density Effect for Efficient Antibacterial Therapy

The current single-atom catalysts(SACs)for medicine still suffer from the limited active site ***,we develop a synthetic method capable of increasing both the metal loading and mass-specific activity of SACs by exchanging zinc with *** constructed iron SACs(h^(3)-FNC)with a high metal loading of 6.27 wt%and an optimized adjacent Fe distance of~4 A exhibit excellent oxidase-like catalytic performance without significant activity decay after being stored for six months and promising antibacterial ***,a“density effect”has been found at a high-enough metal doping amount,at which individual active sites become close enough to interact with each other and alter the electronic structure,resulting in significantly boosted intrinsic activity of single-atomic iron sites in h^(3)-FNCs by 2.3 times compared to low-and medium-loading ***,the overall catalytic activity of h^(3)-FNC is highly improved,with mass activity and metal mass-specific activity that are,respectively,66 and 315 times higher than those of commercial Pt/*** addition,h^(3)-FNCs demonstrate efficiently enhanced capability in catalyzing oxygen reduction into superoxide anion(O_(2)·^(−))and glutathione(GSH)*** in vitro and in vivo assays demonstrate the superior antibacterial efficacy of h^(3)-FNCs in promoting wound *** work presents an intriguing activity-enhancement effect in catalysts and exhibits impressive therapeutic efficacy in combating bacterial infections.

近年来,创新网络作为区域和城市创新驱动力的重要性得到了广泛关注。一些研究将网络关系视为知识在城市内和城市间传播的渠道,并通过网络分析视角洞察城市创新的结构和动态。然而,城市创新的网络机制,特别是数字经济发展背景下的数字赋能城市创新能力提升机制还有待进一步研究。本文利用长三角城市群26个城市2009年—2018年联合申请专利数据构建了城市创新网络,探讨了网络结构属性——小世界和结构洞对城市创新能力的影响。结果表明,小世界与城市创新能力呈“倒U型”关系,数字经济发展水平正向调节了二者之间的“倒U型”关系,结构洞对城市创新能力具有正向影响。本文厘清了创新网络结构对城市创新能力的影响,回应了以往研究关于小世界和结构洞对创新影响的理论争议,为区域创新的理论研究提供了新颖性观点,深化了对城市群创新发展规律的认识,对推进中国城市群协同创新具有重要意义。

Recent Advances in Computational Simulation of Macro-,Meso-,and Micro-Scale Biomimetics Related Fluid Flow Problems

Over the last decade, computational methods have been intensively applied to a variety of scientific researches and engineering designs. Although the computational fluid dynamics （CFD） method has played a dominant role in studying and simulating transport phenomena involving fluid flow and heat and mass transfers, in recent years, other numerical methods for the simulations at meso- and micro-scales have also been actively applied to solve the physics of complex flow and fluid-interface interactions. This paper presents a review of recent advances in multi-scale computational simulation of biomimetics related fluid flow problems. The state-of-the-art numerical techniques, such as lattice Boltzmann method （LBM）, molecular dynamics （MD）, and conventional CFD, applied to different problems such as fish flow, electro-osmosis effect of earthworm motion, and self-cleaning hydrophobic surface, and the numerical approaches are introduced. The new challenging of modelling biomimetics problems in developing the physical conditions of self-clean hydrophobic surfaces is discussed.

House price prediction is of utmost importance in forecasting residential property prices,particularly as the demand for high-quality housing continues to *** predictions have implications for real estate investors,financial institutions,urban planners,and ***,accurately predicting house prices is challenging due to the complex interplay of various influencing *** studies have primarily focused on basic property information,leaving room for further exploration of more intricate features,such as amenities,traffic,and social sentiments in the surrounding *** this paper,we propose a novel approach to house price prediction from a multi-source data fusion *** methodology involves analyzing house characteristics and incorporating factors from diverse aspects,including amenities,traffic,and *** validate our approach using a dataset of 28550 real-world transactions in Beijing,China,providing a comprehensive analysis of the drivers influencing house *** adopting a multi-source data fusion perspective and considering a wide range of influential factors,our approach offers valuable insights into house price *** findings from this study possess the capability to improve the accuracy and effectiveness of house price prediction models,benefiting stakeholders in the real estate market.
