Endoplasmic reticulum stress and autophagy in cerebral ischemia/reperfusion injury:PERK as a potential target for intervention

Several studies have shown that activation of unfolded protein response and endoplasmic reticulum(ER)stress plays a crucial role in severe cerebral ischemia/reperfusion *** occurs within hours after cerebral ischemia,but the relationship between ER stress and autophagy remains *** this study,we established experimental models using oxygen-glucose deprivation/reoxygenation in PC12 cells and primary neurons to simulate cerebral ischemia/reperfusion *** found that prolongation of oxygen-glucose deprivation activated the ER stress pathway protein kinase-like endoplasmic reticulum kinase(PERK)/eukaryotic translation initiation factor 2 subunit alpha(e IF2α)-activating transcription factor 4(ATF4)-C/EBP homologous protein(CHOP),increased neuronal apoptosis,and induced ***,inhibition of ER stress using inhibitors or by si RNA knockdown of the PERK gene significantly attenuated excessive autophagy and neuronal apoptosis,indicating an interaction between autophagy and ER stress and suggesting PERK as an essential target for regulating *** autophagy with chloroquine exacerbated ER stress-induced apoptosis,indicating that normal levels of autophagy play a protective role in neuronal injury following cerebral ischemia/reperfusion *** from this study indicate that cerebral ischemia/reperfusion injury can trigger neuronal ER stress and promote autophagy,and suggest that PERK is a possible target for inhibiting excessive autophagy in cerebral ischemia/reperfusion injury.

Modified Bfat-1 gene and its biological verification in mice by hydrodynamic tail vein injection

Omega-3 polyunsaturated fatty acids （w-3 PUFAs） are essential components required for normal cellular function and have been shown to have important therapeutic and nutritional benefits in humans. But humans or mammals cannot naturally produce w-3 PUFAs, due to the lack of the co-3 fatty acid desaturase gene （fat-1 gene）. Previously, fat-1 gene has been cloned from Caenorhabditis elegans and transferred into mice, pigs and sheep, but not yet into beef cattle. We attempt to transfer it into beef cattle. The object of this paper is to edit the fat-1 gene from C. elegans to express more efficiently in beef cattle and verify its biological function in mice model. As a result, the fat-1 gene from C. elegans was modified by synonymous codon usage and named it Bfat-l. We have demonstrated that degree of codon bias of Brat-1 gene was in- creased in beef cattle. Moreover, Bfat-1 gene could be transiently expressed in mouse liver and muscle, the w-6/co-3 PUFAs ratio of 18 and 20 carbon was decreased significantly in liver （P〈0.05）, and the ratio of 20 carbon decreased significantly in muscle 24 and 72 h after injection （P〈0.05）. This confirms that the Bfat-1 gene modification was successful, and the protein encoded was able to catalyze the conversion of w-6 PUFAs to e0-3 PUFAs.

Effect of rosiglitazone on rabbit model of myocardial ischemia-reperfusion injury

To explore mechanism and protective effect of rosiglitazone on myocardial ischemia reperfusion(I/R) ***:A total of 48 male Japanese white big-ear rabbits were randomly divided into control group(A),I/R group(B),low dose of rosiglitazone group(C),high dose of rosiglitazone group(D).Plasma concentration of and also reduced the concentration of plasma serum creatine kinase(CK),***-sensitivity C-reactive protein(hsCRP).ultrasuperoxide dismutase(SOD),malondialdehyde(MD.A).lactic acid glutathione skin peroxidase (C-SH-PX).nitric oxide(NO)and endothelin(ET) were measured 1 h later after I/***-four hours after I/R the hearts were harvested for pathological and ultrastructural *** of myocardial infarction were ***:Plasma concentration of CK,***,NO. MDA and ET were decreased in C,D group compared with group *** concentration of T-SOD and GSH-Px were increased significantly in C.D group compared with group *** with group *** and ullraslructural changes in C and D group were *** was significant difference in myocardial infarction area between group C.D and group B(P<0.05). Myocardial infarction area and arrhythmia rate were lower in group C,D compare with group B. Rosiglitazone may protect myocardium from I/R injury by enhancing T-SOD and GSH-Px concentration,inhibit inflammatory reaction,and improve endothelial function.

Self-assembly of supramolecular polymers in water from tetracationic and tetraanionic monomers in water through cooperative electrostatic attraction and aromatic stacking

The cooperative electrostatic attraction and π-π aromatic stacking interactions between tetrahedral tetrapyridinium TP and three tetraanionic tetraphenylethylene derivatives TPE-1～3 led to the formation of a new kind of supramolecular polymer networks in water, which have been confirmed by^1 H NMR,fluorescence, isothermal titration calorimetric(ITC) and dynamic light scattering(DLS) experiments. ITC studies show that the contributions of enthalpy and entropy were comparable, reflecting the importance of hydrophobicity in driving the intermolecular aromatic stacking. DLS experiments indicate that the linear supramolecular polymers formed by these tetratopic monomers further aggregated into networks of 10~2-nm size.

The Record-Breaking Extreme Drought in Yunnan Province, Southwest China during Spring-Early Summer of 2019 and Possible Causes

In spring and early summer of 2019, Yunnan Province experienced the most severe seasonal drought on record,with days of extreme drought area exceeding 10^5 km^2 far more than normal. Consistently, the precipitation in each month from February to June is over 30% less than normal, and about 50% less in the most severe drought period(April–June). The rainy season in Southwest China(SWC) started on the third pentad in June 2019, which is the second latest in history. The rainy season in Yunnan started on 24 June, which is the latest(29 days later than normal). On the contrary, the onset of the South China Sea summer monsoon(SCSSM) is abnormally early. The lag time between the start of the rainy season in SWC and the onset of the SCSSM in 2019 is 7 pentads, which is the largest since 1961, much longer than the climate mean(less than 1 pentad). The present study analyzes the possible reasons why the rainy season came extremely late and the drought disaster persisted and intensified after a much early SCSSM, at both seasonal and subseasonal timescales. The abnormally late onset of the rainy season and the second greatest potential evapotranspiration(PET) since 1981 are the direct reasons for the persistent drought. Statistical results show that the water vapor from southwest of Yunnan in April–June contributes more than that from the east at the seasonal scale. In April–June 2019, however, the southern branch trough(SBT) was abnormally weak, the large and strong anticyclonic wind anomaly prevailed over the Bay of the Bengal(BOB), and the meridional water vapor transport to Yunnan was weak. At the subseasonal scale, the weaker SBT lasted the longest, and the strong convection over the BOB came up late despite of an early onset of the SCSSM, which resulted in reduced low-level moisture convergence in Yunnan and development of drought prior to the SCSSM onset. Shift of the principal mode of Pan-Asian monsoon summer precipitation in terms of spatial pattern

This paper documents that the principal mode of Pan-Asian monsoon summer precipitation experienced a prominent interdecadal shift around 1992/1993 in terms of spatial pattern and major driving factors. During 1979-1992 （Period 1, P1）, Pan-Asian monsoon summer precipitation anomalies mainly display a meridional dipole pattern from north to south, whereas in the period 1993-2016 （Period 2, P2）, it shows a meridional tripole pattern instead. The summer precipitation in P1 is primarily associated with a combination of the developing phase （central-eastern Pacific type） and decaying phase （eastern Pacific type） of El Nino-Southem Oscillation （ENSO）; while in P2, it is mainly associated with the decaying phase of central-eastern-Pacific-type ENSO.

Unlocking the stable interface in aqueous zinc-ion battery with multifunctional xylose-based electrolyte additives

The growth of dendrites and the side reactions occurring at the Zn anode pose significant challenges to the commercialization of aqueous Zn-ion batteries（AZIBs）. These challenges arise from the inherent conflict between mass transfer and electrochemical kinetics. In this study, we propose the use of a multifunctional electrolyte additive based on the xylose（Xylo） molecule to address these issues by modulating the solvation structure and electrode/electrolyte interface, thereby stabilizing the Zn anode. The introduction of the additive alters the solvation structure, creating steric hindrance that impedes charge transfer and then reduces electrochemical kinetics. Furthermore, in-situ analyses demonstrate that the reconstructed electrode/electrolyte interface facilitates stable and rapid Zn2+ion migration and suppresses corrosion and hydrogen evolution reactions. As a result, symmetric cells incorporating the Xylo additive exhibit significantly enhanced reversibility during the Zn plating/stripping process, with an impressively long lifespan of up to 1986 h, compared to cells using pure ZnSO4electrolyte. When combined with a polyaniline cathode, the full cells demonstrate improved capacity and long-term cyclic stability. This work offers an effective direction for improving the stability of Zn anode via electrolyte design, as well as highperformance AZIBs.

Bi-extrapolated subgradient projection algorithm for solving multiple-sets split feasibility problem

This paper deals with a bi-extrapolated subgradient projection algorithm by intro- ducing two extrapolated factors in the iterative step to solve the multiple-sets split feasibility problem. The strategy is intend to improve the convergence. And its convergence is proved un- der some suitable conditions. Numerical results illustrate that the bi-extrapolated subgradient projection algorithm converges more quickly than the existing algorithms.
